The effects of human immunoglobulin G on enhancing tissue protection and neurobehavioral recovery after traumatic cervical spinal cord injury are mediated through the neurovascular unit.

BACKGROUND: Spinal cord injury (SCI) is a condition with few effective treatment options. The blood-spinal cord barrier consists of pericytes, astrocytes, and endothelial cells, which are collectively termed the neurovascular unit. These cells support spinal cord homeostasis by expressing tight junction proteins. Physical trauma to the spinal cord disrupts the barrier, which leads to neuroinflammation by facilitating immune cell migration to the damaged site in a process involving immune cell adhesion. Immunosuppressive strategies, including methylprednisolone (MPSS), have been investigated to…

Nanomedicine-Based Approaches for Therapeutic Delivery Across the Blood-Brain Barrier in Neurological Disorders.

Therapeutic delivery to the Central Nervous System (CNS) is challenging for formulation scientists due to the transfer of therapeutics across the Blood-Brain Barrier (BBB). The BBB consists of a selective semi-permeable anatomical structure of the capillary basement membrane, capillary endothelial cells, pericytes, and astrocytes. It restricts the drug transfer from the blood compartment to the brain tissues. Therefore, the desirable effects of various therapeutics in different CNS disorders (like Alzheimer's disease, schizophrenia, Parkinson's disease, etc.) require the transfer of drugs acro…

Single-Nucleus Profiling Reveals a BBB Senescence Unit Driving AD Pathology in Human Brain.

Cellular senescence contributes to Alzheimer's disease (AD) and involves the neurovascular unit, but the molecular mechanisms of cellular senescence in AD pathogenesis remain incompletely understood. Here, we integrate single-nucleus RNA sequencing data from 75 human brain samples to identify a coordinated BBB senescence unit of astrocytes, pericytes, microglia and T cells. Molecular profiling shows inflammatory signaling with maladaptive repair responses in senescent cells. Intercellular communication analysis identifies the SPP1-CD44 axis as central to BBB senescence in AD, with up-regulated…

Ionic regulation of cancer cell stiffness and metastatic colonization via the MRTFA-KCNMB1 axis.

Cellular stiffness impacts multiple steps of cancer metastasis, but mechanisms that regulate the stiffness of cancer cells remain poorly understood. Here, we identified potassium efflux and potassium calcium-activated channel subfamily M regulatory beta subunit 1 (KCNMB1), an auxiliary subunit of the large conductance calcium-activated potassium (BK) channels, as regulators of cellular stiffness downstream of myocardin-related transcription factor A (MRTFA). In primary pericytes, KCNMB1 knockdown increased cellular stiffness, which is consistent with the role of potassium efflux in promoting r…

Dissecting Alzheimer's proteomic landscape through NULISA profiling of brain cell-specific extracellular vesicles.

INTRODUCTION: Blood-based biomarkers are essential for early detection, monitoring, and therapeutic development in Alzheimer's disease (AD) and related dementia (ADRD), but current assays lack brain cell specificity and sensitivity to low-abundant proteins. METHOD: We isolated the following brain cell-derived small extracellular vesicles (sEV) from the plasma of individuals with normal cognition (CN), mild cognitive impairment (MCI), or ADRD: neurons (NDE), astrocytes (ADE), microglia (MDE), oligodendrocytes (ODE), pericytes (PDE), and endothelial cells (EDE). Using NULISAseq, we profiled 122…

Hepatic Stellate Cells Functional Heterogeneity in Liver Cancer.

Hepatic stellate cells (HSCs) are the liver's pericytes, and play key roles in liver homeostasis, regeneration, fibrosis, and cancer. Upon injury, HSCs activate and are the main origin of myofibroblasts and cancer-associated fibroblasts (CAFs) in liver fibrosis and cancer. Primary liver cancer has a grim prognosis, ranking as the third leading cause of cancer-related deaths worldwide, with hepatocellular carcinoma (HCC) being the predominant type, followed by intrahepatic cholangiocarcinoma (iCCA). Moreover, the liver hosts 35% of all metastatic lesions. The distinct spatial distribution and f…

Examining Stromal Cell Interactions in an In Vitro Blood-Brain Barrier Model with Human Umbilical Vein Endothelial Cells.

Understanding the function of the blood-brain barrier (BBB) in health and disease, as well as improving drug delivery across the BBB, remains a critical priority in neuroscience research. However, current in vitro models of the BBB have become increasingly complex and challenging to implement. In this study, we present a simplified microfluidic BBB model in which human umbilical vein endothelial cells (HUVECs) are cultured as a monolayer along a fibrin gel containing human pericytes and astrocytes. Remarkably, within just three days, the 3D co-culture significantly enhanced barrier formation a…

Endothelial cell MHC molecules are necessary and sufficient to reject 3D-printed human skin grafts in an advanced human immune system mouse.

