A comparative study of smooth muscle cell characteristics and myofibroblasts in processus vaginalis of pediatric inguinal hernia, hydrocele and undescended testis.

BACKGROUND: Congenital inguinal hernia, hydrocele and undescended testis (UDT) are associated with patent processus vaginalis. The smooth muscles present in the processus vaginalis aid in the descent of the testis and undergo programmed cell death after testicular descent leading to obliteration. The persisting amount of smooth muscle in the processus vaginalis influences the clinical outcome as inguinal hernia, hydrocele or UDT. Therefore, a study was conducted to evaluate the processus vaginalis in these three conditions to observe the presence and phenotype of smooth muscle cells and the pr

Influence of Epididymal Anomalies and Development of Processus Vaginalis on Testicular Migration And Cryptorchidism. A Narrative Review.

The objective of this narrative review is to show the most relevant aspects of the structure of epidydimal anomalies (EAs) and processus vaginalis (PV) during testicular migration and in patients with undescended testis. Smooth and striated muscles develop around the processus vaginalis (PV) within the gubernaculum during fetal life. The testis migrates through the PV by the means of peristaltic activity generated by those muscles. After migration is completed, the smooth muscle undergoes programmed cell death, resulting in obliteration of PV and leaving only the dartos muscle. Initiation of p

Transgenic overexpression of LARGE induces α-dystroglycan hyperglycosylation in skeletal and cardiac muscle.

BACKGROUND: LARGE is one of seven putative or demonstrated glycosyltransferase enzymes defective in a common group of muscular dystrophies with reduced glycosylation of α-dystroglycan. Overexpression of LARGE induces hyperglycosylation of α-dystroglycan in both wild type and in cells from dystroglycanopathy patients, irrespective of their primary gene defect, restoring functional glycosylation. Viral delivery of LARGE to skeletal muscle in animal models of dystroglycanopathy has identical effects in vivo, suggesting that the restoration of functional glycosylation could have therapeutic applic

The Morphology of the Tongue in the Spinner Dolphin (Stenella longirostris).

Assessments of the tongue morphology contribute to the understanding of evolutionary processes associated with food and feeding habits. In this sense, this study aimed to describe the tongue morphology in the Spinner dolphin (Stenella longirostris) and understand possible morphological associations with its food and feeding habits. Macroscopic dissections and light as well as scanning electron microscopy analyses of three Spinner dolphin tongues were performed. The tongues are elongated and exhibit a dorsal surface divided into three regions, namely the apex, the body and the root. Two types o

Common and Key Differential Pathogenic Pathways in Desminopathy and Titinopathy.

Myofibrillar myopathy (MFM) is a group of hereditary myopathies that mainly involves striated muscles. This study aimed to use tandem mass tag (TMT)-based proteomics to investigate the underlying pathomechanisms of two of the most common MFM subtypes, desminopathy and titinopathy. Muscles from 7 patients with desminopathy, 5 with titinopathy and 5 control individuals were included. Samples were labelled with TMT and then underwent high-resolution liquid chromatography-mass spectrometry analysis. Compared with control samples, there were 436 differentially abundant proteins (DAPs) in the desmin

AAV CRISPR editing rescues cardiac and muscle function for 18 months in dystrophic mice.

Adeno-associated virus-mediated (AAV-mediated) CRISPR editing is a revolutionary approach for treating inherited diseases. Sustained, often life-long mutation correction is required for treating these diseases. Unfortunately, this has never been demonstrated with AAV CRISPR therapy. We addressed this question in the mdx model of Duchenne muscular dystrophy (DMD). DMD is caused by dystrophin gene mutation. Dystrophin deficiency leads to ambulation loss and cardiomyopathy. We treated 6-week-old mice intravenously and evaluated disease rescue at 18 months. Surprisingly, nominal dystrophin was res

Complete primary structure of the I-band region of connectin at which mechanical property is modulated in zebrafish heart and skeletal muscle.

