Mitochondrial fumarate inhibits Parkin-mediated mitophagy.

Here, we explore the potential involvement of fumarate, a metabolite generated from the TCA cycle, as a key regulator of PINK1-Parkin-mediated mitophagy. Fumarate engages in a process called succination, forming S-(2-succino) cysteine with protein cysteine residues. Our research demonstrates that this modification specifically targets the sulfhydryl group of cysteine 323 and 451 residues of human Parkin, leading to the inhibition of its mitochondrial localization and E3 ligase activity, thereby impeding PINK1-Parkin-mediated mitophagy. Notably, our investigation reveals that the succinatable c

Nerve and behavioral responses of mice to various umami substances.

Food contains various taste substances. Among them, umami substances play an important role with regard to the perception of the taste of food, but, few studies have examined the taste characteristics of representative umami substances other than monosodium L-glutamate (MSG). By conducting mouse behavioral studies (the 48-h 2-bottle preference test and the conditioned taste aversion test) and assessing gustatory nerve responses, we investigated the taste characteristics of unique umami substances, including sodium succinate, L-theanine, betaine, and the enantiomer of MSG, D-MSG. Furthermore, w

Impairment of energy metabolism in hippocampus of rats subjected to chemically-induced hyperhomocysteinemia.

Homocystinuria is an inherited metabolic disease biochemically characterized by tissue accumulation of homocysteine (Hcy). Mental retardation, ischemia and other neurological features, whose mechanisms are still obscure are common symptoms in homocystinuric patients. In this work, we investigated the effect of Hcy administration in Wistar rats on some parameters of energy metabolism in the hippocampus, a cerebral structure directly involved with cognition. The parameters utilized were 14CO2 production, glucose uptake, lactate release and the activities of succinate dehydrogenase and cytochrome

Oxygen dependence of flux control of cytochrome c oxidase -- implications for mitochondrial diseases.

The oxygen dependence of cytochrome c oxidase control on succinate oxidation was investigated in saponin-permeabilized muscle fibers and isolated mitochondria from mouse quadriceps muscle applying metabolic control analysis. For this cyanide titrations of the oxygen consumption in the presence of succinate+rotenone were performed at different oxygen concentrations in the medium. While with isolated mitochondria high flux control coefficients were detected only at oxygen concentrations close to the KM value of cytochrome c oxidase, with saponin-permeabilized fibers a significant increase of cyt

[Correlation of changes in the electrical activity and indices of tissue respiration of brain structures in rabbits exposed systematically to vibration].

The electrical activity and tissue respiration of the cortex and subcortex (posterior hypothalamic nucleus, Deiters' vestibular nucleus) have been studied in the rabbits during 3-month exposure to vibration (60H2, for 3h daily). On days 15-30 of exposure to vibration the electrocorticogram demonstrated the prevalence of alfa-waves whereas in the subcortex the teta-waves were dominated. On long-term vibration exposure (2-3 months), the synchronized slow waves were recorded on the EEG. Comparison of a changed electrical activity of the various structures of the brain with the level of oxidizing

Sarcoma-180 cells are more sensitive to heat than are mouse normal tissues: esophagus, stomach, small intestine, large intestine, liver, spleen, and kidney.

Sensitivity of various mouse tissues to heat was determined using mouse sarcoma-180 (S-180) cells and normal tissues: esophagus, stomach, small intestine, large intestine, liver, spleen, and kidney. The in vitro succinate dehydrogenase inhibition (SDI) test was used. The succinate dehydrogenase (SD) activity of tissue fragments was assayed, following exposure to a temperature of 43 degrees C (heat treatment) or 37 degrees C (control) for 1, 2, 5, or 10 hr. The sensitivity to heat treatment was estimated by the percentage of SD activity of the heat-treated cells, compared to that of the control

The chemical conditions on the parent body of the Murchison meteorite: some conclusions based on amino, hydroxy and dicarboxylic acids.

Amino and hydroxy acids have been identified in the Murchison meteorite. Their presence is consistent with a synthetic pathway involving aldehydes, hydrogen cyanide and ammonia in an aqueous environment (Strecker-cyanohydrin synthesis). From the various equilibrium and rate constants involved in this synthesis, four independent estimates of the ammonium ion concentrations on the parent body at the time of compound synthesis are obtained; all values are about 2 x 10(-3) M. Succinic acid and beta-alanine have also been detected in the Murchison meteorite. Their presence is consistent with a synt

Driving impact of organic acids on microbial community dynamics and fermentation performance in Huangjiu.

Huangjiu, a traditional Chinese rice wine, relies on microbial communities for its quality, but the role of organic acids during fermentation is unclear. This study uses flavoromics and metatranscriptomics to explore how organic acids shape microbial composition and metabolism. Comparing samples with high (H) and low (L) organic acid content, 44 flavor compounds were identified, revealing significant group differences. Organic acids altered fungal communities: succinic acid positively correlated with Saccharomyces and Thermomyces, while lactic and tartaric acids showed negative correlations. S

Dysregulated calcium signaling underlies hyposalivation and microbial dysbiosis in Down syndrome.

