Optimizing environmental DNA (eDNA) metabarcoding to maximize species detection while minimizing costs and effort remains a challenge, particularly in high-throughput workflows. In the marine intertidal zone off Point Conception, California, we compared eDNA richness accumulation between filtration approaches that differed in filter type and downstream processing, field and laboratory replicates, and independent and pooled polymerase chain reaction (PCR) replicates. Ten 10-cm2 capsule filters and 10 5-cm2 syringe disc filters were processed using a standardized laboratory workflow for 12S ribo

Ixodes scapularis, the blacklegged deer tick, is the principal vector of Lyme disease in North America. Environmental factors are known to influence regional and seasonal incidence of Lyme disease and possibly the endemicity of the disease to the northeastern and upper mid-western regions of the United States. With a goal to understand the impact of environmental temperature on microbial communities within the tick, we investigated the bacterial microbiome of colony-reared I. scapularis ticks statically incubated at different temperatures (4, 20, 30, and 37°C) at a constant humidity in a contr

Preservation of human blood cells for DNA damage analysis with the comet assay conventionally involves the isolation of mononuclear cells by centrifugation, suspension in freezing medium and slow freezing to -80 °C-a laborious process. A recent publication (Al-Salmani et al. Free Rad Biol Med 2011; 51: 719-725) describes a simple method in which small volumes of whole blood are frozen to -20 or -80 °C; on subsequent thawing, the comet assay is performed, with no indication of elevated DNA strand breakage resulting from the rapid freezing. However, leucocytes in whole blood (whether fresh or fr

In a soil microcosm experiment, the influence of low-level (137)Cs and (90)Sr contamination on the degradation of (14)C-ring-labeled 2,4-dichlorophenoxyacetic acid (2,4-D) was studied. Two differently treated soils (one native soil and one soil sterilized and reinoculated with a biotic soil aliquot) were artificially contaminated with various concentrations of (137)Cs and (90)Sr as nitrate salts. The cumulative doses increased up to 4 Gy for 30 days of incubation in soil microcosms. Changes in microbial community structure were observed with help of the denaturing gradient gel electrophoresis

A Cu2+ fluorescent probe molecule (S) of the "ON-OFF" type was designed and synthesized using 4-(diphenylamino)benzaldehyde and carbohydrazide as starting materials. The structure of S was systematically characterized using IR spectroscopy, HRMS, 1H NMR, and 13C NMR. Spectroscopic experiments demonstrated that the binding stoichiometry of S to Cu2+ was 1:1, with remarkable selectivity and sensitivity for detecting Cu2+. Moreover, the detection limit of 3.5 × 10-8 mol⋅L-1 was significantly lower than those reported for previously developed fluorescent sensors targeting Cu2+. Density functional

During preimplantation development, a wave of genome-wide DNA demethylation occurs to acquire a hypomethylated genome of the blastocyst. As an essential epigenomic event, postfertilization DNA demethylation is critical to establish full developmental potential. Despite its importance, this process is prone to be disrupted due to environmental perturbations such as manipulation and culture of embryos during in vitro fertilization (IVF), and thus leading to epigenetic errors. However, since the first case of aberrant DNA demethylation reported in IVF embryos, its underlying mechanism remains unc

Exercise is an effective approach for primary and secondary prevention of cardiovascular diseases (CVD) and loss of muscular mass and function. Its benefits are widely documented but incompletely characterized. It has been reported that exercise can induce changes in the expression of antioxidant enzymes including Sod2, Trx1, Prdx3 and Gpx1 and limits the rise in oxidative stress commonly associated with CVD. These enzymes can be subjected to epigenetic regulation, such as DNA methylation, in response to environmental cues. The aim of our study was to determine whether in the early stages of a

Atherosclerosis (AS) is a disease induced by multiple factors, including genetic and environmental elements. The aim of the present study is to investigate the comprehensive effects of high cholesterol, high methionine diet, and apolipoprotein E deficiency (ApoE(-/-)) on the pathogenesis of AS. ApoE(-/-) mice were fed with high cholesterol and methionine diet for 15 weeks to induce hyperlipidemia and hyperhomocysteinemia. The methylation levels of genomic DNA (gDNA) and B1 repetitive elements in aortic tissues were measured by both methylation-dependent restriction analysis and nested methylat

Estuarine and river-influenced coastal ecosystems are recognized as important sinks and channels for transfer of heavy metals into the marine environment. Continuous intake of metal pollutants could create chronic exposure situations, perhaps leading to molecular and cellular damage to resident biota, even if environmental concentrations are within regulatory limits. Thus, the incorporation of molecular and genotoxicity biomarkers into environmental monitoring programs has received growing interest, as changes in gene expression are among the earliest detectable responses to pollutant stress a

Rheumatoid arthritis (RA) is an autoimmune disease characterized by chronic synovitis that may progress to irreversible joint destruction and disability, thereby substantially impairing quality of life. RA results from complex interactions among genetic predisposition, environmental exposures, and immune dysregulation; however, current therapies are not curative, and many patients continue to experience pain, morning stiffness, and recurrent inflammation. In recent years, epigenetic mechanisms have emerged as key modulators of RA heterogeneity and disease persistence. Reversible regulatory lay

