Prevalence and risk factors of pulmonary nontuberculous mycobacterial infections in the Zhejiang Province of China.

Risk factors and prevalence of pulmonary nontuberculous mycobacterial (NTM) diseases were retrospectively evaluated in 1208 suspected pulmonary TB patients seeking care at the Affiliated Hospital of Hangzhou Normal University between July 2018 and December 2018. Further analysis of 390 culture-positive cases demonstrated that 358 (358/390, 91.8%) were infected with Mycobacterium tuberculosis (MTB), 24 (24/390, 6.2%) with NTM and eight (8/390, 2.0%) with both MTB and NTM. M. intracellulare was the most prevalent NTM isolated (16/24, 66.7%), followed by M. abscessus (3/24), M. kansasii (2/24), M

Synthesis, Biological Evaluation, Molecular Docking Studies and ADMET Prediction of Oxindole-based Hybrids for the Treatment of Tuberculosis.

INTRODUCTION: With a projected mortality toll of 1.4 million in 2019, tuberculosis (TB) continues to be a significant public health concern around the world. Studies of novel treatments are required due to decreased bioavailability, increased toxicity, increased side effects, and resistance of several first- and second-line TB therapies, including isoniazid and ethionamide. METHODS: This study reports the synthesis of oxindole-based hybrids as potent InhA inhibitors targeting Mycobacterium tuberculosis. The synthesized compounds (5a-5e and 8a-8c) were evaluated for their anti-mycobacterial ac

Discovery of Biofilm-Inhibiting Compounds to Enhance Antibiotic Effectiveness Against M. abscessus Infections.

Background/Objectives: Mycobacterium abscessus (MAB) is a highly resilient pathogen that causes difficult-to-treat pulmonary infections, particularly in individuals with cystic fibrosis (CF) and other underlying conditions. Its ability to form robust biofilms within the CF lung environment is a major factor contributing to its resistance to antibiotics and evasion of the host immune response, making conventional treatments largely ineffective. These biofilms, encased in an extracellular matrix, enhance drug tolerance and facilitate metabolic adaptations in hypoxic conditions, driving the bacte

The use of dual β-lactams to restore susceptibility of Mycobacterium avium complex.

BACKGROUND: The Mycobacterium avium complex (MAC) are non-tuberculous mycobacteria responsible for chronic and debilitating conditions. Guideline-recommended therapy for MAC is a combination of clarithromycin/azithromycin, ethambutol and a rifamycin. However, culture conversion rates with this regimen are 67%. Alternative treatment options are needed. Recent findings of β-lactam combinations in the treatment of other mycobacterial diseases have been promising. The proposed mechanism is an additive inhibition of multiple enzymes in the peptidoglycan synthesis pathway by the β-lactam combination

Managing Complicated Nontuberculous Mycobacteria Infections in Plastic Surgery.

Nontuberculous mycobacteria (NTM) infections after cosmetic surgery have become an increasing concern. These infections are often initially misdiagnosed and treated with standard antibiotic regimens, which fail to resolve the underlying infection, leading to prolonged patient suffering. In this case study, we describe a chronic wound infection caused by Mycobacterium abscessus subsp. bolletii after a muscle-repair abdominoplasty. This case illustrates the diagnostic and therapeutic challenges plastic surgeons face in successfully treating such infections. Initial obstacles included the isolati

Incidence of mycobacteria in pulmonary granulomatous lesions.

Mycobacteria infections are caused by species of the Mycobacterium tuberculosis complex (MTB) and other species called Non-Tuberculosis Mycobacteria (NTM). Identification of mycobacteria species is very important to define treatment and it can be achieved by direct culture. However, the lack of clear protocols regarding the use of culture or molecular tests on specimens diagnosed with granulomatous lesions causes delays in the diagnosis of the etiological agents and, consequently, the definition of the right treatment. This work aimed to characterize the incidence of mycobacteria species in pu

Cutaneous mycobacteriosis in a captive Amazonian manatee Trichechus inunguis.