Individuals with Down syndrome (DS) produce less saliva for unknown reasons resulting in chronic periodontal disease with systemic detrimental effects. Using the (Dp(16)1Yey) mouse model of DS we define the molecular mechanisms of hyposalivation and potential links to periodontal disease. We show that Dp(16)1Yey mice produce less saliva and have a higher immune burden in the salivary glands. We demonstrate that store operated calcium entry (SOCE), required for saliva secretion, is deficient in the salivary glands of Dp(16)1Yey mice. SOCE is also reduced in iPSCs from an individual with DS. We

The Role of HINT3 in Myocardial Ischemia-Reperfusion Injury in Male Mice: Mechanisms Involving SDHA and its Acetylation.

Myocardial ischemia-reperfusion (I/R) injury is characterized by oxidative stress, mitochondrial dysfunction, and cardiomyocyte apoptosis. During I/R, the accumulation and oxidation of succinate contribute to reactive oxygen species (ROS) production, worsening tissue damage. Histidine triad nucleotide-binding protein 3 (HINT3) is identified as a regulator of mitochondrial function and cardiomyocyte survival during I/R. In a mouse I/R model and an oxygen-glucose deprivation/reoxygenation (OGD/R) model, it shows that HINT3 expression is downregulated after I/R. Cardiomyocyte-specific knockout of

Metabolic Reprogramming in Stromal and Immune Cells in Rheumatoid Arthritis and Osteoarthritis: Therapeutic Possibilities.

Metabolic reprogramming of stromal cells, including fibroblast-like synoviocytes (FLS) and chondrocytes, as well as osteoclasts (OCs), are involved in the inflammatory and degenerative processes underlying rheumatoid arthritis (RA) and osteoarthritis (OA). In RA, FLS exhibit mTOR activation, enhanced glycolysis and reduced oxidative phosphorylation, fuelling inflammation, angiogenesis, and cartilage degradation. In OA, chondrocytes undergo metabolic rewiring, characterised by mTOR and NF-κB activation, mitochondrial dysfunction, and increased glycolysis, which promotes matrix metalloproteinase

Drp1 controls complex II assembly and skeletal muscle metabolism by Sdhaf2 action on mitochondria.

The dynamin-related guanosine triphosphatase, Drp1 (encoded by Dnm1l), plays a central role in mitochondrial fission and is requisite for numerous cellular processes; however, its role in muscle metabolism remains unclear. Here, we show that, among human tissues, the highest number of gene correlations with DNM1L is in skeletal muscle. Knockdown of Drp1 (Drp1-KD) promoted mitochondrial hyperfusion in the muscle of male mice. Reduced fatty acid oxidation and impaired insulin action along with increased muscle succinate was observed in Drp1-KD muscle. Muscle Drp1-KD reduced complex II assembly a

Case report: Bilateral carotid body tumors with a concomitant skull-base paraganglioma.

BACKGROUND: Bilateral carotid body tumors with a concomitant skull-base paraganglioma are extremely rare, of which only one case has been reported in the literature to date. CASE PRESENTATION: We present the case of a 35-year-old male with 1 year of hypertension and high levels of dopamine and 3-methoxytyramine. Magnetic resonance imaging (MRI) scans demonstrated three separate masses at the left middle cranial fossa floor and bilateral carotid bifurcation. Genetic testing showed succinate dehydrogenase complex subunit D mutation. The patient underwent the resection of the left skull base mas

Dysregulation of Glucagon Secretion by Hyperglycemia-Induced Sodium-Dependent Reduction of ATP Production.

Diabetes is a bihormonal disorder resulting from combined insulin and glucagon secretion defects. Mice lacking fumarase (Fh1) in their β cells (Fh1βKO mice) develop progressive hyperglycemia and dysregulated glucagon secretion similar to that seen in diabetic patients (too much at high glucose and too little at low glucose). The glucagon secretion defects are corrected by low concentrations of tolbutamide and prevented by the sodium-glucose transport (SGLT) inhibitor phlorizin. These data link hyperglycemia, intracellular Na+ accumulation, and acidification to impaired mitochondrial metabolism

Effect of sulfonamides as carbonic anhydrase VA and VB inhibitors on mitochondrial metabolic energy conversion.

Obesity is quickly becoming an increasing problem in the developed world. One of the major fundamental causes of obesity and diabetes is mitochondria dysfunction due to faulty metabolic pathways which alter the metabolic substrate flux resulting in the development of these diseases. This paper examines the role of mitochondrial carbonic anhydrase (CA) isozymes in the metabolism of pyruvate, acetate, and succinate when specific isozyme inhibitors are present. Using a sensitive electrochemical approach of wired mitochondria to analytically measure metabolic energy conversion, we determine the re
