Precise control of synaptic vesicle luminal pH offers a way to analyze proton-driven mechanisms involved in neurotransmitter loading, vesicle maturation, and synaptic transmission. Current genetically encoded tools either monitor vesicular pH or optogenetically acidify vesicles; here, we address this gap with a light-driven alkalizing actuator. Here, we developed syp-NsXeR, a genetically encoded optogenetic actuator based on the inward proton pump xenorhodopsin NsXeR, targeted to acidic presynaptic compartments through synaptophysin. Confocal imaging demonstrated enrichment of the construct at

The intervertebral disc is an avascular structure composed of the nucleus pulposus, annulus fibrosus, and the superior and inferior cartilage endplates. Located between adjacent vertebrae, it connects them structurally, contributing to spinal stability, load-bearing, shock absorption, and protection of the spinal cord. Intervertebral disc degeneration (IDD) results from various factors, including mechanical overload, trauma, repetitive strain, pregnancy, genetic predisposition, and developmental abnormalities. In the context of increased occupational demands and lifestyle changes, the spine is

Vaccinology originated with 18th century efforts to prevent smallpox by injecting healthy individuals with the infectious contents of cutaneous lesions from smallpox patients (variolation) or from individuals afflicted with cowpox (vaccination). In the late 19th century, vaccination was extended to other diseases after the development of chemical methods to kill pathogens (inactivation) and cell-culture passaging to decrease their virulence (attenuation). Since 1970, advances in immunology, cell subunit purification, and recombinant-DNA genetic engineering have enabled antigen-specific vaccine

Substantial evidence suggests a role for immunotherapy in treating Alzheimer's disease (AD). While the precise pathophysiology of AD is incompletely understood, clinical trials of antibodies targeting aggregated forms of β amyloid (Aβ) have shown that reducing amyloid plaques can mitigate cognitive decline in patients with early-stage AD. Here, we describe what we believe to be a novel approach to target and degrade amyloid plaques by genetically engineering macrophages to express an Aβ-targeting chimeric antigen receptor (CAR-Ms). When injected intrahippocampally, first-generation CAR-Ms have

Non-small-cell lung cancer (NSCLC) afflicts about 2 million people worldwide, with both genetic (familial) and environmental factors contributing to its development and spread. The inadequacy of currently available therapeutic techniques, such as surgery, chemotherapy, and radiation therapy, in addressing NSCLC is reflected in the very low survival rate of this disease. Therefore, newer approaches and combination therapy regimens are required to reverse this dismal scenario. Direct administration of inhalable nanotherapeutic agents to the cancer sites can potentially lead to optimal drug use,

Aging is the greatest risk factor for late-onset Alzheimer's disease (LOAD), which accounts for >95% of Alzheimer's disease (AD) cases. The mechanism underlying the aging-related susceptibility to LOAD is unknown. Cellular senescence, a state of permanent cell growth arrest, is believed to contribute importantly to aging and aging-related diseases, including AD. Senescent astrocytes, microglia, endothelial cells, and neurons have been detected in the brain of AD patients and AD animal models. Removing senescent cells genetically or pharmacologically ameliorates β-amyloid (Aβ) peptide and tau-p

In this study, we compared genetically resistant C57BL/6 and susceptible A/J mice for their resistance to Listeria monocytogenes infection during pregnancy. Intragastric infection with modest numbers of bacterial cells (10(5) CFU) caused reproducible fetal infection and abortion in both mouse strains. Bioluminescence imaging demonstrated dissemination of L. monocytogenes cells from maternal to fetal organs within 3 days of intragastric infection. Although non-pregnant C57BL/6 mice were significantly more resistant to infection than non-pregnant A/J mice, C57BL/6 and A/J mice had similar microb

The genetically obese-hyperglycemic mouse, C57 BL/6J-ob, exhibits hyperglycemia and hyperinsulinemia. We have investigated the in vivo hepatic response to a glucose load in female obese mice and their lean littermates. Within 15 min after the administration of glucose (1.5 g/kg) to fasted lean mice, gluconeogenesis from [14C]alanine markedly decreased, endogenous hepatic levels of alanine and other gluconeogenic precursors increased, and glycogen synthetase was activated by virtue of an increase in the precent of synthetase I. These changes persisted up to 60 min and then returned to fasting v

In this review, we describe and discuss the genetic factors that, up to some point, determine resistance to the infection and control the progression of the disease in HIV-infected individuals. Genetic factors may account for non-progression or slow progression of the disease in some of so called long-term non progressors HIV-infected individuals. In general, this group shows no symptoms for more than 10 years, while their circulating T CD4+ cells levels remain stable and they usually have a low virus load. Even though non-progression and rapid progression phenomenon are still not fully unders