An adult male Amazonian manatee Trichechus inunguis under human care presented with 3 circular cutaneous lesions on the dorsal aspect of the rostrum and between the nostrils (plenum). Initially these lesions were superficial, hypopigmented, without warmth and non-painful. Microbiological cultures of skin swabs isolated Candida sp. and Pseudomonas aeruginosa, and topical treatment with antiseptic, antifungal, anti-inflammatory and antibiotic medication was instituted. This treatment strategy did not lead to any clinical improvement, and after 6 mo, the lesions progressed to a confluent abscess

Enzyme-DNA Interactions Affect the Catalytic Inhibition of Mycobacterial Gyrases by Antibacterial Drugs.

Gyrase and topoisomerase IV, enzymes that play critical roles during DNA replication, are the targets of fluoroquinolones and other antibacterials. Gyrase removes positive supercoils that accumulate ahead of replication forks, while topoisomerase IV untangles daughter chromosomes. Although topoisomerase IV is an essential enzyme in most bacteria, some species, including Mycobacterium tuberculosis and Mycobacteroides abscessus, encode only gyrase. In these species, gyrase is the sole target of fluoroquinolones and is believed to assume the cellular functions of both type II topoisomerases. Alth

Persistent Pathogens Following Minimally Invasive Surgery: Is It Time to Rethink Aldehyde-Based Disinfection?

Atypical mycobacterial (ATM) infections have emerged as a persistent problem in minimally invasive surgery (MIS), presenting as delayed surgical site infections (SSIs) that are difficult to diagnose and treat. We retrospectively reviewed 47 patients referred to our center over an 11-year period with delayed port-site complications following laparoscopic procedures performed elsewhere. Our focus was on evaluating the link between ATM infections and sterilization practices in hospitals, while secondarily considering outcomes and implications for the prevention of the same. The mean age was 43.6

Epidemiology of nontuberculous mycobacteria infection in Asia: A narrative review.

Recent population-based studies conducted in Asia have revealed a notable increase in the overall incidence of nontuberculous mycobacteria (NTM) infection, coinciding with a decline in tuberculosis (TB) cases. The incidence of NTM infection has exceeded that of TB in Japan, with a prevalence rate showing a substantial increase over the years. Similarly, South Korea and Taiwan have witnessed an increase of NTM infection rates, particularly in pulmonary disease. The NTM species isolation rate has increased in the past years globally. The most common NTM species isolated in Asia was Mycobacterium

[Molecular identification of non-tuberculous mycobacteria].

INTRODUCTION: Nontuberculous mycobacteria can be saprophytic, pathogenic or opportunistic. The most common diseases produced by these microorganisms are the post-surgical infections due to anesthetic procedures, infections associated with catheters, disseminated cutaneous diseases and pulmonary and central nervous system diseases that especially affect HIV patients. Identification of the nontuberculous mycobacteria can take several weeks and even then, differentiation of complex members is not possible. OBJECTIVE: The PCR-restriction analysis (PRA) technique was evaluated as a method for geno

Benchmarking deep learning architectures for MALDI-TOF mass spectrometry in infectious disease diagnostics and vector-borne disease surveillance.

Emerging global health threats, from antimicrobial resistance to vector-borne diseases, require scalable diagnostic solutions. Matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry has revolutionized microbial diagnostics but remains underutilized in public health due to algorithmic challenges in interpreting and generalizing complex spectra. Here, we present a systematic benchmarking framework to expand MALDI-TOF's utility for field-deployable diagnostics using diverse deep learning architectures and preprocessing strategies, including approaches newly appli

Synthetically accessible naphthalene-based rigid-core electrolytes as membrane-active antibacterial agents against drug-resistant pathogens.

The growing threat of antimicrobial resistance (AMR) underscores the need for antibacterial scaffolds that act through mechanisms distinct from those of conventional antibiotics. Conjugated oligoelectrolytes (COEs) are membrane-active antimicrobials, although stilbene-containing representatives can present scaffold-specific challenges associated with alkene stability and synthetic accessibility. Here, we report a naphthalene-based rigid-core electrolyte (RCE) platform that removes the phenylene-vinylene linker while retaining potent antibacterial activity. The RCEs were accessed through a two-
