Novel approach to enhance antifungal properties via formation of wavy and Sharp microstructures composed of metal oxide nanoparticles and Cdots.

In this work we have synthesized MgO and ZnO nanoparticles (NPs) and carbon nanodots using green routes. We have used the crystal violet bioassay method to evaluate the antifungal properties of MgO, ZnO NPs, and Cdots against Candida albicans on 2, 5-dimercapto-1, 3, 4-thiadiazole coated microtiter plates. We have examined the intricate mechanisms that govern their efficacy, and have provided insights into their potential therapeutic applications against microbial infections. This study elucidates the structural and functional properties of inorganic metal oxide NPs as well as organic carbon n

CUMS alters behavioral phenotypes and gut mycobiota in mice.

OBJECTIVE: To investigate the effects of chronic unpredictable mild stress (CUMS) on the gut fungal community in mice and its association with anxiety-like behaviors. METHODS: C57BL/6 male mice were randomly divided into a control group and a CUMS group, with 10 mice per group. The CUMS group was exposed to a 3-week regimen of chronic stressors (damp bedding, food and water deprivation, continuous light exposure, tail pinch, cage tilting, restraint, and forced swimming). Anxiety-like behaviors were assessed using the elevated plus maze (EPM) and open field test (OFT). Colonic contents were co

Hexacyclic 6/6/6/5/6/6 Spirosteroids Produced by a Marine-Derived Fungus Galactomyces pseudocandidus and Antifungal Activity against Candida Species.

Two spirosteroids, epoformsterols A and B (1 and 2), featuring a highly fused 6/6/6/5/6/6 hexacyclic framework, were isolated from the marine-derived fungus Galactomyces pseudocandidus. The structures of 1 and 2 were elucidated by NMR, HRMS, and single-crystal X-ray diffraction. The absolute configuration of 1 was established by comparison of calculated and experimental ECD spectra, and that of 2 was assigned by ECD comparison with 1. Compounds 1 and 2 exhibited inhibitory activity against Candida auris and C. albicans with MIC values of 3.12-12.5 μg/mL, and MFC/MIC ratios suggested a fungista

Prochelators modulate azole activity against Candida albicans in a metal-dependent manner.

Candida albicans is an opportunstic fungal pathogen of growing clinical concern, in part due to antifungal drug tolerance. Here we report a thiol-activated prochelation strategy that modulates the activity of azole antifungals against C. albicans in a manner influenced by the degree of azole stress as well as the metal composition of the growth medium. We identify two disulfide-linked prochelators containing aroylhydrazone (AH1-S)2 or thiosemicarbazone (IT1-S)2 structures that impact C. albicans growth in both a standard laboratory strain and a fluconazole-resistant clinical isolate. A combina

Structural basis for transport and inhibition of nucleotide sugar transport in pathogenic fungi.

GDP-Mannose transporters are Golgi-localised solute carriers that are essential for the virulence of pathogenic fungi, serving as critical components of fungal glycosylation pathways. However, the mechanism by which nucleotide sugars are recognised and transported across the Golgi membrane remains unclear, hindering efforts to develop effective inhibitors that could serve as distinct antifungal agents. Here, we present cryo-EM structures of the GDP-Mannose transporter, Vrg4, from Candida albicans in complex with nanobodies in both the cytoplasmic and Golgi-facing states. Structural comparisons

Gastrointestinal bleeding due to disseminated histoplasmosis in a patient under anti-TNF treatment.

A 62-year-old woman, originally from Peru, with rheumatoid arthritis under treatment with anti-tumor necrosis factor (anti-TNF) therapy, was … hickened and necrotic-looking surrounding mucosal edges. Histology showed granulomas and yeast-like fungal structures with methenamine silver staining in both tissues, consistent with disseminated histoplasmosis. Antifungal treatment was initiated with good clinical evolution.

G-quadruplexes as antimicrobial targets.

