Single-round infectious particles enhance immunogenicity of a DNA vaccine against West Nile virus.

DNA vaccines encoding replication-defective viruses are safer than inactivated or live attenuated viruses but may fail to stimulate an immune response sufficient for effective vaccination. We augment the protective capacity of a capsid-deleted flavivirus DNA vaccine by co-expressing the capsid protein from a separate promoter. In transfected cells, the capsid-deleted RNA transcript is replicated and translated to produce secreted virus-like particles lacking the nucleocapsid. This RNA is also packaged with the help of co-expressed capsid protein to form secreted single-round infectious particl…

Primary biliary cirrhosis with multinucleated hepatocellular giant cells: implications for pathogenesis of primary biliary cirrhosis.

Histological and clinical features of syncytial giant cell hepatitis (GCH) are rarely observed in adults, and the disease has been associate … microscopy demonstrated the presence of filamentous strands and particles resembling paramyxovirus nucleocapsids in the cytoplasm of syncytial giant cells. To our knowledge, this is the first case of PBC with histological and clinical evidence of syncytial GCH in an adult, and we submit that it might provide novel clues in the enigma of PBC pathogenesis.

Sequence heterogeneity in the termini of lymphocytic choriomeningitis virus genomic and antigenomic RNAs.

Sequence analysis of lymphocytic choriomeningitis virus L and S RNAs has revealed evidence of heterogeneity within the termini of the genomi … ni may be recognized as functional templates for replication. RNAs extracted from virions, or viral nucleocapsids isolated from acutely infected cells, are similar in the nature and extent of terminal heterogeneity that have been observed. This finding brings into question the function of the conserved sequences located at the termini of arenavirus genomic RNA …

HLA control of the proliferative T lymphocyte response to antigenic determinants on mumps virus. Studies of healthy individuals and patients with type 1 diabetes.

The proliferative T lymphocyte responses to two different mumps antigen preparations, S (nucleocapsid) and V (viral envelope), were characterized. Eight patients with Type 1 (insulin-dependent) diabetes mellitus were all found to be responders to S and V mumps antigen. Among the 64 healthy individuals, 52 were classified as responders to the S antigen and 50 responders to the V antigen. No difference was found between T lymphocytes from Type 1 diabetic patients and those from healthy individuals as regards the effect of indomethacin on the mumps-specific response. The majority of the mumps-spe…

Development of PRRSV-1 specific monoclonal antibody and detection of PRRSV-1 infection.

Porcine reproductive and respiratory syndrome virus (PRRSV) are responsible for reproductive failure in sows and respiratory diseases in pigs, inflicting significant economic losses worldwide. Due to continuous mutation and recombination, PRRSV exhibit remarkable genetic diversity, necessitating detection methods capable of recognizing PRRSV-2 and/or PRRSV-1 strains. In this study, four monoclonal antibodies (mAbs) targeting the PRRSV-1 nucleocapsid (N) protein were generated, and the precise and novel linear B-cell epitopes were identified as 54PHFPL58, 53KPHFP57, and 59AAEDDIRHH67, respectiv…

SARS-CoV-2 N Protein Hijacks the m6A Reader YTHDF2 to Suppress Antiviral Gene Expression.

The m6A RNA methylation pathway plays a critical role in host antiviral defense. Host cells employ m6A readers such as YTHDF2 to regulate viral RNA fate through diverse mechanisms, including degradation, translational control, and immune recognition. However, we found that YTHDF2 is essential for SARS-CoV-2 replication, suggesting that a virus may exploit this host machinery to its advantage. Through integrative RNA-proteome analysis, we identified the SARS-CoV-2 nucleocapsid (N) transcript as the most heavily m6A-modified viral transcript and a direct interactor of YTHDF2. The N protein forms…

A Nanobody-Based Lateral Flow Assay for Point-of-Care Diagnostics.

Lateral flow assays (LFAs) are among the most successful technologies for point-of-care and at-home testing, but further advances are needed to reduce costs and accelerate development. Alpaca-derived nanobodies (Nbs), single-domain antibody fragments, are promising immunoassay reagents across diverse applications. Their small size and ease of recombinant production make them particularly well suited for diagnostics. Here, we present a paper-based LFA targeting the SARS-CoV-2 nucleocapsid (N) protein that exclusively uses Nbs for direct antigen detection. We also demonstrate in-house synthesis …

Engineering a Modular PapMV Nanoparticle Vaccine: Comparative Efficacy of a Covalent and a Non-Covalent N-Antigen Vaccine Against Emerging SARS-CoV-2 Variants.

Background: Despite the effectiveness of current SARS-CoV-2 vaccines, the genetic variability of the viral target has led to the emergence of variants capable of evading vaccine-induced protection. To ensure broader and more durable protection, we investigated the efficacy of a novel vaccine strategy. Methods: This vaccine utilizes the highly conserved nucleocapsid (N) protein as its primary antigen, rather than the spike (S) protein. It incorporates the Papaya Mosaic Virus (PapMV) nanoparticle, a Toll-like receptor (TLR) 7/8 agonist with intrinsic adjuvant properties, as a vaccine platform. R…

SARS-CoV-2 Infection Induces Dopaminergic Neuronal Loss in Midbrain Organoids.

