No genetic effect of alpha1-antichymotrypsin in Alzheimer disease.

Alzheimer disease (AD) is the most common neurodegenerative disorder for individuals over the age of 40. AD has a complex etiology, and it is likely that multiple genes, acting independently and/or interacting, affect the risk of developing AD. Several genes involved with AD have been described already, but only the APOE gene on chromosome 19q has been shown to affect the risk of the common late onset form of AD. alpha1-Antichymotrypsin (AACT) is a major component of the amyloid plaques found in the brains of AD patients, and an allele in its gene has been proposed to increase the risk of deve

Progress of unrelated bone marrow donor search at the University Hospital of Essen (1991-1994).

The development of unrelated bone marrow donor search (UMDS) was analyzed in a retrospective single-center study covering the period from 1991 to 1994. The data files of the 285 enrolled UMDS patients were evaluated for the patients' underlying disease, HLA phenotype frequency, year of search start, UMDS duration/outcome, and the origin of identified donors. Within 3 months after UMDS start, a donor was found in 5%, 10%, 19%, and 49% of the searches started in 1991 (n = 57), 1992 (n = 73), 1993 (n = 77), and 1994 (n = 78, until September), respectively. The total UMDS success rate increased fr

The spread of X-linked shifting gene responsible for G-6-PD deficit in series of 13,710 subjects from 4 districts of Moldavia.

Between 1985 and 1989, a sample of 13,710 subjects (5,825 males and 7,885 females) from the Vrancea, Bacău, Neamţ and Suceava districts were examined for the presence of this pathological gene, 32 male hemizygotes and 13 female heterozygotes and homozygotes being detected. The incidence of G-6-PD deficit in the hemizygotes was of 0.55%. The investigations carried out in the shifting gene carriers' families revealed the way deficit is transmitted to offsprings. Hemizygotes get it from their mother while the female homozygotes either from their mother or father. The frequency of G-6-PD deficit o

Secretor status as a genetic marker in duodenal ulcer--a comparative study.

Idiopathic hypogonadotropic hypogonadism (IHH) is a well-known disorder apart from its pathogenesis, which is still mostly unclear, even though a diverse subgroup of patients with hypogonadotropic hypogonadism and hyposmia/anosmia--the Kallman syndrome--have been partly linked to a mutated gene, known as kal-1 gene. In this study, we aimed to evaluate the sellar region of patients with IHH on magnetic resonance (MR). Pituitary MR of 120 male patients with IHH, diagnosed by a thorough endocrinologic assessment, were compared with pituitary MR of 49 healthy cases selected randomly who underwent

HLA antigen frequencies in HIV-1-related Kaposi's sarcoma.

In an ongoing study of genetic factors that might contribute to disease outcome in HIV-1-infected individuals, HLA antigen (A, B, C, DR and DQ) frequencies in 44 patients with Kaposi's sarcoma (KS) and/or 14 with KS and opportunistic infection (OI) were compared with the frequencies found in 83 HIV-1-seropositive (disease-free) and 87 seronegative homosexual men, or with 50 patients with OI. KS patients had higher frequencies of HLA-B35, -C4, -DR1, and -DQ1 and lower frequencies of HLA-C5 and -DR3 compared to the total control population (N = 170) and to the population at risk (HIV-1-seroposit

Association of class II antigens of HLA with primary glomerulopathies.

HLA antigens of Japanese patients with primary glomerulopathies were determined, and the frequency of each HLA antigen was compared with that of Japanese normal controls. IgA nephropathy with HLA-DR4, idiopathic membraneous glomerulopathy with DR2, lipoid nephrosis with DRw53 and poststreptococcal glomerulonephritis with DR1 were definitely or probably associated. It was suggested that primary glomerulopathies might all be associated with HLA class II and not class I antigens. Japanese patients with HLA-Bw48 and -DRw8 were less susceptible to primary glomerulopathies.

Inherited haemoglobin variants in a South African population.

We present the findings of a survey to determine the prevalence of inherited haemoglobin disorders in the Coloured (mixed ethnic origin) population of South Africa. A variety of haemoglobins was found. Of the structural variants, Hb E and Hb S were the most common, the former probably originating from South-East Asia and the latter from East Africa and possibly Madagascar. The alpha+ (-alpha) thalassaemia haplotype is particularly common with an observed frequency of 0.023. Beta thalassaemia was rather less common, while hereditary persistence of fetal haemoglobin was found for the first time

Linkage of albino and pink-eyed dilution genes in the Mongolian gerbil and other rodents.

The albino (ch) and pink-eyed dilution (p) genes are shown to be linked with a crossover value of 30 +/- 5 percent. The linkage is comparable with similar linkages of the albino and pink-eyed dilution loci in other rodent species. No evidence of linkage was found between nonagouti (a) with either c or p.

Hereditary red cell disorders in Southeast Asian refugees and the effect on the prevalence of thalassemia disorders in the United States.

This investigation established the frequency and prevalence of hemoglobin E, alpha and beta thalassemia, and glucose-6-phosphate dehydrogenase deficiency in a group of Southeast Asian individuals and determined the impact upon the prevalence of thalassemia disorders in the United States. The resettlement program of Southeast Asian refugees has significantly increased the total number of serious thalassemic disorders in the United States. For several generations Southeast Asian children will be the principal thalassemic group.

Are congenital vertebral anomalies and spina bifida cystica aetiologically related?

