Clinical Effectiveness of an Adaptive Treatment Planning Algorithm for Intensity Modulated Radiation Therapy Versus 3D Conformal Radiation Therapy for Node-Positive Breast Cancer Patients Undergoing Regional Nodal Irradiation/Postmastectomy Radiation Therapy.

PURPOSE: Clinical trials support adjuvant regional nodal irradiation (RNI) after breast-conserving surgery or mastectomy for patients with lymph node-positive breast cancer. Advanced treatment planning techniques (eg, intensity modulated radiation therapy [IMRT]) can reduce dose to organs at risk (OARs) in this situation. However, uncertainty persists about when IMRT is clinically indicated (vs 3-dimensional conformal radiation therapy [3DCRT]) for RNI. We hypothesized that an adaptive treatment planning algorithm (TPA) for IMRT adoption would allow OAR constraints for RNI to be met when 3DCRT

Therapeutic and prophylactic effects of radiation therapy in the management of recurrent granulation tissue induced tracheal stenosis: a review on the role of Endobronchial brachytherapy and external beam radiation therapy.

PURPOSE: Granulation tissue-induced tracheal stenosis (mainly secondary to intubation or lung transplantation) is one of the most common etiologies of benign airway obstructions. Recurrence rates after standard treatment options (surgical resection and/or endobronchial interventions) can inadvertently worsen the stricture through the stimulation of more granulation tissue generation (via increased fibroblast activity and roliferation). Low-dose radiotherapy could be a promising tool to prevent granulation tissue formation after surgery and/or endobronchial interventions regarding its establish

Machine learning simulation for radiation effects on MoS₂-Al₂O₃ hybrid nanoliquid with local thermal non-equilibrium conditions.

The current study examines the Soret-Dufour influence on a MoS₂-Al₂O₃ hybrid nanomaterial with LTNECs, using a novel intelligent numerical computing paradigm. The current study uses a simple mathematical model to investigate the features of heat transmission in the absence of LTECs. It can help forecast and optimize medication delivery since temperature and concentration gradients can better direct nanoparticles in blood. The model also aids in understanding heat and mass transmission in stenosed arteries, which is critical for designing medical remedies like hyperthermia therapy and preventin

Rational modulation of immune mechanisms synergizes the anti-tumor effects of targeted radiation therapy in pre-clinical models.

Immunotherapy has revolutionized cancer treatment, offering new hope for many patients. However, while some individuals show remarkable responses, the overall success rate remains limited. This has spurred interest in combination therapies, particularly with established treatments like radiation therapy (RT), to improve outcomes. RT is a cornerstone of cancer therapy and known to influence the immune landscape, yet a systematic characterization of its effects on tumor-infiltrating leukocytes (TILs) and a rationale-based therapy is still lacking. In this study, we employed a diverse set of pre-

Lipidomics of longer-term effects in the murine model after exposure to mixed field radiation and combined infection.

BACKGROUND: Accurate biodosimetry is essential for medical response during nuclear emergencies. However, mixed-field radiation and secondary infections create significant diagnostic challenges. OBJECTIVE: This study aimed to characterize persistent serum lipidomic alterations following mixed-field exposures at long-term time points and evaluate the impact of a delayed secondary infection on radiation-associated signatures to determine the feasibility of serum biomarkers for complex biodosimetry. METHODS: Target serum lipidomics and untargeted metabolomics were performed on male C57BL/6 mice

Effects of probiotics BB-12 and Bifico on radiation-induced brain injury, cognitive dysfunction and gut microbiota composition.

BACKGROUND: Whole brain irradiation (WBI) is an essential treatment for brain tumors and metastases, but often leads to radiation-induced brain injury (RIBI), and cognitive dysfunction, significantly impairing patients' quality of life. Emerging evidence highlights the role of gut microbiota dysbiosis in neurological disorders via the microbiota-gut-brain axis (MGBA). This study investigated the neuroprotective effects of probiotics Bifidobacterium animalis subsp. lactis BB-12 (BB-12) and Bifico on WBI-induced cognitive impairment in mice. METHODS AND RESULTS: After 4 weeks of BB-12 and Bific

Early radiation effects on immediate breast reconstruction: A comparative analysis of 15- and 5-fraction postmastectomy radiotherapy regimens.

