Microporous SPEEK Implants Decorated with Bioactive PG/CS/HA Hydrogels for Improving Osseointegration Potential.

Polyether ether ketone (PEEK) is widely used in orthopedics due to its good mechanical properties and biocompatibility; however, its bioactivity and osseointegration ability are poor. In this article, PEEK materials were dual-modified by surface sulfonation treatment and the loading of active hydrogels (phosphorylated gelatin/chitosan/hydroxyapatite) to improve their bioactivity and osseointegration. Initially, a microstimulated sulfonation process was employed to construct an optimized porous structure on the PEEK surface, with a porosity of 54.63% and an average pore size of 3.38 μm, which s…

Characterization of Sex-Based Differences in Integrin-Mediated Endothelial Cell Adhesion to Bioactive Hydrogels.

Endothelialization promotes thromboresistance in blood-contacting devices, but biomaterial designs often overlook sex differences in the end … e cells. Collectively, these results demonstrate sex-biased endothelial cell responses to bioactive hydrogels mediated by integrin interactions and highlight the importance of incorporating biological sex as a design variable in the development of blood-contacting biomaterials.

One-step fabrication of adhesive, stable, and antioxidant polylipoic acid hydrogels for wound healing.

Lipoic acid-based hydrogels are considered promising candidates for wound dressings due to their antioxidant properties. However, their practical application has been hindered by complex and time-consuming fabrication methods. To address this limitation, a novel oscillation-driven strategy was developed for the rapid and energy-efficient preparation of lipoic acid@polyethyleneimine-x-poly (ethylene glycol) diacrylate-y-protocatechualdehyde-z (LA@PEIx-GyAz) hydrogels. In detail, the polymerization of lipoic acid (LA) is initiated by polyethyleneimine (PEI) to form polylipoic acid (PolyLA), whic…

Application potential of injectable hydrogels in the post-surgical window period following tumor surgery.

Chemotherapy is one of the primary modalities for the treatment of malignant diseases. The outcomes, however, are different between tumors of various origins, which hinder clinical applications. The advantages of chemotherapies in patients with hematological lesions are more obvious than those seen in solid tumors. This might be attributed to the availability of drug concentration and exposure time. Based on this phenomenon, we hypothesis that localized drug administration is expected to be more potential for solid tumors, particularly for the residual tumors in post-operative "window period".

Creation of a rich vascular subcutaneous space for cell transplantation via injectable biological hydrogels.

The subcutaneous space offers an attractive and accessible site for cell transplantation. However, its clinical utility is often hindered by insufficient vascularization. This study evaluated the vascularization potential of three hydrogels; human collagen type I, human fibrin, and alginate implanted subcutaneously in Sprague-Dawley rats. Polylactide-co-glycolide (PLG) scaffolds used as positive controls for foreign body response. Sequential injections were performed at 1, 2, and 4 weeks, and tissues were retrieved for histopathological examination. Human collagen type I and fibrin induced rob…

Synergistic Prevascularization with Proangiogenic Silica Nanoparticles and VEGF-Mimetic Aptamer in Tailored GelMA Hydrogels.

Angiogenesis is a crucial and challenging requirement for the regeneration and repair of damaged tissues, particularly for critical-sized ones. To address this challenge, in this study, we fabricated a cell-communicating gelatin methacryloyl (GelMA) hydrogel using core-shell silica nanoparticles conjugated with roxadustat (FG-4592) and a VEGF-mimetic aptamer (Apt02). This hydrogel promotes tube formation and prevascularization synergistically through both extracellular and intracellular pathways in human umbilical vein endothelial cells (HUVEC), with FG-4592 acting via the extracellular pathwa

Treatment of Denervated Muscle Atrophy by Injectable Dual-Responsive Hydrogels Loaded with Extracellular Vesicles.

Denervated muscle atrophy, a common outcome of nerve injury, often results in irreversible fibrosis due to the limited effectiveness of current therapeutic interventions. While extracellular vesicles (EVs) offer promise for treating muscle atrophy, their therapeutic potential is hindered by challenges in delivery and bioactivity within the complex microenvironment of the injury site. To address this issue, an injectable hydrogel is developed that is responsive to both ultrasound and pH, with inherent anti-inflammatory and antioxidant properties, designed to improve the targeted delivery of ste

Biocomposite Polyvinyl Alcohol/Ferritin Hydrogels with Enhanced Stretchability and Conductivity for Flexible Strain Sensors.

