Expression of collagen type I, II and III in loose body of osteoarthritis.

The expression of collagen type I, II, and III was investigated to evaluate phenotypic change in chondrocytes in loose bodies related to ost … of fibroid tissue on the surface. No positive signal for collagen type III was observed in necrotic osteocytes. There was weakly positive staining for collagen type I in chondrocytes. No positive staining for collagen type II could be seen in the cartilage of loose bodies. Cartilage from the non-osteoarthritic knee joints of four people was negative for the …

Teriparatide therapy for bisphosphonate-related osteonecrosis of the jaw associated with dental implants.

This report describes a case of teriparatide (TPTD) therapy for bisphosphonate (BP)-related osteonecrosis of the jaw induced after implant placement. A 75-year-old woman taking oral BP was referred with uncontrolled osteonecrosis of the mandible related to the implant placement. With conservative treatment, BP was suspended and daily subcutaneous injections of 20 μm/d TPTD were started. After 4 months of the therapy, fixture removal and sequestrectomy were performed. Histological analysis revealed necrotic lamellar bone and empty osteocytic lacunae. In contrast, multiple irregular reversal lin

Jaw osteonecrosis in patients treated with bisphosphonates: an ultrastructural study.

Osteonecrosis of the jaw is a severe bone disorder traditionally associated with periodontal disease, local malignancy, chemotherapy, glucocorticoid therapy, or trauma. Recently a growing number of publications reported the occurrence of osteonecrosis of the jaw in patients undergoing treatment with bisphosphonates. The mechanism by which bisphosphonates might contribute to the development of osteonecrosis of the jaw is far from being fully elucidated. Suppression of bone turnover, infection, tissue hypoxia and cellular toxicity were proposed as possible mechanisms by which bisphosphonates may

Histologic analysis of bony Bankart lesions in recurrent anterior instability of the shoulder.

The histologic examination of bony Bankart lesions was performed with hematoxylin-eosin staining of specimens obtained intraoperatively from 27 patients. We assessed the incidence and extent of degeneration in the bony fragment and the surrounding ligament and the relationship of such pathologic changes to several clinical and radiologic factors. Loss of osteocytes in the bony fragment was defined as osteonecrosis, and loss of fibroblasts in the surrounding ligament was defined as ligament degeneration. Although extensive ligament degeneration was noted in 8 patients (29.6%) (degeneration grou…

Modern analysis of bone loss mechanisms in microgravity.

A summary of results of investigations by the author and a brief review of some literature data on human bone tissue deprived of mechanical loading (spaceflight, hypokinesia) is given. The direction and markedness of changes in bone mass--the bone mineral density and the bone mineral content--in different skeletal segments depend on their position relative to the gravity vector. A theoretically expected bone mass reduction was revealed in the trabecular structures of the bones of the lower part of the skeleton (local osteopenia). In the upper part of the skeleton, an increase in the bone miner

[The histo- and ultrastructural characteristics of the bone tissue in osteogenesis imperfecta].

Histo- and ultrastructure of bone tissue in 28 persons (2 section observations and 26 cases of bone biopsy analysis) with congenital fragility of the bones and dysosteogenesis were studied. Age of the victims ranged from 6 months to 38 years (20 men and 8 women). Against the background of bone rarefaction and decrease in quality of basic bone substance the quantity of osteocytes for a unit of osseous beam area increases. Electron-microscopic investigation revealed changes in osteoblast structure (3 types detected) by the degree of development of granular endoplasmic reticulum. In the 2nd and 3…

Calcium deficiency, pregnancy, and lactation in rats. Microscopic and microradiographic observations on bones.

Adult female rats were subjected to a calcium-depriving regimen (calcium-deficient diet containing oxalate + pregnancy + lactation) to obtain maximum bone mineral mobilization in as short a time as possible. The femur and tibia were investigated by histological, microradiographic and fluorescent microscopic methods. The regimen caused osteoporosis, which varied in severity with the degree of calcium deprivation. Most of the bone tissue removed was taken from the metaphyseal trabeculae and from the endosteal surface of the diaphysis. The remaining bone tissue seemed unchanged. The cells respons

[Morphometric analysis of periosteocytic osteolysis: its application to the diagnosis of hyperparathyroidism].

