Spinach nitrite reductase. Purification and properties of a siroheme-containing iron-sulfur enzyme.

Ferredoxin-nitrite reductase (EC 1.7.7.1.) from spinach has been purified to homogeneity with a specific activity of 110 units/mg of protein. The enzyme, Mr = 61,000 has 3 iron atoms (of which one is in siroheme) and 2 labile sulfides, i.e. 1 (Fe2-S2) per molecule, with absorption maxima at 276, 386 (Soret), 573 (alpha), and 690 nm, with an E386 of 3.97 X 10(4) M-1-cm-1, and A276/A386 absorptivity ratio of 1.8. Anaerobic addition of dithionite results in the loss of the 690 nm peak and the splitting of the 573 nm absorption band into two broad peaks at 545 and 585 nm. Reduction by dithionite i

Liquid Chromatography/tandem Mass Spectrometry-based Target Engagement of Lysine-specific Demethylase 1 Inhibitor TAK-418 Through Detecting Covalent Adducts in the Brain.

Translating pharmacokinetics (PK) and pharmacodynamics (PD) from preclinical to clinical is important in successful development of drugs for central nervous system (CNS) diseases. TAK-418 is a clinical candidate of lysine-specific demethylase 1 (LSD1) inhibitor with a unique mechanism of action; it covalently binds to flavin adenine dinucleotide (FAD) to form a compact adduct formylated FAD (F-FAD), thereby minimizing hematological side effect risks. In this study, we established a liquid chromatography-tandem mass spectrometry (LC/MS/MS)-based approach for evaluating F-FAD as a target engagem

Functional Pathways of the Gut Microbiome Associated with SCFA Profiles in Preclinical Alzheimer's Disease.

Functional activities of the gut microbiome, particularly those contributing to short-chain fatty acid (SCFA) metabolism, play a central role in host-microbe interactions and are linked to neuroinflammatory mechanisms underlying Alzheimer's disease (AD). How microbial metabolic functions relate to SCFA concentrations and cerebral amyloid-β (Aβ) burden during the preclinical stage of AD remains poorly understood. In this study, faecal metagenomes from 87 cognitively unimpaired adults were profiled using HUMAnN3 to generate MetaCyc pathway abundance data, normalised and filtered to retain pathwa

Lacticaseibacillus paracasei 36 attenuates D-GalN/LPS-induced acute liver injury in mice via suppressing the TLR4/NF-κB/MAPK pathway and NLRP3 inflammasome activation through modulating the intestinal microbiota.

INTRODUCTION: Acute liver injury (ALI) is a severe clinical syndrome with high mortality, often triggered by toxins, viruses, or immune-mediated injury. The gut-liver axis serves as a key mediator in the progression of hepatic disorders, and probiotics are considered viable treatment options owing to their impact on modulating intestinal microbiota, reducing inflammation, and alleviating oxidative stress. OBJECTIVES: This study was designed to elucidate the hepatoprotective mechanisms of L. paracasei 36 against D-GalN/LPS-induced ALI in mice, with a specific focus on its roles in inflammation

Auxin biosynthetic key gene CpYUC10B in embryo involved in large-fruited Chinese cherry massive abscission during the pit hardening process.

Chinese cherry [Cerasus pseudocerasus (Lindl.) G.Don] is an economically important Rosaceae fruit crop in China. Massive fruit abscission in the majority of large-fruited Chinese cherry landraces leads to a catastrophic yield loss, hindering the development of the Chinese cherry industry. Herein, we performed systematic physiological analyses to comprehensively characterize the dynamic changes in auxin-centered phytohormones during fruit development across four Chinese cherry landraces exhibiting varying degrees of fruit abscission. These analyses, along with exogenous indole-3-acetic acid (IA

Biochemical, Bioluminescence and Bioreporter Properties of Functionally Improved Firefly Luciferases.

Firefly luciferase (Fluc) is one the most widely used bioluminescent reporters for biological imaging and gene expression analysis, yet its limited stability and high assay cost remain major constraints limiting broader applications. Here, we engineered Fluc variants (N116H, M118L and N116H/M118L) with improved catalytic performance, thermostability, and soluble expression. The N116H variant exhibited the most pronounced enhancement, including a 4.6-fold increase in kcat, a 2.7-fold higher soluble expression yield, a 3.5-fold longer half-life at 37 °C, and 1.25-fold enhanced quantum yield rela

Photocatalytic Fe3O4@CDs Drives Nitrite-Independent extracellular respiration of anammox via efficient bidirectional electron transfer.

