Advances in Optical Tools to Study Taste Sensation.

Taste sensation is the process of converting chemical identities in food into a neural code of the brain. Taste information is initially formed in the taste buds on the tongue, travels through the afferent gustatory nerves to the sensory ganglion neurons, and finally reaches the multiple taste centers of the brain. In the taste field, optical tools to observe cellular-level functions play a pivotal role in understanding how taste information is processed along a pathway. In this review, we introduce recent advances in the optical tools used to study the taste transduction pathways.

Regulation of peripheral inflammation by the central nervous system.

In inflammatory disorders such as rheumatoid arthritis, cytokines and danger signals are sensed by the central nervous system, which adapts behavior and physiologic responses during systemic stress. The central nervous system can also signal the periphery to modulate inflammation through efferent hormonal and neuronal pathways. The brain and spinal cord are involved in this bidirectional interaction. A variety of neuronal pathways that modulate synovial inflammation have been implicated, including the sympathetic and the parasympathetic branches of the autonomic system. Another mechanism, the

Catecholamines and acetylcholine are key regulators of the interaction between microbes and the immune system.

Recent studies suggest that catecholamines (CAs) and acetylcholine (ACh) play essential roles in the crosstalk between microbes and the immune system. Host cholinergic afferent fibers sense pathogen-associated molecular patterns and trigger efferent cholinergic and catecholaminergic pathways that alter immune cell proliferation, differentiation, and cytokine production. On the other hand, microbes have the ability to produce and degrade ACh and also regulate autogenous functions in response to CAs. Understanding the role played by these neurotransmitters in host-microbe interactions may provid

Chronic cough in the Holmes-Adie syndrome: association in five cases with autonomic dysfunction.

The Holmes-Adie syndrome consists of unilateral or bilateral tonic pupils with near light dissociation and tendon areflexia. It is associated with autonomic disturbances affecting sudomotor and vasomotor function. Five such patients are reported on who also had a troublesome chronic dry cough, which was of unknown aetiology and was resistant to a range of treatments. The cough may be related to involvement of afferent or efferent pathways in the vagus. Chronic cough may be an accompaniment in the Holmes-Adie syndrome, like other forms of autonomic dysfunction.

Hypothalamic Neuromodulation of Hypothermia in Domestic Animals.

When an organism detects decreases in their core body temperature, the hypothalamus, the main thermoregulatory center, triggers compensatory responses. These responses include vasomotor changes to prevent heat loss and physiological mechanisms (e.g., shivering and non-shivering thermogenesis) for heat production. Both types of changes require the participation of peripheral thermoreceptors, afferent signaling to the spinal cord and hypothalamus, and efferent pathways to motor and/or sympathetic neurons. The present review aims to analyze the scientific evidence of the hypothalamic control of h

Bidirectional bionic limbs: a perspective bridging technology and physiology.

Precise control of bionic limbs relies on robust decoding of motor commands from nerves or muscles signals and sensory feedback from artificial limbs to the nervous system by interfacing the afferent nerve pathways. Implantable devices for bidirectional communication with bionic limbs have been developed in parallel with research on physiological alterations caused by an amputation. In this perspective article, we question whether increasing our effort on bridging these technologies with a deeper understanding of amputation pathophysiology and human motor control may help to overcome pressing

Interactions between the sympathetic nervous system and angiotensin system in renovascular hypertension.

Renal artery stenosis especially that due to atherosclerotic peripheral vascular disease is increasing in the population. The role of the renin angiotensin aldosterone system in the pathogenesis of renovascular hypertension is widely accepted. There is increasing recognition of the role of the sympathetic nervous system, from both afferent and efferent signaling pathways, as a contributor to development of hypertension. In this review we briefly summarize the experimental evidence for the respective roles of the renin angiotensin aldosterone system and the sympathetic nervous system as well as

Recent concepts in the pathophysiology and evaluation of spasticity.

Spasticity is still a challenge to clinicians and scientists. The participation of Ia, Ib, II, III and IV afferents pathways, as well as those related with presynaptic inhibition. Renshaw cells modulation, and alpha and gamma motoneurons hyperexcitability, in this medical problem are reviewed. A brief overview on the more common neurophysiological tests performed during the evaluation of human spasticity is presented.

A computational model of the mammalian auditory periphery with a closed-loop medial olivocochlear reflex simulating across-channel efferent gain control.

The afferent (ascending) auditory system and how its specialized mechanisms and circuits support ecologically relevant auditory computations such as speech recognition have received considerable attention in decades past. This work has culminated in accurate computational models of early afferent coding alongside a good understanding of how low-level mechanisms (e.g., peripheral tuning) impact auditory perception. In contrast, the auditory efferent (descending) system and its role in auditory perception are much less well understood. To address this gap in knowledge, we describe modifications

Gut-brain communication and obesity: understanding functions of the vagus nerve.

Given the crucial role of the gastrointestinal tract and associated organs in handling nutrient assimilation and metabolism, it has long been known that its communication with the brain is important for the control of ingestive behavior and body weight regulation. It is also clear that gut-brain communication is bidirectional and utilizes both rapid neural and slower humoral mechanisms and pathways. However, progress in understanding these mechanisms and leveraging them for the treatment of obesity and metabolic disease has been hindered by the enormous dimension of the gut mucosa, the complex

Neural mechanism of bradycardiac responses elicited by acupuncture-like stimulation to a hind limb in anesthetized rats.

