Metabolomic Study of Hibernating Syrian Hamster Brains: In Search of Neuroprotective Agents.

Syrian hamsters undergo a reversible hyperphosphorylation of protein τ during hibernation, providing a unique natural model that may unveil … herefore, play a key role in the regulation of hibernation processes. The increase in the levels of ceramides containing more than 20 C atoms was found in torpor animals, reflecting a higher activity of CerS2 during hibernation, linked to neurofibrillary tangle generation and structural changes in the Golgi apparatus. Our results open up the debate about th …

Structure elucidation of sphingolipids from the mycopathogen Paracoccidioides brasiliensis: an immunodominant beta-galactofuranose residue is carried by a novel glycosylinositol phosphorylceramide antigen.

Two major acidic glycolipid components (Pb-1 and Pb-2) have been extracted from the mycopathogen Paracoccidioides brasiliensis, a thermally … e analysis, and exoglycosidase digestion. The compounds were found to be glycosylinositol phosphorylceramides (GIPCs) with the following structures: Pb-2, Manpalpha1-->3Manpalpha1-->2Ins1-P-1Cer; and Pb-1, Manpalpha1-->3[Galfbeta1-->6]Manpalpha1-->2Ins1- P-1Cer. The serologically nonreactive Pb-2 appears to be a biosynthetic intermediate between mannosylino …

Untargeted salivary metabolomics for early prediction of chemotherapy-induced oral toxicities in patients with oral squamous cell carcinoma.

Chemotherapy-induced oral toxicities, including oral mucositis, xerostomia, and dysgeusia, are common complications in patients with oral squamous cell carcinoma (OSCC), often leading to impaired quality of life, treatment interruptions, and poor therapeutic outcomes. Reliable non-invasive biomarkers capable of predicting these toxicities before severe clinical manifestations remain limited. This study investigated salivary metabolomic alterations associated with chemotherapy-induced oral toxicities using untargeted liquid chromatography-quadrupole time-of-flight mass spectrometry (LC-QTOF-MS)

Protective role of the ELOVL2/docosahexaenoic acid axis in glucolipotoxicity-induced apoptosis in rodent beta cells and human islets.

AIMS/HYPOTHESIS: Dietary n-3 polyunsaturated fatty acids, especially docosahexaenoic acid (DHA), are known to influence glucose homeostasis. We recently showed that Elovl2 expression in beta cells, which regulates synthesis of endogenous DHA, was associated with glucose tolerance and played a key role in insulin secretion. The present study aimed to examine the role of the very long chain fatty acid elongase 2 (ELOVL2)/DHA axis on the adverse effects of palmitate with high glucose, a condition defined as glucolipotoxicity, on beta cells. METHODS: We detected ELOVL2 in INS-1 beta cells and mou

Anidulafungin-Induced Suicidal Erythrocyte Death.

BACKGROUND/AIMS: The novel antifungal drug Anidulafungin is used for the treatment of diverse fungal infections including candidiasis and aspergillosis. The traditional antifungal drug amphotericin B has previously been shown to trigger eryptosis, the suicidal death of erythrocytes characterized by cell shrinkage and cell membrane scrambling with phosphatidylserine translocation to the erythrocyte surface. Triggers of eryptosis include increase of cytosolic Ca2+ activity ([Ca2+]i), oxidative stress, ceramide, activated protein kinase C (PKC), casein kinase 1α or p38 kinase and activated caspas

From Obesity to Muscle Insulin Resistance: The Mediating Roles of Intramyocellular Lipids, Inflammation, and Oxidative Stress.

Obesity is highly correlated to muscle insulin resistance (IR), which significantly impacts metabolic health. One of the primary mechanisms connecting obesity to muscle IR is the accumulation of intramyocellular lipids (IMCL). Excessive lipid accumulation in muscle cells, that is, muscle lipotoxicity, leads to the formation of lipid metabolites, such as diacylglycerol (DAG) and ceramides, which disrupt insulin signalling pathways. These metabolites activate protein kinase C (PKC) and other kinases that inhibit insulin receptor substrate (IRS) proteins, subsequently impairing the insulin signal…

Coronary artery bypass graft in a patient with Fabry's disease.