The ocular dimensions in patients suffering from primary angle-closure glaucoma (a.c.g.) have been studied in several clinical series, chiefly in Caucasians. The epidemiology and aetiology of a.c.g. are less well known although genetic factors seem to be involved. Eskimos have recently been shown to constitute a high risk population with respect to a.c.g. Consequently, a series of oculometric, epidemiologic, and genetic studies among Greenland Eskimos was undertaken. Besides the immediate purpose, prevention of blindness in this population, the survey had important general aspects and the foll

INTRODUCTION: Preimplantation genetic testing (PGT) reduces reproductive risks for genetically high-risk families. To date, simultaneous PGT for nuclear and mitochondrial gene variants has rarely been reported. PGT targeting mitochondrial DNA (mtDNA) variants remains challenging and requires further clinical evidence.
CASE REPORT: We herein report a 3-year-old girl with severe global developmental delay. The proband harbored compound heterozygous RNASEH2C variants (maternal c.194G > A and paternal c.433C > T) and a maternally inherited m.3250 T > C variant in MT-TL1 with 72.9% heteroplasmy. P

Pseudorabies (PR) is an infectious disease caused by the pseudorabies virus (PRV), affecting various domesticated and wild animals. Since pigs are the only natural hosts of PRV, PR poses a serious threat to the pig farming industry. Currently, PR is primarily prevented through vaccination with inactivated vaccines or genetically modified attenuated live vaccines. Developing safe and effective genetically engineered vaccines would facilitate the eradication and control of PR. In this study, the PRV vaccine strain Bartha-K61 was used as the reference strain. The gB protein was expressed via the

Conventional wisdom holds that the best way to treat infection with antibiotics is to 'hit early and hit hard'. A favoured strategy is to deploy two antibiotics that produce a stronger effect in combination than if either drug were used alone. But are such synergistic combinations necessarily optimal? We combine mathematical modelling, evolution experiments, whole genome sequencing and genetic manipulation of a resistance mechanism to demonstrate that deploying synergistic antibiotics can, in practice, be the worst strategy if bacterial clearance is not achieved after the first treatment phase

Human immunodeficiency virus type 2 (HIV-2) infections cause severe immunodeficiency in humans, although HIV-2 is associated frequently with reduced virulence and pathogenicity compared to HIV-1. Genetic determinants that play a role in HIV pathogenesis are relatively poorly understood but nef has been implicated in inducing a more pathogenic phenotype in vivo. However, relatively little is known about the role of nef in HIV-2 pathogenesis. To address this, the genetic composition of 44 nef alleles from 37 HIV-2-infected individuals in Portugal, encompassing a wide spectrum of disease associat

STUDY DESIGN: Retrospective cohort.
OBJECTIVES: To determine the relative contributions of anthropometric factors, physical activity, back and neck pain, overall health, and familial aggregation (the combined effects of genetics and childhood environment) to different measurements of adult back muscle function.
SUMMARY OF BACKGROUND DATA: Many methods of muscle testing are employed in the attempt to predict, prevent, treat, and rehabilitate low back pain. Poor correlations between the test results suggest that they are measuring different attributes and have different determinants.
METHODS:

Intervertebral disc disease (IVDD) is a common musculoskeletal disease affecting about 5% of all individuals. It is characterized by lumbar disc herniation, which causes nerve root irritation, either mechanically or via inflammatory mediators, and results in radiating pain, known as sciatica. Numerous studies have been conducted to identify the causes and risk factors for IVDD. Lifting heavy loads, torsional stress, and motor vehicle driving are among the best-identified environmental risk factors. However, it has become evident recently from family and twin studies that genetic factors may al

BACKGROUND: People with human immunodeficiency virus type 1 (HIV-1; PWH) on antiretroviral therapy (ART) and suppressed viremia usually experience a decrease in HIV DNA over time, but about 25% experience an increase. Some also experience intermittent viremia. The reasons for and potential clinical implications of increases in the HIV reservoir remain unclear and a major concern.
METHODS: In this study, we longitudinally characterized the proviral landscape in 4 distinct groups of PWH (n = 40) successfully treated with ART over 10.4 years without any viral failure, presenting either an increa

BACKGROUND: Brucellosis poses a significant threat to animal and human health globally. However, how Brucella subverts the immune response to establish persistent infections remains unclear.
METHODS: We utilized single-cell RNA sequencing (scRNA-seq) to decipher the immune landscape of mice infected with Brucella abortus. Flow cytometry, a transgenic cell line and mouse, and antibody blockage were utilized to explore the relevant mechanisms.
RESULTS: Brucella infection induced significant changes in the composition and signaling pathways of immune cells, and flow cytometry analysis further c

Japanese encephalitis (JE) is a re-emerging viral zoonotic disease transmitted primarily by the bite of infected Culex mosquitoes with ardeid birds as reservoir host for JE virus (JEV). Swine being the amplifier host have been studied extensively, however the role of domestic poultry in JEV epidemiology remains unexplored. Therefore, the present study was undertaken with the aim of quantifying JE viral load and to identify the circulating genotype of JEV in domestic chicken and duck population of India. A total of 206 chicken and 76 duck samples collected across five JE endemic states of India
