IL-17-producing T lymphocytes in lung tissue and in the bronchoalveolar space after exposure to endotoxin from Escherichia coli in vivo--effects of anti-inflammatory pharmacotherapy.

Interleukin (IL)-17 may play a critical role for the innate immune response in mammals. However, little is known about its production in T lymphocytes in comparison with other cells, in lung tissue and in the bronchoalveolar space in vivo. Even less is known about the effects of anti-inflammatory pharmacotherapy on this IL-17 production. In this study on mice we show that one single, intranasal exposure to endotoxin from Escherichia coli increases extracellular IL-17 protein in bronchoalveolar (BAL) samples during 3 days, and is accompanied by a local increase in neutrophils and other inflamma

Novel actin rings within the secretory cells of honeybee royal jelly glands.

We describe a novel cytoskeletal element within secretory cells of an arthropod gland system, the hypopharyngeal gland of the honeybee, Apis mellifera. The hypopharyngeal secretory cells are the source of royal jelly in nurse bees and enzymes in foragers. Each cell possesses an elongate invagination that is occupied by a tubular cuticular structure, the end-apparatus, that accumulates secretion and transfers it into a cuticular microtube and then into a collecting duct. Within the secretory cell, a conspicuous series of actin rings, about 3 μm in diameter, follows the same path as the end-appa

Light and electron microscopy of the cornea in systemic mucopolysaccharidosis type I-S (Scheie's syndrome).

A 37-year-old man with coarse facies, stiff joints, corneal clouding, and normal intelligence sought medical attention. The diagnosis of a systemic mucopolysaccharidosis (MPS) type I-S (Scheie's syndrome) was confirmed by the presence of lysosomal alpha-L-iduronidase deficiency and excessive urinary dermatan and heparan sulfate excretion. The corneal button after perforating keratoplasty of the right eye demonstrated mucopolysaccharides consisting of numerous vacuoles containing fibrillogranular and partly membranebound material in epithelial cells, histiocytes, keratocytes, and extracellular

Early glomerular lesions in amyloidosis. Electronmicroscopic findings.

Our investigations on early glomerular changes in renal amyloidosis showed the following: 1. In some renal biopsies, amyloid was demonstrate … ormed from amyloid precursors brought via the blood stream. 4. Amyloid fibrils may be formed in the extracellular space of the glomerulus under the influence of lysosomal enzymes released from epithelial, mesangial and perhaps endothelial cells, by action of these enzymes on extracellularly deposited amyloid precursors.

Effects of 4-aminopyridine and tetraethylammonium chloride on the electrical activity and cable properties of canine tracheal smooth muscle.

The actions of 4-aminopyridine (4-AP) on passive membrane properties of canine tracheal smooth muscle were compared to those of tetraethylammonium chloride (TEA). Transmembrane potential was studied with intracellular electrodes in a partitioned bath by extracellular current injection. The cells were electrically quiescent and showed rectification to depolarizing pulses. TEA (30 mM) depolarized the muscle and increased membrane resistance, time constant (282-421 msec), space constant (2.16-2.67 mm), decreased rectification and generated action potentials. D600, with TEA, abolished the spikes b

Folded tissue geometry adds an electrodiffusive slowing term to cortical spreading depolarization.

Cortical spreading depolarization (CSD) propagates across cortex at millimeters per minute and can slow in sulcal regions, but the contribution of folded geometry remains unresolved. We tested whether sulcal dipole alignment adds a measurable electrodiffusive delay in a reduced surface model in which extracellular diffusion remains the dominant transport process. The model included overlapping extracellular and intracellular surface compartments, explicit K+, Na+, and Cl- concentrations, Goldman-Hodgkin-Katz membrane voltage, activation-dependent membrane sources, Na+/K+ pump dynamics, dynamic

Mitopherogenesis, a form of mitochondria-specific ectocytosis, regulates sperm mitochondrial quantity and fertility.

Mitochondrial export into the extracellular space is emerging as a fundamental cellular process implicated in diverse physiological activities. Although a few studies have shed light on the process of discarding damaged mitochondria, how mitochondria are exported and the functions of mitochondrial release remain largely unclear. Here we describe mitopherogenesis, a formerly unknown process that specifically secretes mitochondria through a unique extracellular vesicle termed a 'mitopher'. We observed that during sperm development in male Caenorhabditis elegans, healthy mitochondria are exported…

Potassium homeostasis and its disturbances in children.

Although only 2% of the body potassium is present in the extracellular space, its concentration is finely regulated by the internal balance, or distribution of potassium between the intracellular and extracellular compartments, and by the external balance, or difference between intake and output of potassium. Internal balance is modulated by a host of factors, including insulin, epinephrine, extracellular pH and plasma tonicity. Potassium output from the body is mainly determined by renal excretion. Renal secretion of potassium takes place predominantly in the principal cells of late distal an…

Periplasmic Protein Mobility for Extracellular Electron Transport in Shewanella oneidensis.

Extracellular electron transport (EET) supports the survival of specific microorganisms on the Earth's surface by facilitating microbial respiration with diverse electron acceptors. A key aspect of EET is the organization of electron relays, i.e., multi-heme c-type cytochromes (MHCs), within the periplasmic space of microbial cells. In this study, we investigated the mobility of periplasmic electron relays in Shewanella oneidensis MR-1, a model strain capable of EET, using in vivo protein crosslinking to the MHCs. First, we established that crosslinking efficiency correlates with the spatial p

Extracellular Vesicles and Their Role in Skin Inflammatory Diseases: From Pathogenesis to Therapy.