COVID-19 is mainly associated with respiratory symptoms, although several reports show that SARS-CoV-2 affects the nervous system. We evaluated the effects of SARS-CoV-2 infection on human derived midbrain organoids by exposing them to the virus (multiplicity of infection 0.05, 16-h exposure) and analyzing cellular and molecular changes at 4 and 28 days post-infection using immunofluorescence microscopy and RNA sequencing. SARS-CoV-2 nucleocapsid protein preferentially colocalized with tyrosine hydroxylase-positive (TH+) dopaminergic neurons, inducing neurite fragmentation and cellular stress.…

Immune correlates of risk for SARS-CoV-2 infection in children: a prospective, community-based cohort study.

Few studies have characterized immune correlates of SARS-CoV-2 infection risk in children, particularly those with hybrid immunity from vaccination and prior infection. We conduct a prospective community-based cohort study of 1509 U.S. children (2022-2024), performing weekly SARS-CoV-2 PCR testing and measuring baseline binding and neutralizing antibody titers against multiple variants. Higher antibody levels, notably nucleocapsid-binding and Omicron-specific neutralizing antibodies, are significantly associated with reduced risk of SARS-CoV-2 infection after adjusting for age, recent infectio…

Engineered VP2-NC antigen expressed in an NDV vector elicits enhanced protection against the challenge of the novel variant infectious bursal disease virus.

To enhance the efficacy of Newcastle disease virus (NDV)-vectored bivalent vaccines against novel variant infectious bursal disease virus (nVarIBDV), we engineered recombinant NDV strains expressing nVarIBDV VP2 proteins fused with the viral nucleocapsid NC motif, thereby leveraging the NDV assembly pathway. The resulting vaccine candidates, rDM-VP2NC and rDM-VP2mNC, exhibited markedly enhanced VP2 expression in vitro. Furthermore, the NC motif promoted VP2 co‑localization with the NDV M protein near the cell membrane and facilitated its efficient incorporation into budding virus-like particle…

Host transcriptional corepressor TLE1 is commonly exploited by RNA viruses for replication.

Viral genomes encode transcriptional regulators that alter the expression of host genes to facilitate virus replication, and the comprehensi … ved across various RNA viruses, as evidenced by the presence of a bHLH-like motif in the SARS-CoV-2 nucleocapsid protein. Loss-of-function substitutions in the bHLH-like motif attenuated the replication of both IAV and SARS-CoV-2. Our study reveals how RNA viruses exploit the components of the host transcription machinery to facilitate virus replication.

Hyperglycemia in Cats Infected by SARS-CoV-2: Pancreatic Alterations and Potential Antiviral Therapeutics.

Cats represent a susceptible host and a possible translational model for investigating coronavirus pathogenesis and therapeutics. Recent immunohistochemical (IHC) and histopathological studies in both human and feline tissues have demonstrated SARS-CoV-2 nucleocapsid protein (NP) and spike protein expression within pancreatic islet cells, following a classic temporal infection course. Notably, IHC analysis also reveals NP expression within exocrine ductal epithelial cells. Given that ductal epithelium functions as an islet progenitor pool during tissue injury or metabolic stress, pancreotropic…

C5a-C5aR1 axis activation drives neutrophil extracellular trap formation in SARS-CoV-2 N protein-mediated acute kidney injury.

The mechanism of COVID-19-associated acute kidney injury (AKI) remains incompletely understood. This study aimed to investigate the regulatory role of the C5a-C5aR1 axis in neutrophil extracellular trap (NET) formation in SARS-CoV-2 nucleocapsid (N) protein-induced AKI. An AKI mouse model with kidney-specific N protein expression was established using ultrasound-microbubble-mediated technology, and interventions with the C5aR1 inhibitor DF2593A or DNase I (Pulmozyme) were administered to detect renal neutrophil infiltration, NET deposition, C5a-C5aR1 expression, and renal function indices. The…

Rational design, synthesis, and evaluations of HBV capsid assembly modulators featuring a core of 2-amino-5-(2-amino-2-oxoacetyl)-4-methylthiophene-3-carboxamide.

Hepatitis B virus (HBV) remains a major global health challenge, where current therapies offer limited functional cure rates and are often burdened by adverse effects. HBV capsid assembly modulators (CAMs) represent a promising therapeutic approach by targeting viral nucleocapsid formation. Compounds GLP-26 and b were reported as CAMs exhibiting potent antiviral activity. A series of HBV CAMs featuring the core of 2-amino-5-(2-amino-2-oxoacetyl)-4-methylthiophene-3-carboxamide were designed and synthesized by integrating the acetamide fragment of GLP-26 with the pentavalent aromatic amine of b…

Cross-neutralization of SARS-CoV-2 BA.3.2.2 lineage by JN.1 mRNA vaccine-induced immunity.

OBJECTIVES: The SARS-CoV-2 BA.3.2.2 sublineage has emerged globally as the dominant branch of BA.3.2 by late 2025, yet its antigenic relationship with JN.1 vaccine-induced immunity remains unclear. METHODS: We evaluated neutralizing antibody responses in 25 JN.1 mRNA vaccinees against eight variants, stratified by anti-nucleocapsid antibody serostatus. RESULTS: Postvaccination titers increased significantly against all variants in both N antibody-negative and -positive groups. Cross-neutralization against BA.3.2.2 was detected in both groups despite lower titers compared to JN.1. Antigenic c…