Escalating antibiotic resistance and poor options for many parasitic infections, including neglected tropical diseases, demand new antimicrobial strategies. G-quadruplexes (G4s), which are noncanonical nucleic acid structures, are emerging targets in microbial genomes. We summarize evidence for G4 formation in bacteria and parasites, which is provided by phenotypic and chemical-biology datasets, whereas evidence in support of fungal G4 targeting remains nascent, with fewer studies and limited mechanistic validation. After integrating genomics and bioinformatics data, we argue that microbial G4

Elucidating microbial succession dynamics and flavor metabolite formation in korean style spicy cabbage fermentation: Integration of flavoromics, amplicon sequencing, and metagenomics.

Korean style spicy cabbage (KSC) is a prominent fermented vegetable consumed globally. Nevertheless, microbial succession dynamics, interactions, and flavor-core microbiome correlations lack comprehensive understanding. Metabolomics revealed eight taste-active compounds and sixteen aroma-active compounds as key flavor determinants throughout fermentation. Amplicon sequencing elucidated dynamic shifts in bacterial and fungal community structures during KSC fermentation, with subsequent analyses identifying free sugars as the primary drivers of microbial succession. Spearman correlation analysis

Fungal Keratitis Caused by Scedosporium apiospermum in an Immunocompetent Patient: How Challenging Can Treatment get?

This case report underscores the complexities in managing fungal keratitis caused by Scedosporium apiospermum, even in an immunocompetent patient. The pathogen's resistance to many antifungal agents, its ability to invade deep ocular structures, and the presence of posterior chamber involvement added to the treatment challenges. Effective management requires not only timely diagnosis and targeted antifungal therapy, such as voriconazole, but also careful consideration of adjunctive treatments like corticosteroids and surgical interventions to address inflammation and posterior chamber involvem

Systemic mucormycosis caused by Rhizopus microsporus in a captive bottlenose dolphin.

A 6-year-old female bottlenose dolphin (Tursiops truncatus) kept in dolphinarium died after a 3.5-month period of lethargy and inappetence despite antibiotics and supportive care. At necropsy, gross findings included diffuse varying-sized nodules in the lungs and scattered nodules throughout the heart, spleen, mesenteric and hilar lymph node and kidney. Microscopically, the lesions were characterised by disseminated fungal pyogranulomas with numerous intralesional Mucor-like fungi. The fungi structures were demonstrated by Periodic acid-Schiff and Gomori methenamine silver stain. Molecular ana

Idiopathic fibrosing mediastinitis.

Fibrosing mediastinitis is rare in settings where histoplasmosis is not endemic. An idiopathic form of the disease may present with indistinguishable features and requires methodical exclusion of competing differential diagnoses. We report the case of a 30-year old female patient who presented with intermittent haemoptysis for the past 2 years with no constitutional symptoms. Computed tomography of the chest revealed a prominent right bronchial arterial circulation with a mass-like lesion, which encased and attenuated the right pulmonary trunk and adjacent structures. Cytologic, histologic, microbiologic, and electron microscopic characterization of a canine Prototheca wickerhamii infection.

An adult dog was presented for chronic cough and a recent development of ulcerated, erythematous nares with nasal discharge. Cytology of enlarged peripheral lymph nodes revealed many intracellular and extracellular organisms. These round or rarely oval organisms measured approximately 5-9 µm in diameter and frequently contained several globular structures, ranging from deeply basophilic to magenta. A thin, clear halo was present. Smaller 1-2 µm, magenta forms were also observed. Fungal culture yielded small, wet, raised, irregularly shaped, white to pale tan colonies. Microbiologic staining of

Extracellular vesicles released by Candida albicans yeast cells impact candidiasis lethality in insect and murine models.

Extracellular vesicles (EVs) are nano-sized lipid bilayer structures released by diverse cell types. For pathogenic fungi, EVs play critical roles by modulating immune responses and influencing disease progression. Previously, we demonstrated that vaccination with EVs from Candida albicans promoted protection in immunosuppressed mice subsequently infected with lethal doses of this organism. However, the impact of EVs in the presence of fungi is poorly investigated in vivo. Here, we demonstrated that co-injection of C. albicans yeasts and their EVs into Galleria mellonella larvae led to a rapid