The authors indicate the required methodologic conditions for the morphometric analysis of periosteocytic osteolysis, a reliable and specific feature of overactivity of the parathyroid gland. Indeed, significant periosteocytic enlargement has been found in 101 113 cases of primary hyperparathyroidism compared to 92 reference cases. The measurement of the size of the periosteocytic lacunae may be carried out either on hand of a micrometric eyepiece or of a picture analyser. It has to be carried out on decalcified bone sections of 5 microns thickness and it is necessary to measure at least 50 la

Adynamic Bone Disease in Chronic Kidney Disease: From PTH Suppression-Driven Remodeling Phenotype to Osteoanabolic Therapy.

BACKGROUND: Adynamic bone disease (ABD) is an increasingly prevalent form of low-turnover renal osteodystrophy in advanced chronic kidney disease (CKD). ABD was not recognized as a clinical entity prior to the widespread adoption of PTH-targeted therapeutic strategies, and sustained iatrogenic suppression of parathyroid hormone (PTH) activity is now understood as the primary and defining causal mechanism underlying the adynamic phenotype. Uremic toxins, FGF-23-Klotho dysregulation, impaired vitamin D signaling, chronic inflammation, calcium-phosphate imbalance, and osteocyte-mediated abnormali

Single-Cell Transcriptomics of Bone Marrow Stromal Cells in Diversity Outbred Mice: A Model for Population-Level scRNA-Seq Studies.

Genome-wide association studies (GWASs) have advanced our understanding of the genetics of osteoporosis; however, the challenge has been converting associations to causal genes. Studies have utilized transcriptomics data to link disease-associated variants to genes, but few population transcriptomics data sets have been generated on bone at the single-cell level. To address this challenge, we profiled the transcriptomes of bone marrow-derived stromal cells (BMSCs) cultured under osteogenic conditions from five diversity outbred (DO) mice using single-cell RNA-seq (scRNA-seq). The goal of the s

Application of biomaterials in treating early osteonecrosis of the femoral head: Research progress and future perspectives.

Osteonecrosis of the femoral head (ONFH), a progressive pathological process of femoral head ischemia and osteocyte necrosis, is a refractory orthopedic disease caused by multiple etiologies and there is no complete cure at present. With the extension of ONFH duration, osteocyte apoptosis and trabecular bone loss can decrease the load-bearing capacity of the femoral head, which leads to the collapse of the articular cartilage and subchondral bone. Therefore, an urgent clinical need exists to develop effective treatment strategies of early-stage ONFH for maintaining the hip joint function and p

How osteons form: A quantitative hypothesis-testing analysis of cortical pore filling and wall asymmetry.

Osteon morphology provides valuable information about the interplay between different processes involved in bone remodelling. The correct qu … illing that includes osteoblast secretory activity, osteoblast elimination, osteoblast embedment as osteocytes, and osteoblast crowding and redistribution along the bone surface. The mathematical model is first calibrated to symmetric osteon data, and then used to test three mechanisms of asymmetric wall formation against osteon data: (i) delays in the onset …

The effect of Irisin on bone cells in vivo and in vitro.

The myokine Irisin, produced during physical exercise, has an anabolic effect on bone, both in vitro and in vivo. Very recently, using a controlled in vitro 3D cell model to mimic the bone microenvironment aboard the International Space Station, it has been shown that Irisin treatment in microgravity prevents the down-regulation of the transcription factors Atf4, Runx2 and Osterix, as well as Collagen I and Osteoprotegerin proteins, crucial for osteoblast differentiation in physiologic conditions. Irisin action has also been investigated in human subjects, in which it correlates with bone heal

Role of CCN2/CTGF/Hcs24 in bone growth.

Our bones mostly develop through a process called endochondral ossification. This process is initiated in the cartilage prototype of each bone and continues through embryonic and postnatal development until the end of skeletal growth. Therefore, the central regulator of endochondral ossification is the director of body construction, which is, in other words, the determinant of skeletal size and shape. We suggest that CCN2/CTGF/Hcs24 (CCN2) is a molecule that conducts all of the procedures of endochondral ossification. CCN2, a member of the CCN family of novel modulator proteins, displays multi

Reprogramming of Human Limbal Niche Progenitors.

Limbal niche cells (LNCs) possess embryonic stem cell (ESC) characteristics, expressing ESC markers such as Sox2, Nanog, and Oct4; stromal characteristics, expressing neurocrest markers such as CD90; and have angiogenic potential expressing angiogenesis markers such as CD34. In addition, these cells can be induced into adipocytes, chondrocytes, and osteocytes, indicating that they are valuable adult stem cells. However, how these cells may be appropriately passaged and maintained, what signaling pathways are necessary for such regeneration, and how the cells can be used for the treatment of va…