Fabry's disease is a lysosomal storage disease characterized by intracellular accumulation of ceramide trihexoside resulting from alpha-galactosidase A deficiency. While the heart is often involved, coronary artery disease and its management in Fabry's disease patients are extremely rare clinical entities. We report a case of a 72-year-old man with left main disease in Fabry's disease with special consideration of the arterial wall pathology.

Acid sphingomyelinase inhibition restores RPE homeostasis and photoreceptor function in preclinical Stargardt macular degeneration models.

Stargardt disease, which destroys central high-resolution vision in over 2 million people globally, lacks effective therapies. The primary site of damage in Stargardt disease is the retinal pigment epithelium (RPE), which safeguards photoreceptor health and function. Progressive loss of RPE integrity precedes visual deficits, yet insight into mechanisms driving RPE dysfunction and how this influences disease pathogenesis remains elusive. Here, we addressed this in cell-based and pigmented Abca4-/- Stargardt mice models using super-resolution imaging, bioinformatics, and biochemical approaches.

Fungal extracellular vesicles reshape Lactobacillus rhamnosus vesicles to enhance lysosomal homeostasis and resolve intestinal inflammation.

Extracellular vesicles (EVs) are emerging mediators of intercellular and interkingdom communication. While bacterial EVs directly modulate host health, whether exogenous vesicles can reprogram gut microbial vesiculation and bioactivity remains unclear. Using a defined in vitro conditioning system, ScEV exposure altered EV production by Lactobacillus rhamnosus, generating ScEV-conditioned L. rhamnosus vesicles (ScEV-LrEVs). Comparative proteomics and lipidomics revealed extensive remodeling of vesicle cargo, including ~60-fold enrichment of hydroxylated ceramide Cer(t18:0/24:0(2OH)) in ScEV-LrE

Proteomic Profiling of an Exosome-Enriched Extracellular Vesicle Fraction and Structural Characterization of SMPDL3A in the Carcinogenic Liver Fluke Clonorchis sinensis.

Exosomes are important mediators of host-parasite communication and contain diverse molecules that may support the survival of Clonorchis sinensis in the biliary tract. To explore their biochemical properties, exosomes isolated from excretory-secretory products of Korean C. sinensis isolates were characterized through integrated morphological, proteomic, and gene ontology analyses. The vesicles exhibited typical exosomal size ranges and marker profiles, and their protein components were enriched for cytoskeletal, metabolic, and vesicle-trafficking components relevant to epithelial signaling an

Long-term exposure to triclocarban induces splenic injuries in mice: Insights from spatial metabolomics and lipidomics.

Triclocarban (TCC) is a widely used antimicrobial agent and known endocrine-disrupting chemical found in various products. While its potential toxicities on endocrine-related organs have been highlighted in previous studies, the effects of TCC on non-endocrine organs, particularly the spleen, remain largely unknown. Here, we employed a novel approach combining long-term TCC exposure in a mouse model with spatial metabolomics and lipidomics to investigate the effects of TCC on the spleen. Our results showed that TCC exposure significantly altered the splenic organ weight and coefficient and ind

PPARα exacerbates Salmonella Typhimurium infection by modulating the immunometabolism and macrophage polarization.

Salmonella enterica serovar Typhimurium (STm) is a causative pathogen for robust inflammatory gastrointestinal disease and can lead to systemic infection. Eicosanoids, bioactive lipid mediators, play a crucial role in modulating both the induction and resolution of inflammatory responses during an infection. A subset of eicosanoids activates PPARs, nuclear receptor/transcription factors that regulate fatty acid metabolism, lipid body formation, and macrophage function. In this study, we determined that mice lacking PPARα exhibited reduced inflammatory hallmarks of STm infection, including lowe
