Melatonin refines ovarian mitochondrial dysfunction in PCOS by regulating the circadian rhythm gene Clock.

Mitochondrial dysfunction is present in the ovaries of patients with polycystic ovary syndrome (PCOS). Melatonin (MT) has shown promise in treating PCOS by improving mitochondrial dysfunction, though the underlying mechanisms remain unclear. In this study, we first assessed the levels of proteins associated with mitochondrial autophagy and dynamics in ovary granulosa cells (GCs) of PCOS patients and in the ovaries of DHEA-induced PCOS mice. We found abnormal expression of these proteins, indicating the presence of mitochondrial dysfunction in PCOS ovaries. Notably, the expression of the circad

Gene expression regulation and polyadenylation in ulcerative colitis via long-chain RNA sequencing.

BACKGROUND: Ulcerative colitis (UC) is an immune-mediated chronic intestinal disease, with a pathogenesis that remains incompletely understood. The purpose of this study is to analyze the difference of gene expression between UC patients and healthy controls using Oxford Nanopore Technology's long-read RNA sequencing (ONT-RNA-seq) and to explore how alternative polyadenylation (APA) site selection contributes to UC pathogenesis. METHODS: Colon tissue samples from UC and normal controls (NC) were collected, and total RNA was extracted and sequenced using ONT-RNA-seq technology. Various bioinfo

Transcriptomic analysis of Streptococcus pneumoniae serotype 1 reveals serotype-specific gene regulation.

Streptococcus pneumoniae (Sp) is an opportunistic pathogen that colonizes the mucosal surfaces of the human upper respiratory tract. While transcriptomic studies of Sp have become more common, most have focused on laboratory-adapted strains such as D39 or TIGR4. These strains, though widely used in research, may not fully capture the biology of clinical isolates, particularly the hypervirulent serotype 1 (S1). S1 is clinically significant due to its association with invasive disease, epidemic outbreaks and a distinct global distribution, particularly in regions with a high pneumococcal disease

Germline Telomere-Regulating Gene Variants Are Not Associated with Thyroid Cancer in a Saudi Arabian Population.

Background: Germline variants in the telomere-regulating genes (TRG) of the shelterin complex have recently been associated with long telomere length and increased risk of several tumors, including thyroid cancer (TC). We aimed to validate these findings in a different ancestral population. Patients and Methods: We conducted a cross-sectional study in a tertiary care hospital in Saudi Arabia. We included 189 patients with sporadic TC and 63 members of 26 families with at least two members with TC. Whole exome sequencing was performed on genomic DNA isolated from peripheral blood, and bioinform

Preliminary Findings on Post-COVID Cardiovascular Risk: Transcriptional Regulation and Gene Expression Patterns.

Survivors of COVID-19 are facing a new vulnerability of encountering post-COVID complications, including cardiovascular diseases (CVDs). Despite the increasing prevalence of CVDs, post-COVID-induced cardiac abnormalities significantly escalate mortality and morbidity. However, some recent studies have begun to elucidate complex pathological signaling pathways in these conditions, further investigation is required to distinguish key genetic and proteomic regulators in post-COVID states precisely. Gene Expression Omnibus (GEO) databases were used to investigate gene expression profiles in post-C

NFATc3 and PML synergistically regulate tumor-associated gene expression in a SUMOylation-Independent manner.

The nuclear factor of activated T cells 3 (NFATc3) plays a significant role in various cancer-related processes, but its interactions with transcriptional modulators, particularly Promyelocytic Leukemia protein (PML), remain poorly understood. PML, a nuclear scaffold protein, is involved in tumor suppression and transcriptional regulation. This study investigates the interaction between NFATc3 and PML, focusing on the role of SUMOylation and its impact on downstream target genes. In vitro experiments, including mass spectrometry and Co-immunoprecipitation (Co-IP), were conducted to explore thi

PIWIL4 regulates gene expression and piRNA levels in RSV-infected airway epithelial cells.

Respiratory syncytial virus (RSV) is a leading cause of acute lower respiratory tract infections with significant morbidity and mortality in young children, the elderly and immunocompromised hosts. Despite its clinical burden, no effective RSV vaccine or therapy exists for infants, only prophylactic treatment. Small non-coding RNAs have emerged as important regulators of host-pathogen interactions. PIWI-interacting RNAs (piRNAs) are a distinct class of small non-coding RNAs known for maintaining the genome complexity and integrity in gonadal cells. However, there is growing evidence of their r

Pirinixic Acid Protects Against Hemorrhagic Shock by Regulating Neutrophil-Associated Key Genes.

BACKGROUND: The uncontrolled inflammatory cascade triggered by hemorrhagic shock (HS) can exacerbate tissue damage and organ dysfunction. Neutrophils, as the most abundant type of leukocytes, are key mediators of the innate immune response in the early stages of injury. There is a lack of suitable treatments targeting neutrophils in HS. METHODS: We used machine learning algorithms to identify neutrophil-associated key genes in patients with HS and assessed the diagnostic efficacy of these genes. Weighted gene co-expression network analysis and consensus clustering analysis were performed base

Quorum sensing regulates a polysaccharide biosynthesis gene to control cell aggregation in Paracoccus denitrificans.

