Reverse genetics platform for tomato mottle mosaic virus: infectious clone construction and resistance screening in tomato.

BACKGROUND: Recently identified as an emerging pathogen, tomato mottle mosaic virus (ToMMV), belonging to the Tobamovirus genus, has become a major concern for global tomato cultivation. To better understand its genetic characteristics and support future research, we investigated a newly collected ToMMV isolate from symptomatic tomato plants in China and aimed to develop a reliable infectious clone for downstream studies. METHODS AND RESULTS: A ToMMV-positive tomato sample from China was sequenced, revealing 98.5% nucleotide identity with a previously reported isolate from Shandong Province,

Ovulating Female of Nine-Spined Sticklebacks (Pungitius sinensis) Are Attracted to Substances Derived From Male Kidneys.

Male sticklebacks (Gasterosteidae) produce an adhesive protein called "spiggin" in the tubular epithelial cells of their kidneys, which they use to attach plants or plant debris together and build nesting sites. Spiggin is a glycoprotein excreted in urine. A similar substance has been reported in freshwater sculpins (Cottidae), and it is thought that male sculpins that nest under stones use this glycoprotein in their enlarged kidneys to attract females to their nests. Based on the phylogenetic closeness of sticklebacks and sculpins and the physiological similarities in their kidneys, we hypoth

Functional Characterization and Potential Regulatory Role of MdWRKY31 in Cold Tolerance.

Identifying cold-resistance genes is essential for improving the ability of apples (Malus × domestica) to tolerate low temperatures, as cold stress significantly limits their growth and productivity. The MdWRKY31 gene was cloned from apple, and its sequence characteristics, expression pattern, and biological function were systematically investigated. Bioinformatic analysis indicated that MdWRKY31 belongs to the group II WRKY transcription factors and is localized in the nucleus. Expression analysis revealed that MdWRKY31 transcript levels were markedly upregulated under low-temperature stress.

Generating drought- and salt-tolerant tomato germplasm through bioengineering of the ethylene response factor SlERF.J2.

Drought and salinity are key abiotic constraints limiting the growth, development, and yield of tomato (Solanum lycopersicum). Ethylene response factors are crucial regulators that govern plants' adaptation to diverse abiotic and biotic stress stimuli. Nevertheless, the exact role of SlERF.J2 in abiotic stress resistance remains unclear. In this study, clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein 9 gene-editing technology was used to generate slerf.j2 knockout tomato lines to clarify the biological function of SlERF.J2 in regulating tomato's resp

Fusion protein based strategies for developing insect-resistant crops.

Insect pest control in crop production incurs substantial economic costs annually on a global scale. Although broad-spectrum chemical pesticides were once considered the most effective solution, their overreliance has led to adverse effects on beneficial insects, human health, and the environment, as well as the development of pesticide-resistant insect populations. Consequently, there is an urgent need for alternative pest management strategies that minimize pesticide use and reduce unintended impacts on natural enemies, thereby maintaining ecological balance. Host plant resistance plays a pi

OsMDS1 regulates cell elongation and thermotolerance in rice.

Cereal crops often exhibit a tradeoff between thermotolerance, which is important for climate adaptation, and cell elongation, which is crucial for ideal plant architecture and yield. Here, we characterize the rice mutant with dwarfing and short roots/grains 1 (Osmds1), which shows dwarfism, shorter roots, and reduced grain size, along with enhanced thermosensitivity. OsMDS1 encodes a KNOTTED1-like homeobox (KNOX) transcription factor. A 15-bp deletion in the GSE domain of OsMDS1 is identified in Osmds1, which we show to be an allele of Osh15, d6, Oskn3, Shennong9816d, and rlb. OsMDS1 interact

Overexpression of FLZ12 Suppresses Root Hair Development and Enhances Iron-Deficiency Tolerance in Arabidopsis.

Background: The Arabidopsis FCS-LIKE ZINC FINGER (FLZ) family proteins play crucial roles in responses to various biotic and abiotic stresses, but the functions of many family members remain uncharacterized. Methods: In this study, we investigated the function of FLZ12, a member of the FLZ family, using a reverse genetic approach. Results: We found that overexpression of FLZ12 impaired root hair development, as evidenced by marked reductions in both root hair length and number under normal growth conditions. However, deprivation of phosphate could partially restore root hair formation, althoug

A Mini Review of Citrus Rootstocks and Their Role in High-Density Orchards.

Dwarfing is an important agricultural trait for intensive cultivation and effective orchard management in modern fruit orchards. Commercial citrus production relies on grafting with rootstocks that reduce tree vigor to control plant height. Citrus growers all over the world have been attracted to dwarfing trees because of their potential for higher planting density, increased productivity, easy harvest, pruning, and efficient spraying. Dwarfing rootstocks can be used to achieve high density. As a result, the use and development of dwarfing rootstocks are important. Breeding programs in several

From Plant to Yeast-Advances in Biosynthesis of Artemisinin.

Malaria is a life-threatening disease. Artemisinin-based combination therapy (ACT) is the preferred choice for malaria treatment recommended by the World Health Organization. At present, the main source of artemisinin is extracted from Artemisia annua; however, the artemisinin content in A. annua is only 0.1-1%, which cannot meet global demand. Meanwhile, the chemical synthesis of artemisinin has disadvantages such as complicated steps, high cost and low yield. Therefore, the application of the synthetic biology approach to produce artemisinin in vivo has magnificent prospects. In this review,

Biofortification of Cereals and Pulses Using New Breeding Techniques: Current and Future Perspectives.