Connectin, also called titin, is the largest protein with a critical function as a molecular spring during contraction and relaxation of striated muscle; its mutation leads to severe myopathy and cardiomyopathy. To uncover the cause of this pathogenesis, zebrafish have recently been used as disease models because they are easier to genetically modify than mice. Although the gene structures and putative primary structures of zebrafish connectin have been determined, the actual primary structures of zebrafish connectin in heart and skeletal muscles remain unclear because of its large size and th

Morphological analysis of the functional design of the connection between the alimentary tract and the gas bladder in air-breathing lepisosteid fish.

There is considerable controversy in the literature regarding the existence of a "longitudinal slit" versus a "duct" connecting the respiratory gas bladder with the alimentary tract in various species of garfishes (Lepisosteidae). The aim of the present work has been to address these discrepancies on the basis of our own work on longnose gar Lepisosteus osseus in conjunction with a review of the literature. We provide anatomical evidence for a better understanding of the functional advantages of the "longitudinal slit" versus the "duct" in respect to feeding and respiration in juveniles longno

Localization of annexin VI in the adult and neonatal heart.

Annexins are a major family of intracellular Ca(2+)-binding proteins which have been implicated in a variety of cellular functions. In this paper the authors have used confocal microscopy to compare the distribution of annexin VI in vibratome sections of the rat adult left ventricle and striated muscle of the rat oesophagus. It is shown that in rat cardiac myocytes annexin VI is associated with only the sarcolemma and intercalated discs. In contrast, it is demonstrated that in rat skeletal muscle annexin VI is associated with the sarcoplasmic reticulum, in addition to the plasma membrane, sugg

[Surgical anatomy of the bladder neck sphincter and its significance for vaginal surgery of stress incontinence].

The bladder closure mechanism works under the influence of a hydro-aerodynamic force that presses downward ("stress"). This "stress" is caused by the relative weakness of the pelvic floor. The structures running through the urogenital hiatus are compressed by the rectococcygeal and pubo-coccygeal muscles, which close the hiatus. The urogenital diaphragm bridges the slit in the levator ani muscles. It is made of the perineal membrane, the superior fascia, and the smooth and striated muscle lying between the two (M. sphincter urethrovaginalis, M. compressor urethrae). The superior fascia is an e

Composition and central projections of the pudendal nerve in the rat investigated by combined peptide immunocytochemistry and retrograde fluorescent labelling.

The central distribution of the afferent and efferent projections of pelvic striated muscles, the pudendal and sciatic nerves, were systemically investigated in rats by retrograde tracing techniques combined with immunocytochemistry using antibodies to 9 neuropeptides. True Blue was injected into either the pelvic muscles, pudendal or sciatic nerves. Seven days later the spinal cord and dorsal root ganglia (L3-S2 levels) were processed for immunocytochemistry. Injection of tracer into the pelvic muscles labelled dorsomedial, ventral and dorsolateral motoneuron groups of the L6 segment and a fe

Strabismus and eye muscle function.

Studies of external eye muscle morphology and physiology are reviewed, with respect to both motor and sensory functions in concomitant strabismus. The eye muscles have a more complex fibre composition than other striated muscle, and they are among the fastest and most fatigue-resistant muscles in the body. However, it is not generally believed that concomitant strabismus is due to a primary abnormality of the eye muscles or the ocular motor system. The gross anatomy of eye muscles, including the shape and position of the eye muscle pulleys, was not changed in strabismus. The histology of the e

Autologous bone-marrow-derived mesenchymal stem cell transplantation into injured rat urethral sphincter.

OBJECTIVES: To evaluate the functional and histological recovery by autologous bone-marrow-derived mesenchymal stem cell (BMSC) transplantation into injured rat urethral sphincters. METHODS: BMSC were harvested from female Sprague-Dawley retired breeder rats for later transplantation. The cells were cultured, and transfected with the green fluorescence protein gene. The urethral sphincters were injured by combined urethrolysis and cardiotoxin injection. One week after injury, the cultured BMSC were injected autologously into the periurethral tissues. Controls included sham-operated rats and i

The relationship between the metabolic syndrome and energy-utilization deficit in the pathogenesis of obesity-induced osteoarthritis.

