S100A8/9-NLRP3-mediated chronic unresolved inflammation drives cardiac pathologies following invasive pneumococcal disease.

Streptococcus pneumoniae (Spn) is the leading cause of community-acquired pneumonia (CAP). A quarter of hospitalized patients with CAP experience a major adverse cardiac event (MACE), raising their mortality by four to five times compared with pneumonia alone. Patients with CAP continue to face a significantly greater risk of MACE and cardiovascular-associated death during convalescence. However, the reasons responsible for this remain unclear. To elucidate the molecular mechanism(s) of Spn-induced MACE in convalescence, a mouse model of Spn infection and antibiotic rescue was employed. A mark

Minocycline inhibits rosacea-like inflammation through the TLR4-mediated NF-κB signaling pathway in vivo and in vitro.

BACKGROUND: Rosacea is a chronic inflammatory skin disease characterized by multiple intricate pathogenic factors. Previous studies have substantiated the anti-inflammatory properties of minocycline and its potential therapeutic efficacy in treating rosacea. However, further elucidation of the underlying mechanism is warranted. METHODS: HaCaT cells and BALB/c mice were treated with LL37. Moreover, the effect of minocycline on rosacea was explored through the addition of an NF-κB inhibitor (PDTC) or overexpression of Toll-like receptor 4 (TLR4). The expression of related markers was detected b

Empagliflozin protected kidney function in CKD rat through suppressing hypoxic and fibrotic signalings mediated inflammation and EMT.

BACKGROUND: This study tested the hypothesis that PI3K/Akt/GSK3β and TGF-β/Smad2/3 signaling play essential roles in mediating the epithelial-mesenchymal transition (EMT) and fibrosis, resulting in the deterioration of renal function and parenchyma in chronic kidney disease (CKD) rats, which is reversed by early empagliflozin treatment. METHODS AND RESULTS: NRK-52E cells were divided into the A1 (NRK-52E), A2 (NRK-52E + 200 μM p-Cresol), A3 (NRK-52E + 200 μM p-Cresol + 50 μM empagliflozin), B1 (NRK-52E), B2 (NRK-52E + 5 ng/mL TGF-β1) and B3 (NRK-52E + 5 ng/mL TGF-β1 + 50 μM empagliflozin) gro

Neuroinflammatory crosstalk in migraine: consolidated activity of rizatriptan and meloxicam in suppressing CGRP-induced nociception and COX-mediated inflammation.

Migraine is among the most frequently observed neurovascular malady affecting almost 1 billion people globally and is defined by frequent episodes of headache, accompanying neurological problems, and a markedly reduced standard of health. Migraine pathogenesis is directly linked to the CGRP (Calcitonin-gene-related-peptide), a strong vasodilating protein entangled in the exchange of nociceptor signals, and the cyclo-oxygenase (COX) enzymes, influencing inflammatory responses which aggravate migraine manifestations. Although COX inhibiting drugs as well as CGRP receptor blockers have therapeuti

miR-17195 promotes infectious bronchitis virus proliferation and macrophage-mediated inflammation via the PLCβ2-TAK1 axis.

Infectious Bronchitis virus (IBV) causes Infectious bronchitis (IB) is an acute, highly contagious disease primarily affecting chickens and other avian species, characterized by respiratory and renal pathologies. MicroRNAs (miRNAs) play pivotal roles in virus-host interactions and regulate diverse physiological and pathological processes during viral infection. Here, we demonstrate that IBV infection upregulates host-derived miR-17195 in HD11 cells, which targets and suppresses PLCβ2 to enhance replication. Moreover, miR-17195 promotes TAK1 phosphorylation by suppressing PLCβ2, which subsequen

RAPGEF6 is a key biomarker with temporal characteristics in sepsis and exacerbates inflammation and oxidative stress by NF-κB-mediated necroptosis.

Biomarkers identified through integrated bioinformatics approaches often exhibit limited temporal stability during sepsis progression. To address this limitation, temporal changes in the serum transcriptome of sepsis patients were analyzed at multiple time points to investigate underlying pathological mechanisms. The datasets GSE54514 and GSE212092 from the Gene Expression Omnibus (GEO) database were used for gene identification and validation. The key marker RAPGEF6 with temporal characteristics was identified by integrating time-series clustering analysis, differential expression analysis, w

PACAP a mediator of inflammation following trauma exposure and mild traumatic brain injury: Differential effects in males and females.

Individual differences in systemic responses to trauma exposure, posttraumatic stress disorder (PTSD), and/or mild traumatic brain injury (mTBI) may help account for differential risk of adverse sequalae in females and heterogeneity in pathophysiology, symptoms, and treatment responses. Accordingly, this study investigated sex differences in the association between neuroendocrine (pituitary adenylate cyclase-activating polypeptide [PACAP]) and inflammatory markers following lifespan trauma exposure, PTSD, and mTBI in 71 trauma-exposed veteran and non-veteran males (n = 41) and females (n = 30)

Aryl hydrocarbon receptor-induced activation of AIM2 inflammasome is mediated by MOMP and MPT: A vital therapeutic pathway for inflammation.

