Pollution and its impact on wild animals: a meta-analysis on oxidative stress.

Oxidative stress is the unifying feature underlying the toxicity of anthropogenic pollution (e.g., heavy metals, polycyclic aromatic hydrocarbons, and nitrogen-oxides) and the ultimate culprit in the development of many diseases. Yet, there has been no attempt to summarize the published data on wild terrestrial animals to reveal general trends regarding the effects of pollution on oxidative stress. The main findings of this meta-analysis reveal that, as predicted, there is an overall increase in oxidative stress when exposed to pollution. This is mainly due to a weak overall increase of oxidat

Preconceptional stress and racial disparities in preterm birth: an overview.

OBJECTIVE: We reviewed the evidence for three theories of how preconceptional psychosocial stress could act as a contributing determinant of excess preterm birth risk among African American women: early life developmental plasticity and epigenetic programming of adult neuroendocrine systems; blunting, weathering, or dysfunction of neuroendocrine and immune function in response to chronic stress activation through the life course; individuals' adoption of risky behaviors such as smoking as a response to stressful stimuli. METHODS: Basic science, clinical, and epidemiologic studies indexed in M

Drought-activated BZR1 reprograms flavonoid metabolism via transcriptional cascades to amplify baicalin biosynthesis in Scutellaria baicalensis.

Environmental stress triggers a dual response in medicinal plants by inhibiting growth while enhancing secondary metabolite production. However, the regulatory mechanism of this trade-off remains unclear. Here, we identified SbBZR1 as a brassinosteroid (BR)-responsive transcription factor that orchestrates drought resilience and baicalin biosynthesis in Scutellaria baicalensis through integrated multi-omics and metabolic flux analysis. In general, drought stress upregulated BR signaling and SbBZR1 expression. Transgenic Arabidopsis expressing SbBZR1 and its hyperactive mutant (Sbbzr1-1D) exhib

Synthetic microbial communities for engineering climate-smart biofertilizers.

Global agricultural productivity is increasingly destabilized by climate change-driven droughts, floods, extreme heat, and severe storms. Although the climate-smart agriculture (CSA) framework addresses these challenges, implementation has focused mainly on plant genetics and agronomic inputs, leaving the adaptive potential of the crop microbiome underexplored. Here, we examine the agricultural use of synthetic microbial communities (SynComs) through the "crop holobiont" concept, in which plants and their associated microbiota function as an integrated, responsive system rather than through pl

Elevated plasma sulfides are associated with cognitive dysfunction and brain atrophy in human Alzheimer's disease and related dementias.

Emerging evidence indicates that vascular stress is an important contributor to the pathophysiology of Alzheimer's disease and related dementias (ADRD). Hydrogen sulfide (H2S) and its metabolites (acid-labile (e.g., iron-sulfur clusters) and bound (e.g., per-, poly-) sulfides) have been shown to modulate both vascular and neuronal homeostasis. We recently reported that elevated plasma sulfides were associated with cognitive dysfunction and measures of microvascular disease in ADRD. Here we extend our previous work to show associations between elevated sulfides and magnetic resonance-based metr

Integration of postnatal craniofacial development and the effects of diet consistency in rodents.

Phenotypic covariances between traits, shaped by developmental processes and functional integration, play a critical role in the response to natural selection. The influences of environmental factors (e.g. food availability and consistency) on these covariances might be particularly prevalent when functional and anatomical complexes develop. In this study, we investigate the ontogenetic development and plasticity of the craniofacial complex, focusing on the integration between skull and mandible in three rodent species: house mouse, golden hamster and Mongolian gerbil. By raising these species

Pulmonary Endothelial Mechanical Sensing and Signaling, a Story of Focal Adhesions and Integrins in Ventilator Induced Lung Injury.

Patients with critical illness such as acute lung injury often undergo mechanical ventilation in the intensive care unit. Though lifesaving in many instances, mechanical ventilation often results in ventilator induced lung injury (VILI), characterized by overdistension of lung tissue leading to release of edemagenic agents, which further damage the lung and contribute to the mortality and progression of pulmonary inflammation. The endothelium is particularly sensitive, as VILI associated mechanical stress results in endothelial cytoskeletal rearrangement, stress fiber formation, and integrity

Highly selective turn-on chemodosimetric sensing of proline for environmental and hydroponic monitoring.

Proline is an important biomarker in biological regulation and plant stress adaptation, increasing the demand for highly specific analytical detection methods. Herein, we report the use of DMAC as a chemodosimetric probe that exclusively identifies proline in the presence of 19 competing amino acids. The sensing mechanism relies on an irreversible chemical reaction between proline and DMAC, producing a distinct optical response with negligible background interference. This exhibits a highly sensitive limit of detection of 15.8 nM (1.82 ppb), signifying its importance in trace-level quantificat

The impaired unfolded protein-premelanosome protein and transient receptor potential channels-autophagy axes in apoptotic melanocytes in vitiligo.

