Abstract Swallowing of amniotic fluid by late gestation fetuses increases when amniotic fluid volume (AFV) is elevated. Our objectives were to quantitatively characterize fetal swallowing when AFV is elevated above normal to polyhydramniotic levels and to explore the mechanisms that mediate these changes. BACKGROUND & AIMS: The gastric mucosa must have efficient protective mechanisms to maintain physiological intracellular pH. The aim of this study was to investigate the effect of low luminal pH on apical membrane permeability.
METHODS: Chambered Necturus antral mucosa was perfused with Ringer's/95% O2-5% CO2 at pH 7.25. The mucosal side was exposed to pH 4.0-2.0 with four microelectrodes placed in surface cells. Current methods for glucose-stimulated insulin secretion (GSIS) in rodents involve ex vivo perifusion or static incubation of pancreatic islets and pancreatic slices. Ex vivo perfusion and static incubation allow for multiple samples to be taken quickly with efficiency. However, these procedures subject the islets to trauma, intermittent hypoxia, and reduced paracrine insulin signaling, all of which can affect GSIS. These negative effects can be minimized via perfusion through the mice vasculature with an intact pancreas, which provides another comparison method for the different GSIS models.
