Low variability in the capsid gene of a generalist densovirus across lepidopteran hosts and geographic regions.

Viruses are ubiquitous in all ecosystems, and insects can serve as model host systems for understanding their geographic range and host specificity. Lepidoptera are relatively easy to collect and rear and are known to harbor diverse groups of viral pathogens. Much of the research on viruses in Lepidoptera has focused on baculoviruses, but this is beginning to change with better sequencing techniques. In this study, we use genetic tools to characterize the host and geographical range of the Junonia coenia densovirus (JcDV) (Family Parvoviridae, subfamily Densovirinae, genus Protoambidensovirus,…

Taxonomy and molecular phylogeny of Urocleidoides spp. (Monopisthocotyla: Dactylogyridae): Four new species and cophylogenetic patterns in a clade of species parasitizing Characiformes hosts.

Historical reconstruction studies are important for understanding the evolutionary mechanisms associated with different parasite-host systems. Platyhelminths of the classes Monopisthocotyla and Polyopisthocotyla (formerly Monogenoidea or Monogenea) have proven to be excellent models for historical reconstruction studies due to their exceptional parasite specificity, suggesting that cospeciation events are the main pattern observed in these parasite-host systems (i.e., codivergence). However, recent studies indicate that not only cospeciation but also duplication, host-switch, and loss (extinct

Advancing In Vivo Detection of T-Cell Function: Development and Preclinical Evaluation of 89Zr-Ivuxolimab, a Human OX40 PET Tracer.

The variable response to cancer immunotherapies highlights a critical gap in our ability to predict and monitor treatment efficacy. To address this, there is an urgent clinical need for advanced molecular imaging technologies that can noninvasively and precisely assess whole-body immune responses. The OX40 receptor (CD134), a potent costimulatory molecule on T cells, serves as a highly specific marker of T-cell activation, an early and crucial event in immunotherapy efficacy. In this study, we report the development of a human OX40-specific radiotracer based on a clinically evaluated therapeut

Divergent viral phosphodiesterases for immune signaling evasion.

Cyclic dinucleotides (CDNs) and other short oligonucleotides play fundamental roles in immune system activation in organisms ranging from bacteria to humans. In response, viruses use phosphodiesterase (PDE)-mediated oligonucleotide cleavage for immune evasion, a strategy whose diversity has not yet been explored. Here, we use a canonical 2H PDE (2H PDE) structure-based search of prokaryotic and eukaryotic viral sequences to identify an exceptional diversity of 2H PDEs across the virome, including enzymes not detectable with sequence search methods alone. Despite active site conservation, bioch

Observations on the use of ELISA for detection of Babesia bigemina specific antibodies.

An indirect enzyme-linked immunosorbent assay (ELISA) was evaluated to study the cause of the high level of background reactions which hinders the application of ELISA as a field diagnostic test for Babesia bigemina. Different blockers to improve the specificity of the ELISA were compared. THe use of soya milk (25%), gelatin (2.5%) and chicken serum (2%) did not significantly improve the specificity of the test. It was noted that the presence of fibrinogen contributed to the positive ELISA results more than the presence of B. bigemina specific antigen. This conclusion was confirmed by testing

Commentary on the molecular and morphological identification of microfilariae in domestic chickens: Future perspectives.

This letter is a response to the recent article on the morphological and molecular identification of microfilariae in chickens in Northeastern Thailand. We recommend the authors for their important contributions to avian parasitology and offer additional perspectives on molecular techniques, sex-based infection patterns, pathological implications, and host specificity. Emphasis is placed on potential improvements in diagnostic methods and suggestions for future research directions to enhance understanding of filarial infections in poultry and their broader One Health relevance.

Nucleotide Substrate Specificity of Anti-Hepatitis C Virus Nucleoside Analogs for Human Mitochondrial RNA Polymerase.

Nucleoside analog inhibitors (NAIs) are an important class of antiviral agents. Although highly effective, some NAIs with activity against hepatitis C virus (HCV) can cause toxicity, presumably due to off-target inhibition of host mitochondrial RNA polymerase (POLRMT). The in vitro nucleotide substrate specificity of POLRMT was studied in order to explore structure-activity relationships that can facilitate the identification of nontoxic NAIs. These findings have important implications for the development of all anti-RNA virus NAIs.

Colostral transfer of antibodies specific for Hypoderma lineatum proteins.

Antibodies to Hypoderma lineatum were transferred to calves via colostrum. The antibodies transferred in the colostrum demonstrated specificity to all the H. lineatum first-instar proteins which were resolved by non-denaturing polyacrylamide gel electrophoresis and were capable of mediating a Type I hypersensitivity reaction. The kinetic decline of colostrum-acquired antibodies, the effect upon development of an autologous humoral response to H. lineatum and the host or parasite protective role of these antibodies are discussed.

Bartonella entry mechanisms into mammalian host cells.

