Adeno-associated virus (AAV) gene therapy is frequently hindered by dose-limiting toxicities and preexisting neutralizing antibodies (nAbs). Here, we evaluate enhancer polymer (ePL), a cargo-less poly(L-lactic-co-glycolic acid) nanoparticle, the systemic administration of which demonstrated biocompatibility in nonhuman primates (NHPs) and generated a protransduction serum secretome that significantly enhanced AAV uptake in human iPSC-derived cardiomyocytes. Proteomic analysis of human peripheral blood mononuclear cell-derived secretomes identified a transient upregulation of AAV-entry factors,…

Ischaemic stroke is one of the leading causes of disability and death worldwide, yet effective treatment options remain limited. Bilateral hemisphere theta burst stimulation (TBS), a non-invasive brain stimulation technique, has shown considerable therapeutic potential in stroke; however, relevant research is still limited, and the specific mechanisms remain unclear. This study used a middle cerebral artery occlusion (MCAO) model in non-human primates to explore the application of bilateral hemispheric TBS in stroke rehabilitation. By integrating behavioural assessments, electrophysiology, fun…

Both the timing and composition of food intake have gained recognition for their effects on circadian (daily) rhythms in mammals, however the mechanisms by which these effects are exerted are incompletely understood. The present studies focus on synchronization of circadian rhythms to the environmental light-dark cycle, particularly re-entrainment following a phase shift such as occurs with travel across time zones or shift work. To examine the role and therapeutic potential of the peptide hormone leptin in these effects, we create and characterize injectable leptin factories consisting of enc

The cerebellum shapes distributed motor and association networks through precisely organized pathways linking the cerebellar cortex to the deep cerebellar nuclei (DCN), its principal output structures. Whether these cortical and nuclear compartments scale proportionally across primates, and how their relative expansion relates to cerebellar organization, remains unresolved. Here, we generate multimodal, high-resolution cross-species 3D cerebellar atlases in marmoset, macaque, and human by integrating iron-sensitive and diffusion MRI (MAP-MRI) with histological markers, enabling direct delineat…

Strongyloides stercoralis nematode infection occurs in ≈600 million persons worldwide and is listed by the World Health Organization as a neglected tropical disease. Understanding zoonotic potential is critical, especially in areas where humans, domestic animals, and wildlife interact. We explored cross-species sharing of Strongyloides roundworms by analyzing fecal samples from humans, dogs, and nonhuman primates in the Dzanga-Sangha Protected Areas, Central African Republic. We detected positive samples by quantitative PCR and assessed genetic diversity through amplification of the 18S rRNA H…

Yellow fever (YF) remains a major zoonotic disease in tropical regions of the Americas and Africa. In Brazil, surveillance of epizootics in non-human primates (NHPs) has become a cornerstone of YF prevention and control. This study provides a historical overview of NHP epizootic surveillance in Brazil from 1999 to 2025 and evaluates its role as an early warning system for human infections within a One Health framework. A retrospective descriptive epidemiological study was conducted using surveillance data from the Notifiable Diseases Information System (SINAN) and the Wildlife Health Informati…

BACKGROUND: The ways in which Mycobacterium tuberculosis (Mtb) infection affects host metabolism and its implications for disease pathogenesis remain poorly understood. Understanding how Mtb shapes the metabolic state of the host could lead to nutritional and host-directed therapies to improve TB outcomes.
METHODS: We performed high-resolution metabolic and lipid profiling on male rhesus macaques infected with Mtb (n = 17). Serial plasma samples were analysed beginning at the time of infection and up to 15-16 weeks post infection. Observed host metabolic changes were examined in a cohort of M

Male fertility is complex and influenced by genetic, hormonal, environmental and lifestyle factors. However, limitations to human studies necessitate the use of reliable preclinical models to better understand the underlying mechanisms of male fertility. Rhesus macaques (Macaca mulatta), with their close genetic and physiological similarities to humans, offer an invaluable model for male reproductive health studies. The suitability of rhesus macaques for studying male infertility is based on similarities in spermatogenesis, hormonal cycles and the way in which assisted reproductive technologie

In preclinical models, indwelling intravenous catheters and vascular access ports are often essential components of biomedical research aimed at modeling human disease. For instance, animal models of drug self-administration are used for many reasons, including to assess abuse liability, to study physiologic and neurologic consequences of drug exposure, and to examine the efficacy of behavioral and/or pharmacological interventions. The most frequent route of drug self-administration in preclinical animal models is the intravenous route via indwelling intravenous catheters. The present study ex