BACKGROUND: Modern postmastectomy radiotherapy (PMRT) increasingly employs ultra-hypofractionated regimens (26 Gy/5 fractions) as an alternative to standard hypofractionation (15 fractions/40-42 Gy). However, evidence on their safety in the reconstructive setting remains limited. This study compares acute toxicity outcomes between these regimens and a non-irradiated control group following immediate breast reconstruction. METHODS: A single-center cohort study included 203 immediate breast reconstructions in 168 patients (implant = 59, DIEP = 47, LD = 33, BCS = 64) performed between July 2024

Protective effects of cystine and theanine on radiation-induced gastrointestinal syndrome: Modulation of apoptosis, pyroptosis, and crypt survival.

Acute radiation exposure causes severe colonic damage and gastrointestinal (GI) syndrome. Although cystine and theanine (CT) may ameliorate 5 Gy X-ray (0.5531 Gy/min) radiation-induced damage of CT-treated rats (280 mg/kg for 5 days), their precise mechanisms in protecting colonic crypt cells and maintaining epithelial integrity remain unclear. Using a rat model of radiation-induced GI injury following 8-14 Gy exposure, we explored CT treatment effects on crypt survival, apoptosis, DNA damage, and inflammation. CT treatment eliminated bloody diarrhea (p = 0.0075) and decreased ulceration to 6.

Modeling the effects of radiation on the bone tumor microenvironment: opportunities for exploring combination therapies in microphysiologic systems.

Primary bone tumors and bone metastases represent significant challenges in oncology. Radiotherapy is an important adjuvant treatment for several primary bone and musculoskeletal tumors, as well as for palliative care for metastatic bone lesions. While effective in these applications, patients receiving skeletal radiation face a lifelong risk of fragility fracture at the irradiated sites, among other complications. Damage to bone could be reduced by development of tumor-selective radiosensitizers that would enhance the efficacy of radiotherapy, resulting in reducing the radiation dose delivere

Obacunone Photoprotective Effects against Solar-Simulated Radiation-Induced Molecular Modifications in Primary Keratinocytes and Full-Thickness Human Skin.

Solar radiation can cause damage to the skin, leading to various adverse effects such as sunburn, reactive oxygen species production, inflammation, DNA damage, and photoaging. To study the potential of photoprotective agents, full-thickness skin models are increasingly being used as in vitro tools. One promising approach to photoprotection involves targeting the redox-sensitive transcription factor Nrf2, which is responsible for regulating various cellular defense mechanisms, including the antioxidant response, inflammatory signaling, and DNA repair. Obacunone, a natural triterpenoid, has been

Review of compounds that exhibit radioprotective and/or mitigatory effects after application of diagnostic or therapeutic ionizing radiation.

PURPOSE: Exposure to ionizing radiation can be accidental or for medical purposes. Analyzes of the frequency of radiation damage in the general population, it has been determined that by far the most common are those that occur as a result of diagnostic or therapeutic procedures. Damage caused by radiation, either accidentally or for therapeutic purposes, can be reduced by the use of radioprotectors, mitigators or other therapeutic agents. A detailed research of the available literature shows that there is little systematized data of potentially radioprotective and/or mitigating effects of dru

Effects of Gamma-Tocotrienol on Intestinal Injury in a GI-Specific Acute Radiation Syndrome Model in Nonhuman Primate.

The gastrointestinal (GI) system is highly susceptible to irradiation. Currently, there is no Food and Drug Administration (FDA)-approved medical countermeasures for GI radiation injury. The vitamin E analog gamma-tocotrienol (GT3) is a promising radioprotector in mice and nonhuman primates (NHP). We evaluated GT3-mediated GI recovery in total-body irradiated (TBI) NHPs. Sixteen rhesus macaques were divided into two groups; eight received vehicle and eight GT3 24 h prior to 12 Gy TBI. Proximal jejunum was assessed for structural injuries and crypt survival on day 4 and 7. Apoptotic cell death