Protein-based hydrogels with stretchability and conductivity have potential applications in wearable electronic devices. However, the development of protein-based biocomposite hydrogels is still limited. In this work, we used natural ferritin to develop a PVA/ferritin biocomposite hydrogel by a repetitive freeze-thaw method. In this biocomposite hydrogel, ferritin, as a nano spring, forms a hydrogen bond with the PVA networks, which reduces the crystallinity of PVA and significantly improves the stretchability of the hydrogel. The fracture strain of the PVA/ferritin hydrogel is 203%, and the f…

Developing tough, fatigue-resistant and conductive hydrogels via in situ growth of metal dendrites.

Developing hydrogels with high conductivity and toughness via a facile strategy is important yet challenging. Herein, we proposed a new strategy to develop conductive hydrogels by growing metal dendrites. Water-soluble Sn2+ ions were soaked into the gel and then converted to Sn dendrites via an electrochemical reaction; the excessive Sn2+ ions were finally removed by water dialysis, accompanied by dramatic shrinkage of the gel. Based on in situ transformation from metal ions to dendrites, the method integrated the advantages of ionic conductive fillers, such as LiCl (uniform dispersion), and e…

Application and prospects of polysaccharide-based hydrogels in the treatment of intervertebral disc degeneration.

Intervertebral disc degeneration (IVDD) is a primary contributor to chronic low back pain (LBP) and neurological dysfunction. It significantly impairs patients' quality of life. With an aging population and changing lifestyles, the incidence of IVDD has been rising steadily. However, traditional treatments, such as medications, physical therapy, and surgery, primarily alleviate symptoms without fundamentally repairing degenerated discs. Recent advances in tissue engineering and regenerative medicine have led to the development of new therapeutic strategies for IVDD. Among them, hydrogels, whic…

Injection of vocal fold lamina propria-derived hydrogels modulates fibrosis in injured vocal folds.

Vocal fold (VF) fibrosis, often resulting from phonosurgery, radiation, or trauma, causes irreversible voice dysfunction due to excessive ECM deposition and increased tissue stiffness. No FDA-approved treatments for VF fibrosis exist, highlighting the need for novel antifibrotic therapies. TGF-β1 contributes to fibroblast-to-myofibroblast activation, leading to increased ACTA2 expression and collagen production via SMAD3, YAP1, and integrin signaling pathways. Leveraging the principle that local cells respond to tissue-specific signals, our ECM hydrogel, derived from decellularized vocal fold

An Injectable Zwitterionic Hydrogels with Multiple Intermolecular Interactions for Effective Prevention of Abdominal Adhesions.

Postoperative abdominal adhesions are the most common complication following abdominopelvic surgery, posing a significant burden on patients, clinicians, and society. However, current physical barriers often involve a tradeoff between preventing these adhesions and inhibiting inflammation. Herein, a one-stone-two-birds strategy is presented to address this challenge through an injectable intertwined hydrogel containing sulfobetaine, modified aminocaproic acid (A6ACA), and ZnO nanoparticles (PSA-ZnO hydrogel). This intertwined network is stabilized by multiple intermolecular coordination bonds,

Cyanobacterial Ampholyte Hydrogels Developed by the Cationization of Sulfated Polysaccharides and Their Cell-Compatibility.

Sacran is a cyanobacterial supergiant polysaccharide with carboxylate and sulfate groups that exhibits antiallergic and antiinflammatory properties. However, its high anionic functions restrict cell compatibility. Quaternary ammonium groups were substituted to form sacran ampholytes, and the cell compatibility of the cationized sacran hydrogels was evaluated. The cationization process involved the reaction of N-(3-chloro-2-hydroxypropyl)trimethylammonium chloride with the primary amine or hydroxyl groups of sacran. The degree of cationization ranged from 32 to 87% for sugar residues. Hydrogels…

Superhydrophobic and super-stretchable conductive composite hydrogels for human motion monitoring in complex condition.

With the advent of the information age, there is a growing demand for wearable sensing devices. Conventional hydrogels are a class of materials that can hold a large amount of water, with a three-dimensional network of hydrophilic polymerization chains inside. In remote areas or harsh environments, there is an urgent demand for a flexible sensor that is environmentally stable, wearable, and has high mechanical properties. Due to the hydrophilicity of the traditional hydrogel surface, it is easy to adsorb dust or be contaminated by liquid, which limits its further application. As a result, the …

Cold-resistant, highly stretchable ionic conductive hydrogels for intelligent motion recognition in winter sports.

Conductive hydrogels have attracted much attention for their wide application in the field of flexible wearable sensors due to their outstanding flexibility, conductivity and sensing properties. However, the weak mechanical properties, lack of frost resistance and susceptibility to microbial contamination of traditional conductive hydrogels greatly limit their practical application. In this work, multifunctional polyvinyl alcohol (PVA)/carboxymethyl cellulose (CMC)/poly(acrylamide-co-1-vinyl-3-butylimidazolium bromide) (P(AAm-co-VBIMBr)) (PCPAV) ionic conductive hydrogels with high strength an…
