BACKGROUND: Dogs play an important role in the transmission of zoonotic parasites to humans. Among these, gastrointestinal (GI) helminths are widely prevalent and pose a significant public health risk. This study investigated the prevalence, molecular characterization and risk factors analyses of gastrointestinal nematodes in pet and semi-pet dogs in Haryana, India, to assess their potential zoonotic relevance.
RESULTS: A total of 310 faecal samples were collected from pet and semi-pet dogs from five randomly selected blocks of Hisar district of Haryana, India and examined for the presence of

Differentiation between viable and non-viable hookworm ova in environmental samples is necessary in order to implement strategies to mitigate re-infections in endemic regions. In this study, an untargeted metabolic profiling method was developed that utilised gas chromatography-mass spectrometry (GC-MS) in order to investigate hookworm ova viability. Ancylostoma caninum was used to investigate the metabolites within viable and non-viable ova. Univariate and multivariate statistical analyses of the data resulted in the identification of 53 significant metabolites across all hookworm ova samples

Bile acid-like molecules named dafachronic acids (DAs) control the dauer formation program in Caenorhabditis elegans through the nuclear receptor DAF-12. This mechanism is conserved in parasitic nematodes to regulate their dauer-like infective larval stage, and as such, the DAF-12 ligand binding domain has been identified as an important therapeutic target in human parasitic hookworm species that infect more than 600 million people worldwide. Here, we report two x-ray crystal structures of the hookworm Ancylostoma ceylanicum DAF-12 ligand binding domain in complex with DA and cholestenoic acid

Hookworm infection is associated with anaemia and malnutrition in many resource-limited countries. Ancylostoma hookworms have previously been shown to modulate host cellular immune responses through multiple mechanisms, including reduced mitogen-mediated lymphocyte proliferation, impaired antigen presentation/processing, and relative reductions in CD4(+) T cells in the spleen and mesenteric lymph nodes. Syrian hamsters were depleted of CD4(+) for up to 9 days following intraperitoneal injection (200 microg) of a murine anti-mouse CD4 monoclonal IgG (clone GK1.5). CD4(+) T-cell-depleted hamster

This study compared five methods for detecting the eggs of the human parasitic geohelminths Ascaris, Trichuris, and hookworm in infant stool, and describes the epidemiology of infection in infants from a parasite-endemic area. A total of 424 infants 5-11 months old were enrolled from three villages on Pemba Island, Zanzibar. Methods used included the Kato-Katz technique, formol ethyl acetate sedimentation, modified formol ethyl acetate sedimentation, modified Wisconsin floatation, and simple gravity sedimentation. Of methods used alone, Wisconsin floatation and simple gravity sedimentation eac

In the mid nineteenth century, laboratory research in embryology, development, ecology and pathogeny of helminths unexpectedly resulted in a change in the explication of the etiology of some human pathology that was traditionally attributed to environmental factors. The new postulates of the parasitology of helminths put in question the traditional explications as well as the recognized authorities in the fields of clinical medicine, hygiene, and of medical geography, affecting not only the traditional circles where the innovations were discussed, but also the institutions that controlled and

Hookworms are a leading cause of anemia in developing countries, and a strategy aimed at reducing pathology caused by blood-feeding adult parasites would be a valuable addition to global control efforts. This article describes experiments designed to induce resistance to the major clinical sequelae (weight loss and anemia) of Ancylostoma ceylanicum hookworm infection in Syrian golden hamsters of the outbred LVG strain. Previously infected animals acquired long-lived resistance to weight loss and anemia caused by a secondary hookworm infection. Furthermore, transfer of pooled serum from twice-i

Four experiments are described in which hamsters, initially exposed to primary infection with Ancylostoma ceylanicum, were given a homologous challenge and components of the secondary response were quantified and compared to relevant control groups. The initial establishment of the L3 larvae was not prevented in immunized hamsters but their growth was slowed and virtually all larvae were lost within a week of challenge, when the majority were still at the L4 stage of development. The loss of worms was associated with an accelerated mucosal mastocytosis and increased systemic antibody. Thus acq