Extracellular vesicles (EVs), including exosomes, microvesicles, and apoptotic bodies, are released into the extracellular space by almost all known cell types. They facilitate communication between cells by transferring bioactive molecules, which impact both physiological processes and the development of diseases. EVs play a crucial role in the pathogenesis of various diseases by participating in multiple pathological processes. They contribute to disease progression by triggering cytokine release, modulating immune cell activity, and inducing inflammatory and immune responses. Beyond their p…

The secreted host-cell protein clusterin interacts with PmpD and promotes Chlamydia trachomatis infection.

Attachment and uptake into host cells are pivotal steps in the life cycle of the Chlamydiaceae, a family of obligate intracellular pathogens … cells, also binds to secreted clusterin (sCLU), a chaperone-like protein that is secreted into the extracellular space by the host cell, and forms part of the chaperone- and receptor-mediated extracellular protein degradation (CRED) pathway. Using in vitro assays, we demonstrate that sCLU interacts directly with soluble rPmpD. In infection experiments, depletion of …

Metabolic Glycoengineering of Cell-Derived Matrices and Cell Surfaces: A Combination of Key Principles and Step-by-Step Procedures.

Metabolic glycoengineering allows insertion of non-natural monosaccharides into glycan structures during biosynthesis thereby enabling extracellular matrices (ECMs), cell surfaces, or tissues for decoration with functional cues with ultimate spatial control while deploying aqueous and toxicologically benign coupling chemistries. In this work, we discuss relevant methods in the design of metabolic glycoengineered systems, ranging from synthetic procedures to decoration of cell surfaces and ECM components by bioorthogonal chemistries for widespread biomedical applications. Synaptic laminae in the visual system: molecular mechanisms forming layers of perception.

Synaptic connections between neurons form the basis for perception and behavior. Synapses are often clustered in space, forming stereotyped layers. In the retina and optic tectum, multiple such synaptic laminae are stacked on top of each other, giving rise to stratified neuropil regions in which each layer combines synapses responsive to a particular sensory feature. Recently, several cellular and molecular mechanisms that underlie the development of multilaminar arrays of synapses have been discovered. These mechanisms include neurite guidance and cell-cell recognition. Molecules of the Slit,

Lesions produced by the extravasation of urine from the upper urinary tract.

We present six cases which illustrate the spectrum of clinical features, macroscopic findings and light microscopic findings of urine extravasation from the upper urinary tract. The early lesions are characterized macroscopically by an oedematous, glistening or gelatinous appearance to the renal perihilar and peripelvic fat. Light microscopically there is lipolysis with associated foamy macrophages, multinucleate giant cells and lymphocytes. Immunohistochemical staining for Tamm-Horsfall protein is strongly positive in the extracellular space and in the foamy macrophages confirming urine extra…

Morphologic vascular anomalies detected during migraine surgery.

Although there is increasing evidence of migraine headaches having extracranial origins, the exact mechanisms underlying the pathogenesis of surgically treated migraines continue to be poorly investigated and described. We studied the microscopic and ultrastructural characteristics of superficial temporal artery (STA) and occipital (OA) artery in the auriculotemporal and great occipital trigger points of migraine patients to determine their possible role in migraine etiopathogenesis. Fifteen biopsies, 10 of STA and 5 from OAs, were collected intraoperatively during migraine surgery and immedia

Interstitial exclusion of albumin in rabbit lung during development of pulmonary oedema.

The modifications of the macromolecular sieving properties of the pulmonary extracellular tissue matrix were studied in adult anaesthetized rabbits (n = 10) exposed to increased tissue hydration. Exclusion of albumin from the perivascular pulmonary interstitial space was determined by using the continuous infusion method coupled with direct sampling of interstitial fluid performed through the wick technique. The rabbits underwent an intravenous infusion of saline amounting to 10 (n = 5) or 20 % (n = 5) body weight. Extracellular albumin distribution volume was derived from the steady state tis

Targeting the mitochondrial metabolite-dynamics-MDVs-MitoEVs axis: a new frontier in osteoarthritis management.

BACKGROUND: Osteoarthritis (OA) is characterized by progressive cartilage degradation, osteophyte formation, and synovitis. OA progression i … accelerate OA by transferring mitochondrial damage-associated molecular patterns (mtDAMPs) into the extracellular space through mitochondrial-derived extracellular vesicles (mitoEVs). Given the interplay of mitochondrial metabolites, mitochondrial dynamics, and MDVs/mitoEVs, we propose that the metabolite-dynamics-MDVs-mitoEVs axis represents a pivotal mechanism driv …

Phospholipase D1 is a critical mediator of neutrophil extracellular trap formation and venous thrombosis.

Neutrophil extracellular traps (NETs) are a host defense mechanism whereby activated neutrophils release decondensed chromatin and antimicrobial proteins into the extracellular space to trap and kill invading pathogens. While effective in clearing pathogens, NETs also pose pathological risks by exposing self-DNA, histones, granular enzymes, and reactive oxygen species (ROS), contributing to pathologies such as autoimmune diseases, inflammatory disorders, and thrombosis. Here, we identify phospholipase D1 (PLD1), a lipid-signaling enzyme that generates phosphatidic acid (PA), as a critical regu…

The enigmatic world of tear extracellular vesicles (EVs)-exploring their role in ocular health and beyond.

Extracellular vesicles (EVs) are released by all kind of cells into the extracellular space, where they shuttle parental cell-derived molecular cargoes (DNA, RNA, proteins) to both adjacent and distant cells, influencing the physiology of target cells. Their specific cargo content and abundance in liquid biopsies make them excellent candidates for biomarker studies. Indeed, EVs isolated from various body fluids, including blood, pleural fluid, urine, cerebrospinal fluid, saliva, milk, ascites, and tears, have been recognized for their potential as biomarkers in diagnosis, monitoring treatment,…
