Bacillus subtilis ATCC 6051 and ATCC 6633 spores used in bacterial inhibition screening assays for genetic metabolism defects in newborn infants were produced by using liquid synthetic replacement sporulation media. These media allowed a high degree of sporulation, as judged by direct cell counts. Sporulation took place within 23 to 27 h with these media. Also, a more rational procedure for selecting the most sensitive clones of these organisms to the various inhibitors used in the microbiological screening assays is presented.

Mechanisms responsible for regulation of tear film mucus are poorly understood. Humoral factors responsible for stimulation of mucus secretion can be studied in vitro by using the free-swimming urn cell, a normal component of the coelomic fluid of the marine invertebrate Sipunculus nudus. An ointment prepared from fat (sheabutter) obtainable from an indigenous (African) fruit released medicaments faster than either of the two ointment bases recommended by the British Pharmacopoeia. Reduction in particle size of crystalline medicaments incorporated in ointments increased surface area and transfer from the ointment to the substrate, possibly due to greater dissolution in the base, and, therefore, increased the availability of medicament at the ointment-substrate interface.

Two biological assays were conducted in which the antirachitic activity in chicks of 5,6-trans-vitamin D3 added to feed is compared with that of 5,6-cis-vitamin D3. The first commercial kit for the radioisotopic assay of vitamin B(12) in serum-the Phadebas B(12) Test produced higher values than the radioisotopic method of Raven, Robson, Walker, and Barkham (1969) and the Lactobacillus leichmannii microbiological assay. Its normal range was 300-1100 pg/ml and its reproducibility was similar to that of the other radioisotopic method. It should be possible to lower the results obtained by the Phadebas method by modifying its standard curve and to reduce the time taken for the assay by shortening its incubation period.

Because of wide variations in the methods used to assay the potency of pertussis vaccine, the effect of the type of challenge used in the mouse-protection test was studied. A small proportion of patients with peptic ulcer disease harbor a hormone-secreting pancreatic tumor as the cause of the ulcer diathesis, and if this is not recognized, they may suffer undue morbidity and mortality. Close attention to the patient's history, roentgenographic findings and gastric secretory studies will allow identification of ulcerogenic tumors of the pancreas. Other clues to the diagnosis include associated endocrine abnormalities and diarrhea.
