Effects of morphine and naloxone on feline colonic transit.

The effects of endogenous and exogenous opioid substances on feline colonic transit were evaluated using colonic transit scintigraphy. Naloxone (0.3 mg/kg, i.m.) accelerated emptying of the cecum and ascending colon, and filling of the transverse colon. Endogenous opioid peptides thus appear to play a significant role in the regulation of colonic transit. At a moderate dose of morphine (0.1 mg/kg, i.m.), cecum and ascending colon transit was accelerated, while at a larger dose (1.0 mg/kg, i.m.) morphine had no effect. Since naloxone, a relatively nonspecific opioid antagonist, and morphine, a

[The endogenous opiate and nonopiate analgesic system].

A lot of researches indicate that central nervous system not only exist endogenous opiate analgesic system but also possess endogenous nonopiate analgesic system. The opiate and nonopiate analgesic systems can be selectively activated by different environmental or artificial manipulations and modulated the stimulation-produced analgesia, acupuncture analgesia and stress-induced analgesia, including the foot shock-induced analgesia, tail-shock-produced analgesia, ear-shock-induced analgesia, analgesia of cold water swim, analgesia of immobilization or of restraint and analgesia of a-Deoxy-D- gl

Involvement of monoaminergic and peptidergic components in cathinone-induced analgesia.

Evidence has been obtained suggesting that cathinone-induced analgesia depends upon stimulation of alpha-adrenoceptors, followed by release of opioid peptides and by activation of serotonergic pathways. This hypothesis is supported by the following. (1) Cathinone potentiated morphine analgesia and the whole effect was antagonized by naloxone whereas onto the cathinone potentiation was counteracted by phenoxybenzamine. (2) Bestatin potentiated cathinone-induced analgesia and this effect was sensitive to both naloxone and phenoxybenzamine blockade. (3) The analgesic effect of cathinone + bestati

Effect of naloxone, a specific opioid inhibitor, on blood pressure fall during sleep.

The study was designed to investigate the possible role of endogenous opioids in the fall in blood pressure (BP) seen during initial sleep. Seven normal men, ages 20-30 years, were studied for three consecutive nights. Each night, electroencephalogram, chin electromyogram, electrooculogram, heart rate (all continuously), and blood pressure (every 15 minutes) were recorded. Night 1 was used for orientation. On nights 2 and 3, subjects received, in randomized order, an infusion of naloxone 0.2 mg/kg over 1 minute or volume-matched saline. Blood pressure data from the first 4 hours of non-rapid e

Spinal cord stimulation combined with exercise in patients diagnosed with persistent spinal pain syndrome. Study protocol for a randomized control trial.

INTRODUCTION: Administration of spinal cord stimulation to individuals with PSPS-T1/2 may induce supraspinal descending activation. Similarly, exercise is recognized as a fundamental aspect of spinal pain management. Studies have demonstrated its impact on neurophysiological factors, including the release of spinal and supraspinal beta-endorphins, which activate μ-opioid receptors. Therefore, the purpose of this study will be to examine the effect of SCS in combination with lumbo-pelvic stability core training on perceived low back pain, quality of life and disability in PSPS-T2 patients. [Sports, bones and hormones. Multiple interactions].

Classically, sports activities are thought to have a beneficial effect on bone tissues. Actually, there are many interactions between sports … icient pulsatile secretion of luteo-releasing hormone and subsequent hypogonadism. The role of beta-endorphins or catelestrogens on hypothalamic receptors has been suggested as the underlying mechanism. These different observations help provide answers to the different problems raised when providing counselling and care for high level athletes.

Opioid receptors signaling network.

Opioid receptors belong to the class A G-protein-coupled receptors and are activated by alkaloid opiates such as morphine, and endogenous ligands such as endorphins and enkephalins. Opioid receptors are widely distributed in the human body and are involved in numerous physiological processes through three major classical opioid receptor subtypes; the mu, delta and kappa along with a lesser characterized subtype, opioid receptor-like (ORL1). Opioids are the most potent analgesics and have been extensively used as a therapeutic drug for the treatment of pain and related disorders. Chronic admini…

[New findings on endogenous drug receptor ligands].