We propose that the pathogenesis of obesity-induced osteoarthritis may be explained by the metabolic changes in the striated muscle induced by the interaction of insulin resistance and systemic inflammation in obese individuals with metabolic syndrome being osteoarthritis the latest consequence by the physiological changes seen in the metabolic syndrome. Increased levels of TH1 cytokines are produced by activated macrophages in the presence of an acute or chronic infectious disease and suppress the sensitivity of insulin receptors on the membrane of muscle cell and adipocytes. Both cells are a

Free skeletal muscle transplantation to an infarction area: an experimental study in the dog.

In plastic surgery, free skeletal muscle flaps are used for treatment of non-healing injuries like chronic osteitis or radiation ulcers. We wanted to evaluate if the neoangiogenesis induced by these flaps can also constitute a method for revascularization in ischemic heart disease. In diffuse small vessel coronary heart disease, patients are not suitable for bypass surgery because it is impossible to create a microanastomosis with the coronary artery. As an alternative, a free striated muscle flap, transplanted onto the heart, can constitute a source of neovessels. With the induction of extra-

Randomized controlled trial of abductor muscle damage in relation to the surgical approach for primary total hip replacement: minimally invasive anterolateral versus modified direct lateral approach.

INTRODUCTION: Minimally invasive total hip arthroplasty (THA) is claimed to be superior to the standard technique, due to the potential reduction of soft tissue damage via a smaller and tissue-sparing approach. As a result of the lack of objective evidence of fewer muscle and tendon defects, controversy still remains as to whether minimally invasive total hip arthroplasty truly minimizes muscle and tendon damage. Therefore, the objective was to compare the influence of the surgical approach on abductor muscle trauma and to analyze the relevance to postoperative pain and functional recovery. M

Myostatin regulates pituitary development and hepatic IGF1.

Circulating myostatin-attenuating agents are being developed to treat muscle-wasting disease despite their potential to produce serious off-target effects, as myostatin/activin receptors are widely distributed among many nonmuscle tissues. Our studies suggest that the myokine not only inhibits striated muscle growth but also regulates pituitary development and growth hormone (GH) action in the liver. Using a novel myostatin-null label-retaining model (Jekyll mice), we determined that the heterogeneous pool of pituitary stem, transit-amplifying, and progenitor cells in Jekyll mice depletes more

Neural deficits contribute to respiratory insufficiency in Pompe disease.

Pompe disease is a severe form of muscular dystrophy due to glycogen accumulation in all tissues, especially striated muscle. Disease severity is directly related to the deficiency of acid alpha-glucosidase (GAA), which degrades glycogen in the lysosome. Respiratory dysfunction is a hallmark of the disease, muscle weakness has been viewed as the underlying cause, and the possibility of an associated neural contribution has not been evaluated previously. Therefore, we examined behavioral and neurophysiological aspects of breathing in 2 animal models of Pompe disease--the Gaa(-/-) mouse and a tr

Histochemical study of urethral striated musculature in the dog.

The striated musculature of the dog urethra was studied histochemically. Two main groups of muscle fibers could be identified: 1 with slow twitch oxidative fibers, which are fatigue resistant (type 1), and 1 with fast twitch fibers (type 2). The fast twitch fibers were subdivided into glycolytic (fatiguable) and oxidative glycolytic (fatigue resistant) fibers: the latter constitute 20 per cent of all fast twitch fibers. Type 1 constitutes of 35 per cent of the whole musculature and its proportion tends to decrease toward the distal end of the external urethral sphincter. From these observation

Regimen-dependent glucocorticoid effects improve muscle performance without altering CNS physiology in mdx mice.

Duchenne muscular dystrophy (DMD) is a multisystem disorder affecting striated muscle, metabolism and the central nervous system (CNS). Although glucocorticoids remain the standard therapy, muscle-centric evaluations typically fail to capture how dosing regimen and compound selection affect CNS and metabolic phenotypes. Here, we compared daily and weekly dosing of prednisolone and vamorolone in juvenile mdx mice over 6 weeks to determine how these variables influence multisystem outcomes. Multiorgan efficacy and adverse effects were quantified across behavioural, endocrine, metabolic, cardiova