Exposure to hazardous substances from landfill frequently occurs through complex chemical mixtures called leachates with a significant source of environmental and health concern. This study generated simulated leachates from soils collected at three landfill sites, Obajana (OBSL), Ajaokuta (AJSL), and Anyigba (AYSL) simulated leachate and evaluated their genotoxic and molecular effects using Mus musculus as a model organism. The leachates were characterized for physicochemical properties and leachate pollution indices. Mice orally exposed to 4 different (15 - 75 %) concentrations of each test

Progress over the past 30 years has revealed many strengths of the rainbow trout as an alternative model for environmental carcinogenesis research. These include low rearing costs, an early life-stage ultrasensitive bioassay, sensitivity to many classes of carcinogen, a well-described tumor pathology, responsiveness to tumor promoters and inhibitors, and a mechanistically informative nonmammalian comparative status. Low-cost husbandry, for example, has permitted statistically challenging tumor study designs with up to 10,000 trout to investigate the quantitative interrelationships among carcin

Smoking, a pivotal environmental risk factor, drives diseases including lung cancer through genetic and epigenetic alterations. While 2-Naphthylamine (2-NA), a tobacco-derived carcinogen, is established as a bladder carcinogen via DNA damage, its role in lung carcinogenesis remains mechanistically uncharacterized despite epidemiological associations. This study identifies bronchial epithelium as a direct target of 2-NA, demonstrating dose-dependent DNA damage in 16HBE cells (peak at 250 μM, 12 h; P < 0.05), accompanied by S-phase arrest, apoptosis, reduced proliferation, and reactive oxygen sp

This report describes the visible-light-induced one-pot multicomponent regioselective synthesis of a series of 5-aroyl-3-((arylidene)amino)-2-((arylidene)hydrazono)-4-methyl-2,3-dihydrothiazoles as DNA and BSA targeting agents. The multicomponent condensation of thiocarbohydrazide and aldehydes with α-bromo-1,3-diketones, generated in situ by the bromination of unsymmetrical 1,3-diketones with NBS using white LED light as an environmental friendly source in the presence of EtOAc solvent furnished the titled 2,3-dihydrothiazole derivatives in excellent yields. The exact regioisomeric structure

Nonmelanoma skin cancer is the most common cancer in the United States, where DNA-damaging ultraviolet B (UVB) radiation from the sun remains the major environmental risk factor. However, the critical genetic targets of UVB radiation are undefined. Here we show that attenuating PTEN in epidermal keratinocytes is a predisposing factor for UVB-induced skin carcinogenesis in mice. In skin papilloma and squamous cell carcinoma (SCC), levels of PTEN were reduced compared with skin lacking these lesions. Likewise, there was a reduction in PTEN levels in human premalignant actinic keratosis and malig

The routine and often unavoidable exposure to solar ultraviolet (UV) radiation makes it one of the most significant environmental DNA-damaging agents to which humans are exposed. Sunlight, specifically UVB and UVA, triggers various types of DNA damage. Although sunlight, mainly UVB, is necessary for the production of vitamin D, which is necessary for human health, DNA damage may have several deleterious consequences, such as cell death, mutagenesis, photoaging and cancer. UVA and UVB photons can be directly absorbed not only by DNA, which results in lesions, but also by the chromophores that a

Recently, inorganic arsenite (iAs(III)) and its mono- and dimethylated metabolites have been examined for their interference with the formation and repair of benzo[a]pyrene diol epoxide (BPDE)-induced DNA adducts in human cells (Schwerdtle, ., Walter, I., and Hartwig, A. (2003) DNA Repair 2, 1449 - 1463). iAs(III) and monomethylarsonous acid (MMA(III)) were found to be able to enhance the formation of BPDE-DNA adducts, whereas dimethylarsinous acid (DMA(III)) had no enhancing effect at all. The anomaly manifested by DMA(III) prompted us to further investigate the effects of the three trivalent

Environmental toxicants, including chlorinated solvents present in contaminated water sources, can modify human DNA methylation (DNAm), an epigenetic mechanism that underlies biological plasticity in response to environmental perturbations. Given the relevance of these epigenetic mechanisms, this study examines the DNAm profiles of communities near Bussi sul Tirino (Abruzzo, Italy), an area declared a Site of National Interest (SNI) in 2008 due to extensive environmental contamination, including chlorinated solvents. The aim was to determine whether long-standing groundwater contamination is a

Non-invasively collected faecal samples are an alternative source of DNA to tissue samples, that may be used in genetic studies of wildlife when direct sampling of animals is difficult. Although several faecal DNA extraction methods exist, their efficacy varies between species. Previous attempts to amplify mitochondrial DNA (mtDNA) markers from faeces of wild dugongs (Dugong dugon) have met with limited success and nuclear markers (microsatellites) have been unsuccessful. This study aimed to establish a tool for sampling both mtDNA and nuclear DNA (nDNA) from dugong faeces by modifying approac