Demyelination and remyelination play key roles in spinal cord injury (SCI), affecting the recovery of motor and sensory functions. Research in rodent models is extensive, but the study of these processes in non-human primates is limited. Therefore, our goal was to thoroughly study the histological features of demyelination and remyelination after contusion injury of the cervical spinal cord in Macaca fascicularis. In a previous study, we created an SCI model in M. fascicularis by controlling the contusion displacement. We used Eriochrome Cyanine staining, immunohistochemical analysis, and tolu…

Yellow fever (YF) causes severe morbidity and mortality in Africa and South America. It is an arthropod-borne viral disease endemic to tropical regions of Africa and South America. Yellow fever virus (YFV) is transmitted by mosquitoes and frequently affects both non-human primates (NHPs) and humans. Neotropical primates (NTPs) are generally more severely afflicted by YFV than African primates. Asian primates appear not to be susceptible to this disease. Susceptibility varies among NTP species: asymptomatic infections are described in some NTP species, whereas severe epizootic mortality events …

Visual object memory is a fundamental element of various cognitive abilities, and the underlying neural mechanisms have been extensively examined especially in the anterior temporal cortex of primates. However, both macroscopic large-scale functional network in which this region is embedded and microscopic neuron-level dynamics of top-down regulation it receives for object memory remains elusive. Here, we identified the orbitofrontal node as a critical partner of the anterior temporal node for object memory by combining whole-brain functional imaging during rest and a short-term object memory …

In the last decade, geopolitical instability across the globe has increased the risk of a large-scale radiological event, when radiation biomarkers would be needed for an effective triage of an irradiated population. Ionizing radiation elicits a complex response in the proteome, genome, and metabolome and hence can be leveraged as rapid and sensitive indicators of irradiation-induced damage. We analyzed the plasma of total-body irradiated (TBI) leukemia patients (n = 24) and nonhuman primates (NHPs; n = 10) before and 24 h after irradiation, and we performed a global metabolomic study aiming t…

PURPOSE: The sphingosine-1-phosphate receptor-1 (S1PR1) is involved in regulating responses to neuroimmune stimuli. There is a need for S1PR1-specific radioligands with clinically suitable brain pharmcokinetic properties to complement existing radiotracers. This work evaluated a promising S1PR1 radiotracer, [18F]TZ4877, in nonhuman primates.
PROCEDURES: [18F]TZ4877 was produced via nucleophilic substitution of tosylate precursor with K[18F]/F- followed by deprotection. Brain PET imaging data were acquired with a Focus220 scanner in two Macaca mulatta (6, 13 years old) for 120-180 min followin…

Radiation injury, either from radiotherapy or a mass-casualty event requires a health care system that can efficiently allocate resources to patients. We conducted a comprehensive transcriptome analysis of whole blood from a nonhuman primate model that received upper thoracic radiation (9.8-10.7 Gy). Blood samples were collected at multiple time points, extending up to 270 days post-irradiation with a minimum n = 6 for initial time points (Day 3-Day 40) and a total number of n = 28 primates. No males receiving the higher dose survived to Day 270. Using the Elastic Net model in R we found that …

Early life adverse experiences, including childhood maltreatment, are major risk factors for psychopathology, including anxiety disorders with dysregulated fear responses. Consistent with human studies, maltreatment by the mother (MALT) leads to increased emotional reactivity in rhesus monkey infants. Whether this persists and results in altered emotion regulation, due to enhanced fear learning or impaired utilization of safety signals as shown in human stress-related disorders, is unclear. Here we used a rhesus model of MALT to examine long-term effects on state anxiety and threat/safety lear

IN BRIEF: Minipuberty is a transient activity period of the hypothalamic-pituitary-gonadal axis in the postnatal and infant period including surging serum concentrations of reproductive hormones. Increasing evidence points to an important role of this period for maturation of the testes and thereby for male reproductive function.

INTRODUCTION: The primary objective of this article was to determine if any histological alterations occur in the round window (RW) and adjacent anatomical structures of an animal model with normal hearing when a cochlear implant (CI) electrode array is inserted. Furthermore, this article aims to relate these histological alterations to the auditory changes generated.
METHODS: Cochlear implantation was performed, following the principles of minimally traumatic surgery (MTS), in 15 ears of nonhuman primates (Macaca fascicularis) (Mf) with normal hearing. Auditory brainstem-evoked potentials (A…

PURPOSE: To determine whether short-latency changes in multifocal electroretinography (mfERG) observed in experimental glaucoma (EG) are secondary solely to retinal ganglion cell (RGC) loss or whether there is a separate contribution from elevated intraocular pressure (IOP).
METHODS: Prior to operative procedures, a series of baseline mfERGs were recorded from six rhesus macaques using a 241-element unstretched stimulus. Animals then underwent hemiretinal endodiathermy axotomy (HEA) by placing burns along the inferior 180° of the optic nerve margin in the right eye (OD). mfERG recordings were
