Evaluation of some neuropharmacological effects of Caladium bicolor aiton (araceae) leaf extracts in mice.

Caladium bicolor Aiton (Araceae) is used in ethnomedicine for the treatment of boils, wound ulcers and convulsion. This study investigated the effects of the leaf extracts on some neuropharmacological parameters. The leaves were collected, dried, powdered and then extracted by maceration in methanol to yield the whole extract (WE). Extraction was also done using n-hexane, ethyl acetate and methanol in a Soxhlet apparatus to obtain n-hexane (HE), ethyl acetate (EA) and methanol (ME) extracts. Preliminary phytochemical screening was done using the whole extract. Some neuropharmacological evaluat

Inhibition of mTORC1 Signaling Reverts Cognitive and Affective Deficits in a Mouse Model of Parkinson's Disease.

Non-motor symptoms, including cognitive deficits and affective disorders, are frequently diagnosed in Parkinson's disease (PD) patients and are only partially alleviated by dopamine replacement therapy. Here, we used a 6-hydroxydopamine (6-OHDA) mouse model of PD to examine the effects exerted on non-motor symptoms by inhibition of the mammalian target of rapamycin complex 1 (mTORC1), which is involved in the control of protein synthesis, cell growth, and metabolism. We show that rapamycin, which acts as an allosteric inhibitor of mTORC1, counteracts the impairment of novel object recognition.

Deletion of ovarian hormones induces a sickness behavior in rats comparable to the effect of lipopolysaccharide.

Neuroimmune factors have been proposed as the contributors to the pathogenesis of sickness behaviors. The effects of female gonadal hormones on both neuroinflammation and depression have also been well considered. In the present study, the capability of deletion of ovarian hormones to induce sickness-like behaviors in rats was compared with the effect lipopolysaccharide (LPS). The groups were including Sham, OVX, Sham-LPS, and OVX-LPS. The Sham-LPS and OVX-LPS groups were treated with LPS (250 μg/kg) two hours before conducting the behavioral tests. In the forced swimming (FST), the immobility

Prodepressive effect induced by microinjections of MCH into the dorsal raphe: time course, dose dependence, effects on anxiety-related behaviors, and reversion by nortriptyline.

Melanin-concentrating hormone (MCH) administered within the rat dorsal raphe nucleus (DRN) has been shown to elicit prodepressive behaviors in the forced-swim test. The present study was designed to evaluate the time course (30 and 60 min) and dose dependence (25-100 ng) of this effect, and whether it would be antagonized by an intra-DRN microinjection of the MCH-1 receptor antagonist ATC0175 (ATC, 1 mmol/l) or intraperitoneal pretreatment with the noradrenergic antidepressant nortriptyline (20 mg/kg). The results showed that the behavioral effect of MCH was time and dose dependent as immobili

Isolation of anxiolytic principle from ethanolic root extract of Cardiospermum halicacabum.

Cardiospermum halicacabum roots have been used traditionally for the treatment of epilepsy and anxiety disorders. The purpose of this study was to characterize the putative phytoconstituents present in the ethanolic root extract having anxiolytic activity using an elevated plus-maze (EPM) and light dark transition model. Control mice were orally treated with an equal volume of vehicle (4% gum acacia), and positive control mice were treated with diazepam (1mg/kg). In the EPM test, out of pool of 19 master fractions (MF) only MF-14, 16 and 17 significantly (30mg/kg, p<0.05, p<0.01 and p<0.001) i

Early Postnatal Androgenic Activity Inhibition Produces Differential Long-Term Alterations in Anxiety-Like Behavior in Young Adult Male and Female Wistar Rats.

Sex differences in anxiety-like behavior are influenced by gonadal steroids, yet the contribution of early postnatal androgen signaling and testosterone metabolic pathways to the long-term organization of these behaviors remains unclear. Wistar male and female rats received daily s.c. injections from postnatal day (P) 1 to P5 of vehicle, flutamide, letrozole, or finasteride. In young adult animals, locomotor activity and anxiety-like behavior were evaluated via the open field test (OFT) and elevated plus maze (EPM). Females showed higher locomotor activity than males in both tests. Control fem

Electroacupuncture Alleviates Pain and Anxiety in CFA-Induced Mice by Suppressing Glutamatergic Neurons in the Spinoparabrachial Pathway.

The spinoparabrachial tract is a major ascending pathway that transmits nociceptive signals from the spinal cord to the brain, but whether electroacupuncture (EA) relieves pain-anxiety comorbidity by modulating this pathway remains unclear. Here, we investigated the effects of EA on mechanical allodynia and anxiety-like behaviors in Complete Freund's adjuvant (CFA)-induced inflammatory pain mice using von Frey testing, the open field test, and the elevated plus maze, combined with chemogenetic manipulation of glutamatergic projections from the spinal dorsal horn (SDH) to the lateral parabrachi

Daily Administration of Psilocin Mucate (L-130) Produces a Favorable Safety Profile and Anxiolytic Effects in Rodents Exposed to Chronic Unpredictable Mild Stress.

Anxiety disorders are chronic health conditions affecting the quality of life of millions of people. Psilocin, the active moiety of psilocybin, provides an anxiolytic effect; however, when orally administered as psilocybin, it only offers a moderate level of bioavailability and less predictable pharmacokinetics, potentially making effects after absorption variable and increasing the risk of adverse hallucinations, depending on the dose. As such, we investigated a recently developed stable salt of psilocin, psilocin mucate (L-130), which delivers increased bioavailability and, thus, more precis

Psilocybin restores behavioral and neuroplastic deficits induced by chronic stress in rats.