Understanding how mitochondrial dysfunction (MD) triggers damage-associated molecular pattern (DAMP) signaling could advance therapies for inflammatory diseases. While aryl hydrocarbon receptor (AHR) is implicated in MD-related inflammation, its mechanistic role remains unclear. 8:2 fluorotelomer alcohol (8:2 FTOH), a per- and polyfluoroalkyl substance compound, was identified as an exogenous ligand for AHR. We demonstrate that AHR ligands drive mitochondrial outer membrane permeabilization (MOMP) and mitochondrial permeability transition (MPT) via transcriptional and non-transcriptional activ

Linking sleep microstructure, blood markers of inflammation and metabolism, and cognition: mediation analysis in the osteoporotic fractures in men study.

The sleep electroencephalographic (EEG) microstructure is explicitly related to brain functions, such as sleep spindle and memory consolidation. On the other hand, given the crosstalk between the central nervous system and the body, including inflammation regulation and metabolic systems, there is a gap in understanding the bidirectional relationship between sleep and these systems that contribute to cognitive health. We used data from the Osteoporotic Fractures in Men (MrOS) Study, a six-site cohort study of community-dwelling men 65 years or older. We analyzed 1898 participants who participa

Gentiopicroside ameliorates synovial inflammation and fibrosis in KOA rats by modulating the HMGB1-mediated PI3K/AKT signaling axis.

BACKGROUND: Knee osteoarthritis (KOA) is a degenerative joint disease characterized by synovial inflammation and fibrosis. Gentiopicroside (GPS), one of the main active ingredients of Gentiana macrophylla, is widely used in anti-inflammatory and anti-fibrotic therapies. However, the exact mechanism by which GPS treats synovial inflammation and fibrosis in KOA remains unclear. METHODS: Fibroblast-like synoviocytes (FLSs) were stimulated with lipopolysaccharide (LPS) to induce inflammation and fibrosis, and CCK-8 was performed to determine the viability of GPS-treated FLSs, using immunofluoresc

Mechanoreceptor Piezo1 channel-mediated interleukin expression in conjunctival epithelial cells: Linking mechanical stress to ocular inflammation.

PURPOSE: Mechanical stress on the ocular surface, such as from eye-rubbing, has been reported to lead to inflammation and various ocular conditions. We hypothesized that the mechanosensitive Piezo1 channel in the conjunctival epithelium contributes to the inflammatory response at the ocular surface after receiving mechanical stimuli. METHODS: Human conjunctival epithelial cells (HConjECs) were treated with Yoda1, a Piezo1-specific agonist, and various allergens to measure cytokine expression levels using qRT-PCR. Piezo1 activation-induced intracellular signaling pathways were also investigate

Inhibitory Effect of a Novel Non-Steroidal AMPamide on Inflammation and Sebogenesis by Suppressing TLR4 and TLR6-Mediated Signaling Pathway.

Seborrheic dermatitis (SD) is a chronic inflammatory skin disorder that primarily affects areas with increased sebaceous gland activity, and is characterized by erythematous scaly lesions. Malassezia utilizes sebum lipids to produce free fatty acids that may disrupt the epidermal barrier and trigger inflammation in eczematous lesions. However, the pathogenesis and mechanisms underlying the exaggerated inflammatory response and sebogenesis regulation in SD remain unknown. Activation of pattern recognition receptors, including Toll-like receptors (TLRs), is crucial for initiating innate immunity

N-glycosylation of SCAP exacerbates hepatocellular inflammation and lipid accumulation via ACSS2-mediated histone H3K27 acetylation.

Sterol regulatory element binding protein (SREBP) cleavage-activating protein (SCAP) is a widely expressed membrane glycoprotein that acts as an important modulator of lipid metabolism and inflammatory stress. N-glycosylation of SCAP has been suggested to modulate cancer development, but its role in nonalcoholic steatohepatitis (NASH) is poorly understood. In this study, the N-glycosylation of SCAP was analyzed by using sequential trypsin proteolysis and glycosidase treatment. The liver cell lines expressing wild-type and N-glycosylation sites mutated SCAP were constructed to investigate the N

Selectively Antagonizing the NOD1-Mediated Inflammatory Signaling Pathway Mitigates the Gastric Inflammation Induced by Helicobacter pylori Infection.

Helicobacter pylori (H. pylori) infection is characterized by the complex interplay between H. pylori and gastric disorders. It has been established that NOD1 can be activated by the peptidoglycan (PGN) present in the cell wall of H. pylori, serving as a key mediator of inflammation and initiating the RIP2/NF-κB and MAPK inflammatory signaling pathways. In this article, we reported on the development of a 2-chloroquinazolin-4-ol derivative 66 as a potent and selective antagonist of both human and mouse NOD1, which effectively inhibited the expression of inflammatory cytokines (IL-6, TNF-α) and