Most biofilm research has centered on a few motile model organisms that form thick biofilms. During dispersal, motile bacteria actively escape through motility; however, nonmotile species likely rely on the modulation of adhesive interactions to regulate detachment. In this study we show that the nonmotile soil organism Paracoccus denitrificans maintains thin biofilms through constitutive quorum sensing (QS). We show that a LuxI/LuxR-type QS system actively suppresses cell-to-cell adhesion to maintain a thin biofilm architecture, while maintaining a basal level of cell-surface adhesion. We ide

Regulation of testosterone synthesis by circadian clock genes and its research progress in male diseases.

The circadian clock is an important internal time regulatory system for a range of physiological and behavioral rhythms within living organisms. Testosterone, as one of the most critical sex hormones, is essential for the development of the reproductive system, maintenance of reproductive function, and the overall health of males. The secretion of testosterone in mammals is characterized by distinct circadian rhythms and is closely associated with the regulation of circadian clock genes. Here we review the central and peripheral regulatory mechanisms underlying the influence of circadian clock

Phosphoglucomutase 5 gene transcripts are expressed by the human placenta and differentially regulated in placental dysfunction.

The placenta plays an essential role facilitating nutrient, gas and waste exchange between the maternal and fetal systems for optimal fetal growth. When placental development is impaired and the placenta dysfunctional, serious pregnancy complications such as fetal growth restriction and preeclampsia may arise. Previously, phosphoglucomutase-5 (PGM5) transcripts were found to be highly elevated in the blood of patients whose pregnancies were complicated by fetal growth restriction and preeclampsia. As both conditions feature placental insufficiency, here we aimed to characterise PGM5 levels in

Regulation of reactive oxygen species and the role of mitochondrial apoptotic-related genes in rheumatoid arthritis.

Previous research suggests mitochondrial apoptosis alleviates rheumatoid arthritis (RA), but the role of mitochondrial apoptosis-related genes (MARGs) is unclear. Urgent exploration of RA-related mitochondrial apoptosis biomarkers is needed. Gene Expression Ontology (GEO)-derived RA datasets were used to identify differentially expressed genes (DEGs) compared to normal controls, intersected with MARGs to obtain differentially expressed mitochondrial apoptosis-related genes (DE-MARGs). Three ML algorithms screened diagnostic biomarkers. A nomogram was built and validated by receiver operating c

An ISWI-related chromatin remodeller regulates stage-specific gene expression in Toxoplasma gondii.

ATP-dependent chromatin remodellers are specialized multiprotein machines that organize the genome in eukaryotic cells and regulate its accessibility by repositioning, ejecting or modifying nucleosomes. However, their role in Toxoplasma gondii is poorly understood. Here we show that T. gondii has evolved two divergent proteins within the imitation switch (ISWI) family: TgSNF2h and TgSNF2L. TgSNF2h specifically forms a core complex with the transcription factor AP2VIII-2 and the scaffold protein TgRFTS. Depletion of TgRFTS phenocopies the knockdown of TgSNF2h, restricting access to chromatin an

The characterization and validation of regulated cell death-related genes in chronic rhinosinusitis with nasal polyps.

BACKGROUND: Regulated cell death (RCD), a genetically controlled process mediated by specialized molecular pathways (commonly termed programmed cell death), plays pivotal roles in diverse pathophysiological processes. However, the landscape and functional implications of RCD subtypes in chronic rhinosinusitis with nasal polyps (CRSwNP) remain poorly characterized. This study aimed to systematically investigate the involvement of RCD mechanisms in the pathogenesis and progression of CRSwNP. METHODS: Transcriptomic datasets (GSE136825, GSE23552, GSE198950, GSE196169, GSE156285) related to CRSwN

rBCG-LTAK63 enhances protection against tuberculosis by inducing autophagy and circadian gene regulation.

Tuberculosis (TB) remains a global public health challenge, with the current BCG vaccine providing limited efficacy in adults, and available treatments being lengthy and debilitating. To overcome these challenges, we have previously developed a recombinant BCG strain expressing the detoxified E. coli Heat-Labile Toxin (LTAK63), providing increased protection in mouse models and reduced lung pathology. Here, using systems biology and RNA sequencing of lung tissues in a murine model, we uncover the molecular mechanisms underlying rBCG-LTAK63's increased protection. Immunization triggered early a

Gene regulation by mating depends on time, diet, and body region in female Aedes aegypti.

Aedes aegypti is a major vector of several arboviruses that cause human mortality and morbidity. One method for controlling the spread of these viruses is to control mosquito reproduction. During mating, seminal fluid molecules and sperm are transferred and these stimuli influence female post-mating physiology and behavior. Yet, little is known about the mechanisms underlying these post-mating responses. To fill this gap, short-read RNA sequencing was used to identify differentially expressed genes between unmated (control) and mated females in the head/thorax (HT), abdomen (Ab) and the lower