Radiological records of 104 patients with multiple vertebral anomalies without apparent spina bifida and 112 infants with spina bifida cystica were surveyed and the incidences of hemivertebrae and of rib, vertebral body, and vertebral arch fusions were recorded. The distributions of these four anomalies along the vertebral axis were found not to be statistically different between the two age groups. This is additional evidence to support the hypothesis that multiple vertebral anomalies and anencephaly-spina bifida cystica are aetiologically related.

Shortfall of exome analysis for diagnosis of Shwachman-Diamond syndrome: Mismapping due to the pseudogene SBDSP1.

Shwachman-Diamond syndrome characterized by metaphyseal dysplasia, pancreatic insufficiency, and pancytopenia is caused by biallelic mutations in SBDS. Gene conversion between SBDS and its pseudogene SBDSP1 is the major cause. Here, we report two unrelated patients with Shwachman-Diamond syndrome who were shown to be compound heterozygotes for relatively frequent pathogenic alleles (the 258+2T>C allele and another allele composed of 183-184TA>CT and 201A>G) using an established polymerase chain reaction sequencing assay with SBDS-specific primers. Exome analysis of the patients showed discrepa

Analysis of Polymorphism rs1042522 in TP53 Gene in the Mothers of Twins and of Singletons: A Population-Based Study in Rio Grande do Sul, Brazil.

Although the birth of twins has always attracted attention, there are no known genetic or environmental factors that can determine the birth of monozygotic (MZ) twins. And even for dizygotic (DZ) twins, genetic influences are not completely understood. A previous study from our group has shown that the C allele of polymorphism rs1042522 in the TP53 gene was more frequent in the mothers of twins than in the mothers of singletons in a small village in South Brazil. In order to clarify whether this was an isolated factor, we performed a population-based, observational case-control study. Samples

Gene-Environment Correlation Between the Dopamine Transporter Gene (DAT1) Polymorphism and Childhood Experiences of Abuse.

In the present study, we investigated the possible gene-environment correlation between the dopamine transporter gene (DAT1) polymorphism and childhood experiences of abuse and neglect. Genetic information was obtained from 1,442 male and 2,178 female twins and their siblings drawn from a Finnish population-based sample. The Childhood Trauma Questionnaire was used to measure the childhood experiences of abuse and neglect. In men, the DAT1 polymorphism was associated with having experienced sexual abuse in childhood, such that men with the 9R9R genotype reported less sexual abuse experiences th

A versatile omnibus test for detecting mean and variance heterogeneity.

Recent research has revealed loci that display variance heterogeneity through various means such as biological disruption, linkage disequilibrium (LD), gene-by-gene (G × G), or gene-by-environment interaction. We propose a versatile likelihood ratio test that allows joint testing for mean and variance heterogeneity (LRT(MV)) or either effect alone (LRT(M) or LRT(V)) in the presence of covariates. Using extensive simulations for our method and others, we found that all parametric tests were sensitive to nonnormality regardless of any trait transformations. Coupling our test with the parametric

[Genetic conditioned changes in activity of 5,10-methylenetetrahydrofolate reductase (MTHFR) and recurrent miscarriages].

5,10-methylenetetrahydrofolate reductase (MTHFR) is the key enzyme in folate, methionine and homocysteine metabolism. The disturbances in MTHFR activity could be the cause of increased serum level of homocysteine. Hyperhomocysteinemia is a risk factor of changes in coagulation cascade through direct cytotoxic influence on endothelium, atherogenesis, activation of coagulation factor V and VII, increased level of thrombin and platelets aggregation. Genetic disturbances in MTHFR enzyme activity in the presence of polymorphic variants of its gene are responsible for homocysteine augmentation and c

CDKN2A mutations and melanoma risk in the Icelandic population.

BACKGROUND: Germline CDKN2A mutations have been observed in 20-40% of high risk, melanoma prone families; however, little is known about their prevalence in population based series of melanoma cases and controls. METHODS: We resequenced the CDKN2A gene, including the p14ARF variant and promoter regions, in approximately 703 registry ascertained melanoma cases and 691 population based controls from Iceland, a country in which the incidence of melanoma has increased rapidly. RESULTS: We identified a novel germline variant, G89D, that was strongly associated with increased melanoma risk and app

Extended evaluation of serotonin transporter gene functional polymorphisms in subjects with post-stroke depression.

OBJECTIVE: As an extension of our previous observation, relating a serotonin transporter gene-linked promoter region (5-HTTLPR) diallelic functional polymorphism (short [S] and long [L] alleles) to the risk of post-stroke major depression (PSD), this study investigated the role of 2 other functional polymorphisms of the serotonin transporter gene (5-HTT) in the same sample of subjects with PSD. METHOD: In a clinical sample of 26 patients with PSD and 25 unrelated nondepressed stroke patients of Caucasian descent, we examined the frequencies of a functional single nucleotide variant (A/G) with

Beta-globin gene cluster haplotypes and alpha-thalassemia in sickle cell disease patients from Trinidad.

In this study, we have determined the frequency of beta(S) haplotypes in 163 sickle cell disease patients from Trinidad. The alpha(3.7) globin gene deletion status was also studied with an observed gene frequency of 0.17. Among the 283 beta(S) chromosomes analyzed, the Benin haplotype was the most prevalent (61.8%) followed by Bantu (17.3%), Senegal (8.5%), Cameroon (3.5%), and Arab-Indian (3.2%), while 5.7% of them were atypical. This beta(S) haplotypes distribution differed from those previously described in other Caribbean islands (Jamaica, Guadeloupe, and Cuba), in agreement with the known…