Vascularized skins were 3D-printed using single donor human fibroblasts, pericytes, keratinocytes and endothelial cells (ECs), the latter either unmodified (WT-ECs) or deleted of MHC molecules (KO-ECs). Adult MISTRG6 immunodeficient mice neonatally inoculated with adult human hematopoietic stem cells (HSCs) received printed skin allogeneic to the HSCs and were boosted 3 weeks post-grafting with human peripheral blood mononuclear cells (PBMCs) autologous to the HSCs. HSC inoculation alone produced low levels of circulating human myeloid and lymphoid cells without affecting grafts; PBMC boosting…

Impact of direct oral anticoagulants on blood-brain barrier integrity.

INTRODUCTION: In cerebral infarction, major prognostic factors include hemorrhagic transformation and cerebral edema, both partially associated with blood-brain barrier (BBB) disruption. Anticoagulation therapy, especially with direct oral anticoagulants (DOACs), is linked to these outcomes, suggesting that the effect of DOACs on BBB integrity may influence stroke prognosis and recovery. METHODS: Two in vitro BBB models-a monolayer model and a co-culture model-were constructed using rat brain endothelial cells (RBECs), pericytes, and astrocytes. Each model was treated with one of three DOACs:…

Human pluripotent stem cell-derived fetal hepatic stellate cells promote vascularization and maturation in liver organoids.

Human-induced pluripotent stem cell (hiPSC)-derived liver organoids (LOs) are valuable for studying human liver organogenesis but face challenges in faithfully recapitulating certain processes, like vasculogenesis, due to the lack of specific cell components. Hepatic stellate cells (HSCs), which are liver-specific pericytes and might be crucial for liver vasculogenesis, remain underutilized in developmental studies because of their disease-related status and inefficient generation process. Here, we present an efficient method for generating hiPSC-derived HSCs (hiPSC-HSCs) resembling the transc…

Thyroid Hormone T4 Alleviates Traumatic Brain Injury by Enhancing Blood-Brain Barrier Integrity.

Traumatic brain injury (TBI) disrupts the blood-brain barrier (BBB), resulting in increased permeability, neuronal loss, and cognitive dysfunction. This study investigates the therapeutic potential of thyroid hormone (T4) to reduce BBB dysfunction following moderate fluid percussion injury. T4 injection (intraperitoneal) after TBI restores the levels of pericytes and endothelial cells vital for BBB integrity, reduces edema by downregulating AQP-4 gene expression, and enhances levels of the tight junction protein ZO-1. T4 counteracts the TBI-related increase in MMP-9 and TLR-4, significantly re…

Vascular motion in the dorsal root ganglion sensed by Piezo2 in sensory neurons triggers episodic pain.

Spontaneous pain, characterized by episodic shooting or stabbing sensations, is a major complaint among neuropathic pain patients, yet its mechanisms remain poorly understood. Recent research indicates a connection between this pain condition and "clustered firing," wherein adjacent sensory neurons fire simultaneously. This study presents evidence that the triggers of spontaneous pain and clustered firing are the dynamic movements of small blood vessels within the nerve-injured sensory ganglion, along with increased blood vessel density/angiogenesis and increased number of pericytes around blo…

Placental growth factor deficiency initiates obesity- and aging-associated metabolic syndrome.

Obesity often leads to inadequate angiogenesis in expanding adipose tissue, resulting in inflammation and insulin resistance. We explored the role of placental growth factor (PlGF) in metabolic syndrome (MS) using mice models of type 2 diabetes, high-fat diet, or aging. Reduced serum PlGF levels were associated with decreased insulin sensitivity and development of MS features. PlGF was localized within endothelial cells and pericytes of adipose tissue. In vitro, low PlGF levels in hypoxic conditions worsened oxidative stress, apoptosis, and reduced autophagy. This was associated with a reducti…

Heterogeneous fibroblasts contribute to fibrotic scar formation after spinal cord injury in mice and monkeys.

Spinal cord injury (SCI) leads to fibrotic scar formation at the lesion site, yet the heterogeneity of fibrotic scar remains elusive. Here we show the heterogeneity in distribution, origin, and function of fibroblasts within fibrotic scars after SCI in mice and female monkeys. Utilizing lineage tracing and single-cell RNA sequencing (scRNA-seq), we found that perivascular fibroblasts (PFs), and meningeal fibroblasts (MFs), rather than pericytes/vascular smooth cells (vSMCs), primarily contribute to fibrotic scar in both transection and crush SCI. Crabp2 + /Emb+ fibroblasts (CE-F) derived from …

Characteristics of blood-brain barrier heterogeneity between brain regions revealed by profiling vascular and perivascular cells.

The blood-brain barrier (BBB) protects the brain and maintains neuronal homeostasis. BBB properties can vary between brain regions to support regional functions, yet how BBB heterogeneity occurs is poorly understood. Here, we used single-cell and spatial transcriptomics to compare the mouse median eminence, one of the circumventricular organs that has naturally leaky blood vessels, with the cortex. We identified hundreds of molecular differences in endothelial cells (ECs) and perivascular cells, including astrocytes, pericytes and fibroblasts. Using electron microscopy and an aqueous-based tis…