Vitiligo is an autoimmune skin disease, characterized by depigmentation and epidermal melanocytes loss. The specific mechanisms underlying vitiligo have not been fully understood. As a result, treating vitiligo is a dermatological challenge. Recently, much attention has been paid to the dysfunction and interaction of organelles under environmental stress. The impaired organelles could generate misfolded proteins, particularly accumulated toxic premelanosome protein (PMEL) amyloid oligomers, activating the autoimmune system and cause melanocyte damage. Unfolded protein response (UPR) dysfunctio

Emerging Role for the PERK/eIF2α/ATF4 in Human Cutaneous Leishmaniasis.

Leishmania parasites utilize adaptive evasion mechanisms in infected macrophages to overcome host defenses and proliferate. We report here that the PERK/eIF2α/ATF4 signaling branch of the integrated endoplasmic reticulum stress response (IERSR) is activated by Leishmania and this pathway is important for Leishmania amazonensis infection. Knocking down PERK or ATF4 expression or inhibiting PERK kinase activity diminished L. amazonensis infection. Knocking down ATF4 decreased NRF2 expression and its nuclear translocation, reduced HO-1 expression and increased nitric oxide production. Meanwhile,

Effects of L-NAME and air exposure on mitochondrial energetic markers, thyroid hormone receptor/regulator system and stress/ease-responsive receptor expression in the brain/gut axis of zebrafish.

As a signal molecule, nitric oxide (NO) has several physiological actions in fish. However, the action of NO on the brain/gut axis, a classic inter-organal axis that bridges the gastrointestinal tract and the CNS, still requires more understanding. The short-term in vivo action of a NO inhibitor, N-omega-nitro-L-arginine methyl ester (L-NAME), on mitochondrial energetic markers and the receptor expression of thyroid hormone (TH) and neuroendocrine hormones involved in stress/ease response was tested in the brain/gut axis of zebrafish exposed to either in non-stressed or air-exposed condition.

The role of the corticotropin-releasing hormone and its receptors in the regulation of stress response.

Stress is an essential part of everyday life. The neuropeptide corticotropin-releasing hormone (CRH, also called CRF and corticoliberin) plays a key role in the integration of neuroendocrine, autonomic and behavioral responses to stress. The activation of the hypothalamic-pituitary-adrenal axis (HPA axis) by neurons of the paraventricular hypothalamic nucleus (PVN), the primary site of synthesis CRH, triggers stress reactions. In addition to the hypothalamus, CRH is widespread in extrahypothalamic brain structures, where it functions as a neuromodulator for coordination and interaction between

Shorter telomeres with high telomerase activity are associated with raised allostatic load and impoverished psychosocial resources.

Recent work has linked psychological stress with premature cellular aging as indexed by reduced leukocyte telomere length. The combination of shorter telomeres with high telomerase activity (TA) may be indicative of active cell stress. We hypothesized that older individuals characterized by shorter telomeres with high TA in unstimulated leukocytes would show signs of high allostatic load and low levels of protective psychosocial resources. We studied 333 healthy men and women aged 54-76 y who underwent laboratory testing in which we measured cardiovascular, neuroendocrine, and inflammatory res

Cyclic di-adenosine monophosphate as a potential regulator of sporulation and solventogenesis in Clostridium beijerinckii: a hypothesis.

Cyclic di-adenosine monophosphate (c-di-AMP) is a versatile bacterial signaling molecule involved in the regulation of cell wall metabolism, response to osmotic stress, growth, maintenance of DNA integrity, potassium homeostasis, and sporulation. Recent studies have implicated c-di-AMP in the regulation of sporulation and solventogenesis in Clostridium beijerinckii, a model solventogenic Clostridium species (SCS). In light of emerging findings, in this review, we hypothesize that c-di-AMP plays a central role in regulating solventogenesis and sporulation in C. beijerinckii, a mechanism that ma

A mycorrhiza-inducible phosphate transporter SlPT3 regulates phosphate uptake, iron homeostasis, and arbuscule development in tomato under combined nutrient stress conditions.

Interactions between phosphate (Pi) and zinc (Zn) or iron (Fe) nutrition in plants have been widely studied; however, the underlying mechanisms of their cross-talks in arbuscular mycorrhizal (AM) plants remain obscure. Here, we examine the physiological and molecular responses of tomato (Solanum lycopersicum L.) to the combination of Pi, Zn and/or Fe nutrient stresses during symbiosis with Rhizophagus irregularis, revealing the existence of a tripartite Pi-Zn-Fe cross-talk in AM symbiosis. Interestingly, the mycorrhiza-activated SlPT3, a member of the PHOSPHATE TRANSPORTER 1 (PHT1) gene family

Protein S-sulfhydration: Unraveling the prospective of hydrogen sulfide in the brain, vasculature and neurological manifestations.

Hydrogen sulfide (H2S) and hydrogen polysulfides (H2Sn) are essential regulatory signaling molecules generated by the entire body, including the central nervous system. Researchers have focused on the classical H2S signaling from the past several decades, whereas the last decade has shown the emergence of H2S-induced protein S-sulfhydration signaling as a potential therapeutic approach. Cysteine S-persulfidation is a critical paradigm of post-translational modification in the process of H2S signaling. Additionally, studies have shown the cross-relationship between S-sulfhydration and other cys
