The value of laboratory testing in uveitis.

Accurate diagnosis of uveitis is of great importance since the treatment for the various uveitis entities may differ considerably. In a large number of cases the clinical picture is sufficient to make an adequate diagnosis. There are cases in which the diagnosis cannot be made on clinical grounds alone and support is needed from laboratory tests. Only a limited number of tests have been proven to be useful as a diagnostic or prognostic aid. These include HLA-B27 typing in patients presenting with anterior uveitis and testing for angiotensin converting enzyme and lysozyme in case of suspected s

Renin-angiotensin system in sepsis.

The time course of the components of the renin-angiotensin system was investigated in the plasma of three patients on the intensive care unit. Two of them, which were both polytraumatized, suffered from adult respiratory distress syndrome (ARDS). All patients had sepsis and impaired pulmonary and renal function. Plasma samples were investigated for up to two weeks, in which time all three patients showed a decrease in their angiotensin converting enzyme (ACE) plasma concentration. Two of the patients with deteriorating renal function had three to four times elevated angiotensinogen (Ao) plasma

Familial elevation of serum angiotensin converting enzyme.

We report here a familial clustering of elevated serum angiotensin converting enzyme (ACE) levels. The patient is a 58-year-old Japanese female who had been in excellent health until age 45 when she developed an occlusion of the left central retinal vein. She was otherwise in excellent health, and no laboratory abnormality except a marked elevation of serum ACE level (625 nmol/min/ml; normal range; 22-40 nmol/min/ml of serum) was found. Her blood pressure was within normal limits (140/80 mmHg). There was no evidence for the diagnosis of sarcoidosis, Gaucher's disease, leprosy, hyperthyroidism,

Cardiovascular effects of discrete intrahypothalamic and preoptic injections of bradykinin.

Blood pressure and heart rate were monitored during discrete injections of bradykinin (5 nmol; 100-300 nl) in the hypothalamus and preoptic area of halothane anesthetized rats. In the paraventricular nucleus, bradykinin produced bradycardia without effecting blood pressure. The decrease in heart rate was abolished by pretreatment with methylatropine (IP), suggesting that the parasympathetic nervous system mediates this response. In contrast, in the dorsomedial and posterior hypothalamic nuclei, bradykinin increased both heart rate and blood pressure; methylatropine pretreatment (but not adrena

Gallium-67 in the evaluation of sarcoidosis: correlations with serum angiotensin-converting enzyme and bronchoalveolar lavage.

Gallium-67 (67Ga) scanning was assessed for its usefulness in the evaluation and follow-up of 54 patients with sarcoidosis, both treated and untreated. Scans were repeated in 23 subjects. Serum levels of angiotensin-converting enzyme (ACE) were determined concurrently in all 54 patients and bronchoalveolar lavage was performed in 29 patients. Gallium-67 scan was effective in the detection and assessment of lesions not revealed by traditional methods of investigation, particularly those affecting the mediastinum, spleen, and salivary glands. The scan also enabled fibrotic lesions, which do not

Cryo-EM structure of locked spike glycoprotein from bat SARS-like coronavirus WIV1, molecular dynamics and biophysics across host range.

As made evident by severe acute respiratory syndrome coronavirus 1 (SARS-CoV-1) and SARS-CoV-2 pandemics, the possibility of a SARS-like coronavirus (SL-CoV) emerging again in humankind after zoonotic spillover represents a significant global health threat. Given the role of spike (S) glycoprotein in mediating SL-CoV cell entry and cross-species transmission, there remains urgent need of structural information on SL-CoV S to guide therapeutic countermeasure development. Among SL-CoVs, bat-derived WIV1 is capable of using as receptor the angiotensin converting enzyme 2 (ACE2) from a variety of

Tiliroside From Pavonia malacophylla (Link & Otto) Garcke as an Anti-SARS-CoV-2 Cell Entry Agent.

Flavonoids are recognized for interfering with stages of SARS-CoV-2 life cycle. Then, tiliroside (kaempferol 3-O-glucoside-6''-E-coumaroyl), an O-glycosylated flavonoid isolated from Pavonia malacophylla (Link & Otto) Garcke, was investigated for its ability to block SARS-CoV-2 viral entry. Tiliroside was chemically characterized and tested for inhibition of the SARS-CoV-2 Spike receptor-binding domain (RBD)-human ACE2 interaction using a bioluminescent immunoassay. At 10 µM, it inhibited 63% of RBD:ACE2 binding. Flow cytometry and pseudovirus assays assessed Spike internalization in endotheli

Inducible CD147 up-regulation boosts extended SARS-CoV-2 infection triggering severe COVID-19 independent of ACE2.

The high mortality caused by severe COVID-19 poses great challenges to the public health. However, the underlying pathogenesis of severe cases remains unclear. Here, we find that SARS-CoV-2 infection boosts CD147 inducible up-regulation in the lung tissues of virus-infected rhesus macaques coupled with down-regulated membrane-bound ACE2, which conduces to extended virus infection and severe pathological lesions. Specifically, SARS-CoV-2 infection enhances the expression of transcriptional factor aryl hydrocarbon receptor and facilitates its nucleus translocation, which causes CD147 gene transc

Association of ACE I/D polymorphism with blood pressure response to valsartan-hydrochlorothiazide combination therapy in hypertensive patients from Khyber Pakhtunkhwa, Pakistan.