Human eosinophilic enteritis (EE) may result from hypersensitivity to the excretory/secretory (ES) antigens of adult Ancylostoma caninum. The origin of several antigens were identified by probing adult A. caninum with mouse monoclonal antibodies (MoAbs), sera from mice vaccinated with ES antigens and sera from human EE patients. Six MoAbs (AC/ES 1-6) were produced against ES antigens, two being IgG3 and four IgM. Western blots demonstrated four different antigen specificities: MoAb AC/ES 1 bound strongly to an ES product at about 30 kDa; AC/ES 2 recognized a broad band ranging from 50-200 kDa;

Twenty-six mixed-breed (14 males, 12 females) dogs were used in a double-blind study to evaluate the effect of milbemycin oxime against naturally acquired infection with Ancylostoma caninum. Dogs were ranked and paired, on the basis of number of hookworm eggs/g of feces, and treatment was randomly assigned. Each dog was given either the study drug or placebo (1 tablet/11.4 kg [0.5 mg/kg] of body weight). Eggs per gram of feces enumeration was done on days 3 and 7 after treatment, and dogs were euthanatized on day 7. On day 3, 5 of the 13 dogs in the milbemycin-treated group had hookworm eggs i

Ancylostoma ceylanicum infection in golden hamsters (Mesocricetus auratus) caused marked biochemical and histopathological derangements. Jejunum, the primary site of infection, showed pronounced alterations compared with liver. Though the biochemical composition of jejunum was not significantly altered, activities of a few lysosomal enzymes were enhanced during hookworm infection. Marked damage to mitochondrial and microsomal membranes was reflected in changes in the activities of the marker enzymes from jejunal tissue. Lipid content, especially phospholipids and neutral lipids of hepatic tiss

A model of human hookworm infection has been developed which shows that dogs currently infected with small numbers of hookworms are considerably resistant to a challenge infection with a large inoculum of infective larvae. Two groups of dogs were infected with 500 larvae and four weeks after infection one group together with a previously uninfected control group were infected with 5,000 larvae and followed for six weeks. When compared with the secondary infection control dogs, faecal egg excretion and adult worm burdens were reduced by an average of 83 and 78% respectively. Infections had no s

Six parasitic helminths detected were Ancylostoma sp., Opisthorchis felineus, Toxocara mystrax, Hepaticola hepatica, Taenia sp. and Hydatigera taeniaeformis. The prevalence of H. hepatica, T. mystrax and H. taeniaeformis is associated with rodents hunted by cats, and that of O. felineus with fish fed to cats. The role of cats as primary host and reservoir for these helminths, which could be of zoonotic importance, is briefly mentioned.

Albendazole was given to some men for the first time. 131 patients (43 children) received different dosages, without clinical intolerance. Biological parameters were normal. Each study was made in two parts: - determination of the good dosage, - administration in a double-blind study (except for Strongyloides Stercoralis). Albendazole has a good efficiency against Enterobius Vermicularis, Trichuris trichiura, Ascaris lumbricoides and Necator americanus. Strongyloides Stercoralis seems to be killed by this drug, but complementary studies will be necessary.

The dominant intestinal helminths in the Iranian villages of Kargar Malleh (hookworms), Bandar Abbas (hookworms) and Jazin (Ascaris lumbricoides and Trichuris trichiura), were recorded with their frequency distributions, in 80, 40 and 111 villagers, respectively in 1977-78. Anthelminthics were then used to remove all intestinal stages which were identified and counted. Analyses showed clumped or over-dispersed patterns in all cases. Each community contained 'wormy persons', 1 to 3% of the individuals carried 11, 16, 30, 38 or 84% of all the worms collected. 'Wormy persons' could only be identi

Serum protein of Swiss albino mice infected with Ancylostoma caninum larvae, were analysed electrophoretically. Twenty mice were infected with a dose of 1000 larvae and the alterations in serum protein during an infection period of 30 days were recorded and compared with uninfected controls. The result showed a significant decrease in albumin and gamma globulin with an increase in beta globulin. There were no significant changes in the alpha-1 and alpha-2 globulins. The total globulins increased and A/G ratio decreased. These changes appeared to be remarkable on the 9th day after infection.