Cereals and pulses are consumed as a staple food in low-income countries for the fulfillment of daily dietary requirements and as a source of micronutrients. However, they are failing to offer balanced nutrition due to deficiencies of some essential compounds, macronutrients, and micronutrients, i.e., cereals are deficient in iron, zinc, some essential amino acids, and quality proteins. Meanwhile, the pulses are rich in anti-nutrient compounds that restrict the bioavailability of micronutrients. As a result, the population is suffering from malnutrition and resultantly different diseases, i.e.

Genotyping by Sequencing Revealed QTL Hotspots for Trichome-Based Plant Defense in Gossypium hirsutum.

Cotton possesses certain physical features, including leaf and stem trichomes that help plants deter damage caused by insect pests, and to some extent, from abiotic factors as well. Among those features, trichomes (pubescence) hold a special place as a first line of defense and a managemental tool against sucking insect pests of cotton. Different insect pests of cotton (whiteflies, aphids, jassids, and boll weevil) severely damage the yield and quality of the crop. Likewise, whiteflies, aphids, jassids, and other insect pests are considered as potential carriers for cotton leaf curl viruses an

Quantifying the seasonal drivers of transmission for Lassa fever in Nigeria.

Lassa fever (LF) is a zoonotic disease that is widespread in West Africa and involves animal-to-human and human-to-human transmission. Animal-to-human transmission occurs upon exposure to rodent excreta and secretions, i.e. urine and saliva, and human-to-human transmission occurs via the bodily fluids of an infected person. To elucidate the seasonal drivers of LF epidemics, we employed a mathematical model to analyse the datasets of human infection, rodent population dynamics and climatological variations and capture the underlying transmission dynamics. The surveillance-based incidence data o

Ascorbic acid deficiency leads to increased grain chalkiness in transgenic rice for suppressed of L-GalLDH.

The grain chalkiness of rice (Oryza sativa L.), which determines the rice quality and price, is a major concern in rice breeding. Reactive oxygen species (ROS) plays a critical role in regulating rice endosperm chalkiness. Ascorbic acid (Asc) is a major plant antioxidant, which strictly regulates the levels of ROS. l-galactono-1, 4-lactone dehydrogenase (L-GalLDH, EC 1.3.2.3) is an enzyme that catalyzes the last step of Asc biosynthesis in higher plants. Here we show that the L-GalLDH-suppressed transgenic rice, GI-1 and GI-2, which have constitutively low (between 30% and 50%) leaf and grain

Analysis of the MdMYB1 gene sequence and development of new molecular markers related to apple skin color and fruit-bearing traits.

MdMYB1, a key transcription factor determining apple skin color, coordinately regulates genes in the anthocyanin pathway. In this study, we analyzed the MdMYB1 gene and its relationship to apple skin color and fruit-bearing traits to better understand this gene and its application to apple breeding. A previously reported MdMYB1 dCAPS marker failed to identify alleles of the MdMYB1 gene in 'Fuji', a very important apple cultivar. In this study, we revealed that the polymorphic site related to the MdMYB1 dCAPS marker is heterozygous in 'Fuji'. In addition, two new polymorphic sites related to ap

Regulatory divergence of homoeologs underlies network optimization for fiber improvement in domesticated cotton.

Polyploidy is prominent in plant evolution and in many of the world's most important crops, yet how domestication reshapes the regulation of duplicated genes (homoeologs) to generate superior agronomic traits remains incompletely understood. Here, we integrate population genomics, stage-resolved transcriptomics, expression quantitative trait locus (eQTL) mapping, and coexpression network analysis across 161 semiwild and 376 cultivated accessions of allotetraploid cotton (Gossypium hirsutum) to dissect the regulatory consequences of domestication. We show that domestication increases both the f

The OsUVR8-OsNAC3-OsERF117 signaling module mediates metabolic acclimation and climate adaptation in rice.

Ultraviolet-B (UV-B; 280 to 315 nanometers) radiation increasingly threatens crop productivity, yet the genetic basis of plant adaptation remains poorly understood. We delineate a UV-B signaling module in rice that links photoreceptor activation to transcriptional reprogramming and metabolic acclimation. The AP2/ERF transcription factor OsERF117 acts as a central regulator, directly activating flavonoid and melatonin biosynthetic genes to drive photoprotective metabolite accumulation and enhance UV-B stress tolerance. Promoter variation in OsERF117 defines 10 haplotypes across 4093 rice access

Trichome acylsugar defenses are equally effective against leaf-mining and free-living insects.

Trichomes, epidermal projections, are crucial components of plant defense systems. Whether these epidermal defenses are effective against leafminers, which infest the internal mesophyll layers, remains unclear. Using the devastating invasive tomato pest, Phthorimaea absoluta (tomato leafminer), and the trichome-rich wild tomato, Solanum pennellii, we demonstrate that trichome-produced acylsugars effectively inhibit the growth of endophagous P. absoluta larvae feeding inside leaves. Chromatin accessibility directly governs flavonoid biosynthesis and indirectly orchestrates cannabinoid production in Cannabis.

Secondary metabolites in hemp enhance its pharmaceutical and nutraceutical value, yet the epigenetic regulatory network underlying secondary metabolite biosynthesis remains poorly understood in hemp. Here, we profiled the inflorescences of two cultivars with different trichome density by integrating metabolomics, transcriptomics and ATAC-seq. Multi-omics data revealed pronounced differences in metabolites (491 differentially accumulated metabolites (DAMs)), transcripts (8343 differentially expressed genes (DEGs)), and chromatin accessibility (11376 different accessibility genes, (DAGs)) betwee