The Gram-negative genus Bartonella comprises arthropod-borne pathogens that typically infect mammals in a host-specific manner. Bartonella bacilliformis and Bartonella quintana are human-specific pathogens, while several zoonotic bartonellae specific for diverse animal hosts infect humans as an incidental host. Clinical manifestations of Bartonella infections range from mild symptoms to life-threatening disease. Following transmission by blood-sucking arthropods or traumatic contact with infected animals, bartonellae display sequential tropisms towards endothelial and possibly other nucleated

Factors associated with diversity, quantity and zoonotic potential of ectoparasites on urban mice and voles.

Wild rodents are important hosts for tick larvae but co-infestations with other mites and insects are largely neglected. Small rodents were trapped at four study sites in Berlin, Germany, to quantify their ectoparasite diversity. Host-specific, spatial and temporal occurrence of ectoparasites was determined to assess their influence on direct and indirect zoonotic risk due to mice and voles in an urban agglomeration. Rodent-associated arthropods were diverse, including 63 species observed on six host species with an overall prevalence of 99%. The tick Ixodes ricinus was the most prevalent spec

Host-associated Bifidobacterium longum exhibit gastrointestinal resilience and antagonistic activity against Salmonella serovars.

Probiotics are increasingly explored as alternatives to antibiotics for preventing and controlling infectious diseases in livestock, including enteritis, pneumonia, and sepsis. However, most commercially available probiotic strains originate from humans, which can limit their colonization efficiency and functional performance in livestock due to host-specificity. In this study, we isolated twelve Bifidobacterium longum strains from healthy dairy calves and evaluated their probiotic potential with a focus on gastrointestinal resilience and antimicrobial activity. Comparative genomic and phyloge

The molecular mechanisms of autophagy-virus interactions: from antiviral defense to viral counterstrategies.

Autophagy is a highly conserved cellular degradative pathway in eukaryotes that exerts a dual regulatory role in the host antiviral defense. On the one hand, as an essential component of the host antiviral defense network, autophagy participates in the direct degradation of viral components, synergistically modulates innate immune signaling pathways including TLRs, RLRs, and cGAS-STING, and regulates adaptive immune responses such as MHC class I/II-mediated antigen presentation. On the other hand, diverse viruses have evolved sophisticated immune evasion mechanisms during long-term coevolution

The final gate: bacterial subversion of autophagosome-lysosome fusion as a conserved immune evasion strategy.

Autophagy is a vital component of the host cell intracellular defense arsenal, culminating in the fusion of autophagosomes with lysosomes to degrade invading pathogens. While autophagosome formation has been extensively studied, recent insights reveal that the final fusion step constitutes a critical immunological bottleneck that is highly vulnerable to microbial sabotage. In this review, we synthesize evidence from diverse pathogens, including Mycobacterium tuberculosis, Salmonella enterica, Treponema pallidum, Helicobacter pylori, Coxiella burnetii, Yersinia pestis, and Porphyromonas gingiva

Structural characterization of zinc-bound Zmp1, a zinc-dependent metalloprotease secreted by Clostridium difficile.

Proteases are commonly secreted by microorganisms. In some pathogens, they can play a series of functional roles during infection, including maturation of cell surface or extracellular virulence factors, interference with host cell signaling, massive host tissue destruction, and dissolution of infection-limiting clots through degradation of the host proteins devoted to the coagulation cascade. We previously reported the identification and characterization of Zmp1, a zinc-dependent metalloprotease secreted by Clostridium difficile, demonstrated that Zmp1 is able to degrade fibrinogen in vitro,

Primary cell culture of Echinococcus granulosus developed from the cystic germinal layer: biological and functional characterization.

Cell cultures of parasitic helminths are an invaluable tool for investigations of basic biological processes, as well as for development of improved chemotherapeutic agents and molecular interactions between host and parasite. We carried out a simple and efficient methodology to isolate Echinococcus granulosus germinal cells which were maintained for at least 4 months while cultivated in the presence of reducing agents and hormones. Microscopic analysis of the primary cell culture revealed the presence of cells with similar Echinococcus germinal cell morphology and behaviour. Population doubli

Optimizing Lankesterella Detection in Avian Hosts: A Comparative Analysis of Microscopic and Molecular Techniques.

Lankesterella Labbe, 1899 is a genus of blood parasites found in reptiles, amphibians, and birds that has been poorly understood. In birds, it was originally classified as Hepatozoon Miller, 1908 or Atoxoplasma Garnham, 1905, but recent studies have revealed the diversity, prevalence, and host specificity of Lankesterella using integrative approaches, particularly the 18S rDNA gene. The introduction of new diagnostic protocols focusing on mitochondrial genes requires assessing the comparability of different diagnostic methods. This study aims to compare microscopy and molecular detection metho…

Prediction of the occurrence of leprosy reactions based on Bayesian networks.

INTRODUCTION: Leprosy reactions (LR) are severe episodes of intense activation of the host inflammatory response of uncertain etiology, today the leading cause of permanent nerve damage in leprosy patients. Several genetic and non-genetic risk factors for LR have been described; however, there are limited attempts to combine this information to estimate the risk of a leprosy patient developing LR. Here we present an artificial intelligence (AI)-based system that can assess LR risk using clinical, demographic, and genetic data. METHODS: The study includes four datasets from different regions o