The effects of acupuncture-like stimulation of a hind limb on heart rate were examined in anesthetized rats. An acupuncture needle, having a diameter of either 160 or 340 microm, was inserted into the skin and underlying muscles at a depth of about 5 mm and twisted right and left twice every second for 1 min. Stimulation by a needle with a diameter of either 160 or 340 microm produced a decrease in heart rate. Severance of the femoral and sciatic nerves ipsilateral to the hind-limb stimulation completely abolished the bradycardiac response. Also, heart rate was significantly decreased by acupu

Differential c-fos expression in the nucleus of the solitary tract and spinal cord following noxious gastric distention in the rat.

c-Fos has been used as a marker for activity in the spinal cord following noxious somatic or visceral stimulation. Although the viscera receive dual afferent innervation, distention of hollow organs (i.e. esophagus, stomach, descending colon and rectum) induces significantly more c-Fos in second order neurons in the nucleus of the solitary tract and lumbosacral spinal cord, which receive parasympathetic afferent input (vagus, pelvic nerves), than the thoracolumbar spinal cord, which receives sympathetic afferent input (splanchnic nerves). The purpose of this study was to determine the contribu

Functional variability of sacral roots in bladder control.

OBJECT: Sacral roots are involved in sensory, autonomic, and motor innervation of the lower limbs and perineum. Theoretically, it can be assumed that the S-3 root level innervates the bladder; however, clinical practice shows that this distribution can vary. Few researchers have studied this variability. METHODS: The authors conducted a retrospective study involving 40 patients who underwent surgery requiring an electrophysiological exploration of the sacral roots. They performed stimulations for the monitoring of muscular (3 Hz, 1 V) and bladder responses under cystomanometry (30 Hz, 10 V).

Transplantation of Xenopus laevis ears reveals the ability to form afferent and efferent connections with the spinal cord.

Previous comparative and developmental studies have suggested that the cholinergic inner ear efferent system derives from developmentally redirected facial branchial motor neurons that innervate the vertebrate ear hair cells instead of striated muscle fibers. Transplantation of Xenopus laevis ears into the path of spinal motor neuron axons could show whether spinal motor neurons could reroute to innervate the hair cells as efferent fibers. Such transplantations could also reveal whether ear development could occur in a novel location including afferent and efferent connections with the spinal

Diffusion tensor imaging of brainstem tumors: axonal degeneration of motor and sensory tracts.

OBJECT: Diffusion tensor (DT) imaging has been used to predict postoperative motor function in patients with supratentorial tumors. The authors sought to determine whether DT imaging and white matter tractography could detect axonal degeneration in patients with brainstem tumors. METHODS: A cross-sectional, retrospective study of 7 patients with brainstem tumors and 8 healthy volunteers was performed. The DT imaging data were normalized and regions of interest (ROIs) with the highest probability of sensory and motor connections were selected using the Talairach Atlas to identify the 3D millim

A distinct anatomical network of cortical areas for analysis of motion in far peripheral vision.

We defined cortical areas involved in the analysis of motion in the far peripheral visual field, a poorly understood aspect of visual processing in primates. This was accomplished by small tracer injections within and around the representations of the monocular field of vision ('temporal crescents') in the middle temporal area (MT) of marmoset monkeys. Quantitative analyses demonstrate that the representation of the far periphery receives specific connections from the retrosplenial cortex (areas 23v and prostriata), as well as comparatively stronger inputs from the primary visual area (V1) and

Facial nerve injury induces facilitation of responses in both trigeminal and facial nuclei of rat.

A study was made of the effects of facial nerve transection on trigeminal stimulation- evoked field potentials in the principal trigeminal (Pr5) and facial nuclei (7) in rats. Although the transected branch of the facial nerve contains pure motoric efferents, it resulted in enhanced responses in both Pr5 and 7. These electrophysiological results suggest a functional circuitry involving the whiskers, trigeminal nerve, Pr5 and 7 and the facial nerve as efferent. The disconnection (opening) of this loop results in enhanced responsiveness of the neurons in both Pr5 and 7.

Receptive field scatter, topography and map variability in different layers of the hindpaw representation of rat somatosensory cortex.

We recorded neurons extracellularly in layers II/III, IV, and V of the hindpaw representation of primary somatosensory cortex in anesthetized rats and studied laminar features of receptive fields (RFs) and representational maps. On average, RFs were smallest in layer IV and largest in layer V; however, for individual penetrations we found substantial deviations from this rule. Within the hindpaw representation, a distinct rostrocaudal gradient of RF size was present in all layers. While layer V RFs were generally largest independent of this gradient, layer IV RFs recorded caudally representing

Arm-trunk coordination in the absence of proprioception.

During trunk-assisted reaching to targets placed within arm's length, the influence of trunk motion on the hand trajectory is compensated for by changes in the arm configuration. The role of proprioception in this compensation was investigated by analyzing the movements of 2 deafferented and 12 healthy subjects. Subjects reached to remembered targets (placed approximately 80 degrees ipsilateral or approximately 45 degrees contralateral to the sagittal midline) with an active forward movement of the trunk produced by hip flexion. In 40% of randomly selected trials, trunk motion was mechanically

Selective potentiating effect of RS14203 on a serotoninergic pathway in anesthetized rats.

The usefulness of selective inhibitors of type 4 phosphodiesterase (PDE4) in the treatment of inflammation and pulmonary diseases is limited by their side effects: nausea and vomiting. We studied the effect of three structurally diverse PDE4 inhibitors on the vagal nerve afferent and efferent fibers in anesthetized rats. The effects of RS14203, (R)-rolipram, and CT-2450 were evaluated on the von Bezold-Jarisch reflex (vagal afferent fibers) and in a model of vagal electrical stimulation (vagal efferent fibers). All three PDE4 inhibitors were administered at 1, 10, or 100 microg/kg (iv) 15 min