Anaerobic ammonium oxidation (Anammox) process relying on extracellular electron transfer (EET) overcomes nitrite scarcity in practical wastewater, but inefficient EET rate constrains nitrogen removal. Herein, Fe3O4 with carbon dot shell (Fe3O4@CDs) was self-assembled with anammox bacterial as a photocatalytic hybrid. Building on bacteria inward uptake of photogenerated electrons from CDs and outward transfer of respiratory electrons to Fe3O4, a bidirectional electron pathway was established firstly. This novel route achieved efficient nitrite-independent Anammox, enabling direct and rapid rem

Autofluorescence spectroscopy and multispectral autofluorescence microscopy for characterization of lupus nephritis in renal tissues.

The study investigates lupus nephritis in label-free mode using simultaneous autofluorescence spectroscopy and multispectral autofluorescence microscopy. Lupus nephritis (LN) is an autoimmune disease with proteinuria as the most typical symptom and indicator of renal damage and disease activity. This study includes the development and application of a micro-spectro-endoscope on the renal tissues of murine LN, covering three stages, advanced end-stage (proteinuric), early onset disease (antibody-positive), and healthy with no disease manifestations (young). The micro-spectro-endoscope exploits

Glucose Oxidoreductases for Glucose Monitoring: A Bird's Eye View of Glucose-Sensing Enzymes II.

Biosensors, utilizing glucose oxidoreductases as biological recognition elements (BREs), underpin current blood glucose monitoring (BGM) and continuous glucose monitoring (CGM) technologies. Glucose oxidase remains the gold standard due to its high selectivity and stability, yet its oxygen dependency limits it to first- and second-generation sensing. Fungal flavin adenine dinucletotide (FAD)-dependent glucose dehydrogenase (GDH) has emerged as the standard for mediator-based second-generation sensors due to its oxygen insensitivity, though it requires further engineering to match the stability

First Experience with Hypothermic Oxygenated Perfusion in Human Uteri: Feasibility and Metabolic Characterization.

Background: Uterus transplantation (UTx) is an emerging treatment for absolute uterine factor infertility. However, the use of deceased donors is limited, and donation after circulatory death (DCD) has not yet been utilized. Ischemic injury remains a major barrier, particularly compared with living donor procedures. Hypothermic oxygenated perfusion (HOPE), which has shown protective effects in heart, liver, and kidney transplantation, may offer similar benefits for uterine grafts. Methods: We report the first series applying HOPE to human uteri to improve preservation and enable metabolic inju

Untargeted metabolomics reveals organism specific biomarkers of carbapenem resistance in Klebsiella pneumoniae and Escherichia coli.

Antimicrobial resistance (AMR) is one of the greatest global concerns due to the increase in the rate of AMR infections and the lack of development of antimicrobial agents to combat AMR. The development of resistance to carbapenems among common pathogenic bacteria, including Klebsiella pneumoniae and Escherichia coli, is particularly concerning because carbapenems are relied upon for the treatment of Gram-negative infections. Metabolomics offers an approach to identify biomarkers associated with metabolic mechanisms of carbapenem resistance. We analyzed 512 annotated metabolites from a total o

Dietary choline deficiency potentiates Helicobacter pylori-driven gut-liver dysfunction via microbial metabolic rewiring.

BACKGROUND & AIMS: Approximately half of the global population is infected with Helicobacter pylori (H. pylori), yet the severity of clinical manifestations post-infection exhibits substantial heterogeneity, a disparity strongly correlated with host dietary and nutritional status. Choline deficiency, a widespread but frequently neglected condition, demonstrates poorly characterized mechanistic connections with H. pylori-induced pathological damage. METHODS: C57BL/6 J mice were subjected to choline-deficient diet (CDD) and H. pylori infection for 8- and 16-week intervals to model the associati

FMO5 Plays a Sex-specific Role in Goblet Cell Maturation and Mucus Barrier Formation.

BACKGROUND & AIMS: The intestine plays a key role in metabolism and nutrient and water absorption and provides both physical and immunological defense against dietary and luminal antigens. The protective mucosal lining in the intestine is a critical component of the intestinal barrier that, when compromised, can lead to increased permeability, a defining characteristic of inflammatory bowel disease, among other intestinal diseases. Here, we define a new role for the flavin-containing monooxygenase (FMO) family of enzymes in maintaining a healthy intestinal epithelium. METHODS: Using Caenorhab

Validation of mitochondrial FMN as a predictor for early allograft dysfunction and patient survival measured during hypothermic oxygenated perfusion.

Hypothermic oxygenated machine perfusion (HOPE) preconditions liver grafts before transplantation. While beneficial effects on patient outcomes were demonstrated, biomarkers for viability assessment during HOPE are scarce and lack validation. This study aims to validate the predictive potential of perfusate flavin mononucleotide (FMN) during HOPE to enable the implementation of FMN-based assessment into clinical routine and to identify safe organ acceptance thresholds. FMN was measured in perfusate samples of 50 liver grafts at multiple time points. After transplantation, patients were followe
