Combined transcriptome and metabolome analyses of metformin effects reveal novel links between metabolic networks in steroidogenic systems.

Metformin is an antidiabetic drug, which inhibits mitochondrial respiratory-chain-complex I and thereby seems to affect the cellular metabolism in many ways. It is also used for the treatment of the polycystic ovary syndrome (PCOS), the most common endocrine disorder in women. In addition, metformin possesses antineoplastic properties. Although metformin promotes insulin-sensitivity and ameliorates reproductive abnormalities in PCOS, its exact mechanisms of action remain elusive. Therefore, we studied the transcriptome and the metabolome of metformin in human adrenal H295R cells. Microarray an

Amplification of R-spondin1 signaling induces granulosa cell fate defects and cancers in mouse adult ovary.

R-spondin1 is a secreted regulator of WNT signaling, involved in both embryonic development and homeostasis of adult organs. It can have a dual role, acting either as a mitogen or as a tumor suppressor. During ovarian development, Rspo1 is a key factor required for sex determination and differentiation of the follicular cell progenitors, but is downregulated after birth. In human, increased RSPO1 expression is associated with ovarian carcinomas, but it is not clear whether it is a cause or a consequence of the tumorigenic process. To address the role of Rspo1 expression in adult ovaries, we ge

Disentangling neural processes of egocentric and allocentric mental spatial transformations using whole-body photos of self and other.

Mental imagery of one's body moving through space is important for imagining changing visuospatial perspectives, as well as for determining how we might appear to other people. Previous neuroimaging research has implicated the temporoparietal junction (TPJ) in this process. It is unclear, however, how neural activity in the TPJ relates to the rotation perspectives from which mental spatial transformation (MST) of one's own body can take place, i.e. from an egocentric or an allocentric perspective. It is also unclear whether TPJ involvement in MST is self-specific or whether the TPJ may also be

From models to mechanisms: odorant communication as a key determinant of social behavior in rodents during illness-associated states.

Pheromones and other social odor cues convey rich information among rodents. Social investigation is described as a key element in olfactory communication, which involves motivated approaches to conspecifics and other socially relevant stimuli. This behavior is activated by the detection of social cues to gather information about conspecifics for subsequent strategies such as avoidance or further approach, thereby determining the extent and nature of physical contact that ensues. This feature indicates a useful way for describing the process of social communication in distance-based manner. In

Osteological and dental markers of health in the transition from the Late Antique to the Early Medieval period in Croatia.

The purpose of this study is to analyze health at the transition from the Late Antique (LA) to the Early Medieval (EM) period in Croatia. Results of the analyses of skeletal remains are compared with historical and archaeological data to test the hypothesis that the transition was catastrophic. An additional objective is to determine whether the transition was a uniform process, or differentially affected the past inhabitants of Croatia because of various local considerations. To accomplish this, four markers of health: cribra orbitalia, linear enamel hypoplasia, nonspecific periostitis, and t

Markers of antioxidant defence system and lipid peroxidation in peripheral blood of female patients with chronic idiopathic urticaria.

Oxidative stress is an important event in lesional skin of patients with chronic idiopathic urticaria (CIU). In the present study, we assessed blood oxidant/antioxidant status of patients suffering from CIU with positive response to autologous serum skin test (ASST) and with negative ASST, to improve our understanding of biological processes and the part of oxidative stress in this disease. Activities of manganese superoxide dismutase (MnSOD), copper-zinc superoxide dismutase (Cu/ZnSOD), glutathione peroxidase (GSH-PX), and catalase (CAT) as indices of enzymatic antioxidant capacity, as well a

Placental Insulin-like Growth Factor 1 Insufficiency Drives Neurodevelopmental Disorder-Relevant Behavioral Changes with Sex-Specific Vulnerabilities.

Preterm birth, placental insufficiency, and other perinatal adversities lead to the loss of placental support including critical hormones, such as Insulin-like growth factor 1 (IGF1), required for neurodevelopment. Decreased IGF1 and preterm birth are associated with neurodevelopmental disorder risk, including autism spectrum disorder. Whether placental Igf1 insufficiency drives neurodevelopmental risks is not understood. To understand these mechanisms, placental-targeted CRISPR manipulation in mice was employed to induce placental Igf1 insufficiency. Subsequently, embryonic forebrain developm

Histopathological Features of Ossified Keloids and Their Association With Recurrence: The Role of Inflammation and Foreign Bodies.

Heterotopic ossification within keloid tissue is an uncommon but clinically relevant phenomenon. However, its histopathological characteristics and etiological mechanisms remain poorly understood. To investigate the histological features of ossified keloids and assess potential contributing factors, with a focus on chronic inflammation and foreign body reaction. We retrospectively analyzed 39 histologically confirmed ossified keloids and compared them with two non-ossified control cohorts: a frequency-matched group (n = 78) adjusted for age, sex, and anatomical site, and a broader institutiona

SPATIO-TEMPORAL VARIATION OF PROCHRISTIANELLA SP. (CESTODA: TRYPANORHYNCHA) IN THE HOST OCTOPUS MAYA (CEPHALOPODA: OCTOPODA) AND ITS RELATIONSHIP WITH THE REPRODUCTION.

