Missense Mutations Modify the Conformational Ensemble of the α-Synuclein Monomer Which Exhibits a Two-Phase Characteristic.

α-Synuclein is an intrinsically disordered protein occurring in different conformations and prone to aggregate in β-sheet structures, which are the hallmark of the Parkinson disease. Missense mutations are associated with familial forms of this neuropathy. How these single amino-acid substitutions modify the conformations of wild-type α-synuclein is unclear. Here, using coarse-grained molecular dynamics simulations, we sampled the conformational space of the wild type and mutants (A30P, A53P, and E46K) of α-synuclein monomers for an effective time scale of 29.7 ms. To characterize the structur

A Narrative Review on Biochemical Markers and Emerging Treatments in Prodromal Synucleinopathies.

Alpha-synuclein has been associated with neurodegeneration, especially in Parkinson's disease (PD). This study aimed to review clinical, biochemical, and neuroimaging markers and management of prodromal synucleinopathies. The prodromal state of synucleinopathies can be better understood with PD pathophysiology, and it can be separated into premotor and pre-diagnostic phases. The incidence of PD in patients with prodromal phase symptoms ranges from 0.07 to 14.30, and the most frequently studied pathology is the REM behavioral disorder (RBD). Neuroimaging markers are related to dopamine denervat…

Multiple system atrophy: Diagnostic challenges and a proposed diagnostic algorithm.

Multiple system atrophy (MSA) is a heterogenous condition, presenting with core clinical features of autonomic dysfunction, parkinsonism, and/or cerebellar ataxia. The presence of alpha-synuclein glial cytoplasmic inclusion is the hallmark of MSA. It shares a common pathological origin with Parkinson's disease (PD) and Lewy body dementia (DLB) and they are collectively grouped as "synucleinopathies." The pathological synuclein protein is now well- recognized in skin biopsies of these patients. Besides the pathological findings, radiological investigation is a useful diagnostic tool. Brain MRI …

Lack of synucleins induces an alteration of lipid peroxidation in the brain.

Family proteins - α-, β, γ-synucleins shown to play important roles in metabolism, signal transduction and dopamine handling. Aggregated α-synuclein is neurotoxic and involved in pathogenesis of Parkinson's disease. The mechanism toxicity of aggregated α-synuclein includes lipid peroxidation, oxidative stress and ferroptosis but effect of monomeric synucleins on the basal lipid peroxidation is unclear. Using acute brain slices and primary cortical co-culture of neurons and astrocytes from α-, β- and γ-synuclein deficient mice and live cell imaging we studied how lack of synucleins changes the

SNCA-targeted transcriptional repression therapy alleviates pathological and behavioral perturbations in a Parkinson's disease mouse model.

Alpha-synuclein (SNCA) overexpression is implicated in Parkinson's disease (PD) pathogenesis, making SNCA downregulation a promising therapeutic strategy. We developed a SNCA-targeted transcriptional repression therapy using an all-in-one lentiviral vector (LV) carrying deactivated CRISPR/(d)Cas9, gRNA targeted at SNCA-intron1, and either the catalytic domain of DNA-methyltransferase3A (DNMT3A), or an engineered repressor molecule, a fusion of MeCP2's transcription repression domain (TRD) and KRAB. Therapeutic efficacy was evaluated following co-administration of the therapeutic and model vect…

INTRODUCTION: Lewy Body Dementia (LBD) is a Parkinsonian disorder which often leads to Lower Urinary Tract Symptoms (LUTS), especially an Ov … d of constipation, with 44% suffering from fecal incontinence. DISCUSSION: LBD is characterized by alpha-synuclein aggregates in the cerebral cortex, thus explaining associated cognitive impairment. The most commonly found LUTS is stress incontinence. We also found voiding difficulties in smaller proportion, sometimes associated with prostatism. Topographically, …

Modulators selectively reshape α-synuclein phase transitions.

Protein phase transitions govern numerous diseases, including neurodegenerative disorders. In Parkinson's disease, distinct species of the protein α-synuclein undergo phase transitions from disordered to ordered β-rich states. The emerging species and transitions between them can be reshaped by modulators, e.g., small molecules, peptides or antibodies. Here, we use coarse-grained simulations to understand the effect of modulators on the thermodynamics and kinetics of α-synuclein phase transitions. Each protein is represented as a morphing particle that transforms from a soft sphere (disordered

Nanoscale interrogation of α-Synuclein oligomers: Bridging biophysics, interface science, and neurobiology.

α-Synuclein (α-syn) is an intrinsically disordered presynaptic protein. In synucleinopathies, it undergoes a structural transition into β-sheet-rich conformers that promote the formation of amyloid fibrils and pathological inclusions. Although fibrillar aggregates have been studied extensively, soluble oligomers, which may have the greatest neurotoxic potential, remain poorly understood because of their transient nature and structural heterogeneity. This review critically examines α-syn oligomeric species, with particular emphasis on advanced biophysical and surface characterization techniques

In situ NMR and integrative proteomics reveal the interaction signature of serum α-synuclein.

Alpha-synuclein (αS) plays a central role in several neurodegenerative diseases. Although predominantly neuronal, αS is also present in peripheral fluids such as serum and cerebrospinal fluid (CSF), where it may contribute to disease propagation. However, its extracellular interactions remain poorly understood. By combining in situ NMR spectroscopy and bottom-up MS proteomics, we reveal that monomeric αS exhibits distinct interactomes in serum versus CSF. Both N- and C-terminal αS regions bind to serum components, while in CSF, αS remains largely unbound. The N- and C-terminal interactions are…

Characterization of proteoforms within α-synuclein complexes in living cells by cleavable cross-linking combined with top-down mass spectrometry.

α-Synuclein (α-syn) exists as diverse proteoforms that interact with distinct partners to perform various biological functions. Therefore, top-down mass spectrometry (MS) analysis of proteoform components within α-syn complexes is crucial for understanding their functions. However, the purification of α-syn complexes directly from cell or tissue lysates leads to the loss of weak or transient interacting partners present in the native cellular environment. In vivo cross-linking could fix native interactions before cell lysis, but the top-down MS identification of covalently cross-linked protein

Structural dynamics of α-Synuclein: Multi-scale imaging insights into pathological progression across Synucleinopathies.

The misfolding and aberrant aggregation of alpha-synuclein (α-syn) constitute the central pathological hallmark of a spectrum of synucleinopathies, including Parkinson's disease, dementia with Lewy bodies, and multiple system atrophy. A continuous ultrastructural conformational evolution from disordered monomers through toxic oligomers to amyloid fibrils is linked to the formation of Lewy pathology and the progressive functional decline of neurons. This review integrates structural dynamics revealed by multi-scale electron microscopy (EM) techniques-including transmission electron microscopy, …

Mass spectrometric identification of novel truncated α-synuclein species following optimized immunoprecipitation from human brain tissue.

α-synuclein is a protein central to neurodegenerative diseases, and its functions are affected by multiple posttranslational modifications. Mass spectrometry is powerful for the characterization of α-synuclein forms but requires prior efficient immunoprecipitation conditions. In this study, we refined the immunoprecipitation of α-synuclein from human brain tissues by evaluating key parameters that influence recovery and specificity. We assessed the performance of tosyl-activated magnetic beads versus sheep antibody beads, identifying the optimal bead type for enhanced binding efficiency. Vario