BACKGROUND: Interindividual variability in antihypertensive response may be partly explained by genetic variation. The angiotensin-converting enzyme insertion/deletion polymorphism (ACE I/D; rs1799752) has been associated with variable response to renin-angiotensin-aldosterone system-targeting therapies, but evidence for valsartan-hydrochlorothiazide (Val-HCTZ) combination therapy remains limited, particularly in South Asian populations. METHODS: In this multicenter prospective cohort study, 645 hypertensive patients from Khyber Pakhtunkhwa, Pakistan, receiving fixed-dose Val-HCTZ were enroll

Bioluminescent label-free molecular switch for SARS-CoV-2 detection.

BACKGROUND: Simple, single-step platforms for virus detection are still important for improving diagnostic testing beyond laboratory settings. Molecular switch platforms, which generate measurable signals upon specific target binding, represent a promising strategy for virus detection due to their modularity, high sensitivity, and potential adaptability to emerging pathogens. RESULTS: Here, we present CovSwitch, a novel one-step bioluminescent molecular switch platform engineered for rapid, sensitive, and potentially point of care detection-compatible detection of severe acute respiratory syn

Boosting angiotensin-converting enzyme (ACE) in microglia protects against Alzheimer's disease in 5xFAD mice.

Genome-wide association studies have identified many gene polymorphisms associated with an increased risk of developing late-onset Alzheimer's disease (LOAD). Many of these LOAD risk-associated alleles alter disease pathogenesis by influencing innate immune responses and lipid metabolism of microglia (MG). Here we show that boosting the expression of angiotensin-converting enzyme (ACE), a genome-wide association study LOAD risk-associated gene product, specifically in MG, reduces amyloid-β (Aβ) plaque load, preserves vulnerable neurons and excitatory synapses, and significantly reduces learnin

Discovering the natural source-derived antihypertensive compounds aspiring current therapeutic targets by computer-based drug design.

Hypertension, a primary global health concern, heightens the risk of heart, brain, and kidney diseases and contributes to premature mortality. The present paper thoroughly explores the potential therapeutic use of natural compounds, drawing on Avicenna's ancient medical philosophy to highlight the role of bodily humor imbalances and lifestyle factors in hypertension. A key aspect of our study is using a computer-aided drug design (CADD) approach, which we employ to identify novel antihypertensive bio-compounds, focusing on current therapeutic targets, including angiotensin I-converting enzyme,

Structure-activity relationship and molecular interaction mechanisms of the ACE-inhibitory tripeptide LL-X.

Investigating the molecular interaction mechanisms of angiotensin-converting enzyme (ACE) inhibitory peptides is valuable in antihypertensive drug development. In this study, we designed two novel peptide variants, Leu-Leu-Phe (LLF) and Leu-Leu-Pro (LLP), by substituting the C-terminal tyrosine (Tyr) in casein-derived Leu-Leu-Tyr (LLY) with phenylalanine (Phe) and proline (Pro), respectively, to investigate the impact of C-terminal amino acid properties of peptide on ACE inhibition. The ACE inhibitory activity, inhibition kinetics, peptides-ACE binding mechanisms, and LLF and LLP conformationa

Diagnosis and Treatment of Inherited Renal Tubular Dysgenesis Caused by ACE Gene Mutation: A Single-Center Experience.

INTRODUCTION: Autosomal recessive renal tubular dysgenesis (ARRTD) is a rare and severe condition, and current methods for early diagnosis and treatment of this disease remain unclear. Here, we describe 3 patients with ARRTD. CASE PRESENTATION: The 3 cases come from three independent pedigrees and harbor completely different ACE gene mutation types. Two infants died of the disease shortly after birth despite treatment attempts, while one infant showed improvement with symptomatic therapies including blood pressure elevation, respiratory support, diuretics, and CRRT and was discharged. During

Human transferrin receptor can mediate SARS-CoV-2 infection.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection has been detected in almost all organs of coronavirus disease-19 patients, although some organs do not express angiotensin-converting enzyme-2 (ACE2), a known receptor of SARS-CoV-2, implying the presence of alternative receptors and/or co-receptors. Here, we show that the ubiquitously distributed human transferrin receptor (TfR), which binds to diferric transferrin to traffic between membrane and endosome for the iron delivery cycle, can ACE2-independently mediate SARS-CoV-2 infection. Human, not mouse TfR, interacts with

Pharmacological inhibition of the ACE/Ang-2/AT1 axis alleviates mechanical ventilation-induced pulmonary fibrosis.

Mechanical ventilation (MV) is an essential therapy for acute respiratory distress syndrome (ARDS) and pulmonary fibrosis. However, it can also induce mechanical ventilation-induced pulmonary fibrosis (MVPF) and the underlying mechanism remains unknown. Based on a mouse model of MVPF, the present study aimed to explore the role of the angiotensin-converting enzyme/angiotensin II/angiotensin type 1 receptor (ACE/Ang-2/AT1R) axis in the process of MVPF. In addition, recombinant angiotensin-converting enzyme 2(rACE2), AT1R inhibitor valsartan, AGTR1-directed shRNA and ACE inhibitor perindopril we