Although harvest of Octopus maya (Mexican four-eyed octopus) is one of the most important fisheries in the Yucatan Peninsula, little is known about the parasites of these cephalopods and how they affect host physiological processes. We analyzed the spatio-temporal variation of infection of O. maya by the cestode larva Prochristianella sp. (found in the anterior salivary glands) and its relationship with the gonad development stages (GDSs) of the host. From August 2009 to May 2010, 439 specimens of O. maya were caught in 2 localities on the northern coast of the Yucatán Peninsula: Dzilam de Bra

Influence of sex on global myocardial ischemia tolerance and mitochondrial function in circulatory death donor hearts.

Donation after circulatory death (DCD) donor hearts are not routinely used for heart transplantation (HTx) because of ischemic damage, which is inherent to the DCD process. HTx outcomes are suboptimal in males who received female donor hearts. The exact mechanism for suboptimal outcomes from female donor hearts has not been defined. Differential susceptibility to ischemia tolerance, which would play a significant role in DCD donation, could be a reason but has not been studied. We studied the influence of sex on global myocardial ischemia tolerance and mitochondrial function. Sprague-Dawley ra

Intersectional effects of race/ethnicity, sex, and APOE4 on plasma inflammatory profiles in Hispanic and non-Hispanic white adults.

BACKGROUND: Despite known links between inflammation and Alzheimer's disease (AD), little is understood about how inflammatory processes vary across racial/ethnic and sex groups, particularly in relation to the major genetic risk factor apolipoprotein E ε4 (APOE4), whether these differences contribute to AD disparities among understudied Hispanics. OBJECTIVE: This study investigated the interactive effects of race/ethnicity, sex, and APOE4 status on plasma inflammatory markers (IL-5, IL-6, IL-10, TNF-α). Secondary analyses examined whether these inflammatory markers were associated with cogni

Establishment of Serum Cholesterol Sulfate Reference Intervals and Evaluation of Its Diagnostic and Metabolic Associations in Chinese Adults.

Background/Objectives: Cholesterol sulfate (CS) is a multifunctional signaling molecule implicated in diverse physiological and pathological processes. Its clinical translation as a biomarker is hindered by the lack of established reference intervals, unclear disease-specific alterations, and undefined relationships with routine laboratory parameters. This study aimed to establish a serum CS reference range for Chinese populations, evaluate its associations with various diseases and clinical laboratory parameters, and define its clinical diagnostic utility. Methods: A total of 642 subjects wer

Annexin A6 in Eye-Lavage Extracellular Vesicles-a Candidate Biomarker for Sex-Specific Differences in Mild Hypoxic-Ischaemic Brain Damage-Findings from an Explorative Neonatal Mouse Model Study.

Extracellular vesicles (EVs) play important roles in cell communication, including via transfer of cargo proteins, and are promising biomarkers for pathobiological processes. Reliable biomarkers for mild neonatal hypoxia ischaemia (HI) are scarce. Wrongly determined HI severity often leads to adverse long-term neurological outcomes, with more detrimental effects in males. Therefore, identification of sex-specific markers for HI is of great interest. We recently reported the potential of EVs from eye lavage as novel non-invasive biomarkers of neonatal HI brain injury, differing in proteomic car

Temperature-Dependent Alternative Splicing of Precursor mRNAs and Its Biological Significance: A Review Focused on Post-Transcriptional Regulation of a Cold Shock Protein Gene in Hibernating Mammals.

Multiple mRNA isoforms are often generated during processing such as alternative splicing of precursor mRNAs (pre-mRNA), resulting in a diversity of generated proteins. Alternative splicing is an essential mechanism for the functional complexity of eukaryotes. Temperature, which is involved in all life activities at various levels, is one of regulatory factors for controlling patterns of alternative splicing. Temperature-dependent alternative splicing is associated with various phenotypes such as flowering and circadian clock in plants and sex determination in poikilothermic animals. In some s

The Clinic of Identifications in the Different Processes of Metamorphosis Into Woman.

This article examines, from a psychoanalytical perspective, the function of identification in the relationship between the subject of the unconscious and his body, his body image, and the other. To this effect, the article leans on the clinic of the metamorphosis into a woman in psychosis, both in the way that it is presented by patients in the context of treatment, and in the form of testimonies extracted from literature. It demonstrates how specular identification allows the subject to unify himself, so long as there is an avoidance of possible deformations of the psychical body, including f

Microarray expression profile analysis of long noncoding RNAs in premature brain injury: A novel point of view.

Long noncoding RNAs (lncRNAs) are abundant in the central nervous system and have a key role in brain function as well as many neurological disorders. However, the regulatory function of lncRNAs in the premature brain has not been well studied. This study described the expression profile of lncRNAs in premature mice using microarray technology. 1999 differentially expressed lncRNAs and 955 differentially expressed mRNAs were identified. Gene Ontology (GO) and pathway analysis showed that these lncRNAs were involved in multiple biological processes, including the nervous system development and