Psychedelics have emerged as a promising novel therapeutic approach for major depressive disorder (MDD). Altered activity and structural atrophy of the prefrontal cortex, hippocampus, and limbic structures are associated with depressive disorders. Psilocybin may reverse the loss of synaptic connections and restore the function of these brain regions. In this study, we investigated the effects of psilocybin on rat behavior, hippocampal neurogenesis, expression level of brain-derived neurotrophic factor (BDNF) and hypothalamic-pituitary-adrenal (HPA) axis activity. Psilocybin administered in two

Effects of environmental enrichment during adolescence on social defeat effects.

Social defeat (SD) is a well established model that increases addiction vulnerability accompanied by depressive- and anxiety-like behaviors. Environmental enrichment (EE) has been shown to enhance resilience and mitigate stress-induced behavioral alterations. Here, we investigated the protective effects of EE during adolescence, both before and during SD encounters, on stress-induced anxiety, depression and the increased conditioned rewarding effects of a subthreshold cocaine dose in adulthood. We employed the social interaction test (SIT) to categorize mice into resilient and susceptible phen

IL-6 Inhibition Partially Ameliorates Maternal Immune Activation-Induced Autism-Like Behavioral Abnormalities in Mice.

Prenatal maternal immune activation (MIA) has been implicated in autism spectrum disorder (ASD) pathogenesis, with interleukin-6 (IL-6) identified as a key inflammatory mediator. We investigated the therapeutic potential of IL-6 inhibition in an MIA mouse model induced by Toxoplasma gondii soluble tachyzoite antigen (STAg). Adult MIA offspring received systemic administration of the IL-6-neutralizing antibody (MP5-20F3) or isotype control, followed by behavioral assessments one week later. Open field and elevated plus maze tests revealed heightened anxiety-like behaviors in the STAg offspring,…

Adolescent activity-based anorexia has a substantial and prolonged impact on social behavior in young adult female rats.

Activity-based anorexia (ABA) is a rodent model of anorexia nervosa (AN) that induces several key components of AN, including voluntary reduction in food intake, reduced body weight, hyperactivity, and alterations to the hypothalamic-pituitary-adrenal (HPA) axis. Previous research has demonstrated persistently increased anxiety-like behavior in the elevated plus maze (EPM), a test measuring avoidance of novel and open areas in adult female rats that experienced ABA during adolescence and are weight-restored in adulthood. Whether the same behavioral effects of two bouts of adolescent ABA emerge

Limited Cheese Intake Paradigm Replaces Patterns of Behavioral Disorders in Experimental PTSD: Focus on Resveratrol Supplementation.

Currently, the efficacy of drug therapy for post-traumatic stress disorder or PTSD leaves much to be desired, making nutraceutical support a promising avenue for treatment. Recent research has identified the protective effects of resveratrol in PTSD. Here, we tested the behavioral and neurobiological effects of combining cheese consumption with resveratrol supplements in an experimental PTSD model. Using the elevated plus maze test, we observed that cheese intake resulted in a shift from anxiety-like behavior to depressive behavior, evident in increased freezing acts. However, no significant c…

Chronic MAP4343 reverses escalated alcohol drinking in a mouse model of alcohol use disorder.

Alcohol use disorders can be driven by negative reinforcement. Alterations of the microtubule cytoskeleton have been associated with mood regulation in the context of depression. Notably, MAP4343, a pregnenolone derivative known to promote tubulin assembly, has antidepressant properties. In the present study, we tested the hypothesis that MAP4343 may reduce excessive alcohol drinking in a mouse model of alcohol dependence by normalizing affect during withdrawal. Adult male C57BL/6J mice were given limited access to voluntary alcohol drinking and ethanol intake escalation was induced by chronic

The age-dependent effect of pre-pubertal castration on anxiety-like behaviour in male rats.

Adolescence is considered to be a critical period of sex hormone action (re)organising the brain and determining the behavioural phenotype. Such organisational effects in the brain might be the cause of sex differences in some behavioural features. In this experiment, we aimed to examine the role of pubertal sex hormones in development of anxiety in male rats. Male rats underwent gonadectomy prior to puberty onset, and were tested for explorative and anxiety-like behaviour in adolescence as well as in young adulthood. In adolescence, but not in adulthood, gonadectomised rats spend by 50% more

Increased 4R tau expression and behavioural changes in a novel MAPT-N296H genomic mouse model of tauopathy.

The microtubule-associated protein tau is implicated in various neurodegenerative diseases including Alzheimer's disease, progressive supranuclear palsy and corticobasal degeneration, which are characterized by intracellular accumulation of hyperphosphorylated tau. Mutations in the tau gene MAPT cause frontotemporal dementia with parkinsonism linked to chromosome 17 (FTDP-17). In the human central nervous system, six tau isoforms are expressed, and imbalances in tau isoform ratios are associated with pathology. To date, few animal models of tauopathy allow for the potential influence of these

Estradiol decreases rat depressive behavior by estrogen receptor beta but not alpha: no correlation with plasma corticosterone.

17β-Estradiol (E2) may influence some of the sex differences in stress-related neuropsychiatric disorders, such as anxiety and depression. E2 may also modulate the hypothalamic-pituitary-adrenal axis function as a cortisol response to stress in women. This study explored the role of E2 (10 and 50 μg/rat) and selective estrogen receptor modulators: diarylpropionitrile (DPN, 10 μg/rat) and propyl pyrazole triol (PPT, 10 μg/rat), in anxiety and depressive behaviors in ovariectomized rats using an animal model. The study also examined the relationship between rats' affective behavior and the plasm