Since the discovery of endogenous ligands for opiate receptors, endorphins and enkephalins some 30 years have elapsed. The fact that one of the breakdown products of the common precursor (proopiomelanocortin, POMC) ACTH and opioid peptides, i.e. ACTH 1-24 has a saluretic effect (balancing perhaps sodium retention after activation of the adrenal cortex) was not generally accepted yet although MSH as well as some fragments of the ACTH molecule have a natriuretic effect. The endogenous digitalis-like immunoreactive factor, DLF (even ACTH displays some DLF activities) which inhibits NaK-ATPase ori…

Dexamethasone differentially alters naltrexone effects on vasopressin and oxytocin release during tail electroshock.

The origin of endogenous opioid peptides that inhibit release of vasopressin (VP) and oxytocin (OT) into the bloodstream after tail electroshock was investigated. We hypothesized that endogenous opioid peptides derived from the anterior pituitary reduced secretion of VP and OT during this stimulus. To test this hypothesis, dexamethasone (DEX) was used to preferentially suppress release of endorphins with ACTH from the anterior pituitary. We evaluated the effects of an opiate receptor antagonist, naltrexone, on the rise in plasma [VP] and [OT] after tail electroshock in male Sprague-Dawley rats…

Pheochromocytoma in Old World Primates (Macaca mulatta and Chlorocebus aethiops).

Pheochromocytoma, a rarely reported adrenal gland neoplasm in Old World primates, was diagnosed in 5 rhesus macaques (Macaca mulatta) and 2 African green monkeys (Chlorocebus aethiops) from 3 research institutions. Age and sex were available for 6 primates. Two males and 4 females were affected, ranging in age from 9 to 31 years. All neoplasms were unilateral and, in the cases reporting the affected gland, 4 involved the right adrenal gland and 2 involved the left. Diagnosis was established by characteristic histologic features. Immunohistochemically, neoplastic cells in all cases expressed ch

Immunohistochemical demonstration of dopamine receptor D2R in the primary cilia of the mouse pituitary gland.

Dopamine regulates the synthesis and secretion of prolactin and α-MSH/β-endorphin in lactotrophs and melanotrophs, respectively. While a predominant dopamine receptor, D2R, is known to be expressed in both the anterior and intermediate lobes of the pituitary gland, no previous immunohistochemical studies have shown the existence of D2R in the plasma membrane of pituitary endocrine cells. The present study clearly demonstrated a selective localization of the D2R immunoreactivity in primary cilia of lactotrophs and melanotrophs in the mouse adenohypophysis. Another immunoreactivity of D2R was fo

Central role of p53 in the suntan response and pathologic hyperpigmentation.

UV-induced pigmentation (suntanning) requires induction of alpha-melanocyte-stimulating hormone (alpha-MSH) secretion by keratinocytes. alpha-MSH and other bioactive peptides are cleavage products of pro-opiomelanocortin (POMC). Here we provide biochemical and genetic evidence demonstrating that UV induction of POMC/MSH in skin is directly controlled by p53. Whereas p53 potently stimulates the POMC promoter in response to UV, the absence of p53, as in knockout mice, is associated with absence of the UV-tanning response. The same pathway produces beta-endorphin, another POMC derivative, which p

A case of post-Japanese encephalitis with partial hypothalamic dysfunction showing repetitive hyperthermia in summertime.

Japanese encephalitis (JE) virus is a mosquito-borne virus belonging to the flavivirus family, including the West Nile and St Louis encephalitis viruses endemic to North America. JE virus is prevalent in East Asian countries and can cause acute lethal encephalitis. Although vaccination programs have decreased the incidence of JE in Japan, the cases that do occur are often fatal or associated with considerable clinical sequelae. We report, for the first time to our knowledge, a patient who had repetitive bouts of hyperthermia in the summertime after recovery from acute JE. An insulin challenge
