Exploratory Computational Assessment of Lactiplantibacillus plantarum K25-Derived Fatty Acid Metabolites Against Leishmania donovani Centrin.

Probiotic derived metabolites have emerged as promising bio-active compounds with diverse antimicrobial and therapeutic applications. In the present study, an integrative metabolomic and computational approach was employed to investigate fatty acid metabolites produced by Lactiplantibacillus plantarum K25 and evaluate their potential activity against virulence associated targets to Leishmania donovani, the causative agent of visceral leishmaniasis (kala-azar). L. plantarum K25 exhibited robust growth across linoleic acid concentrations ranging from 1 to 10% (w/v), with maximum metabolite produ

Arachidonic acid-induced H+ and Ca2+ increases in both the cytoplasm and nucleoplasm of rat cerebellar granule cells.

1. Arachidonic acid (AA) exerts multiple physiological and pathophysiological effects in the brain. By continuously measuring the intracellular pH (pH(i)) and Ca2+ levels ([Ca2+]i) in primary cultured rat cerebellar granule cells, we have found, for the first time, that 20 min treatment with 10 microM AA resulted in marked increases in Ca2+ and H+ levels in both the cytosol and nucleus. 2. A much higher concentration (40 mM) of another weak acid, propionic acid, was needed to induce a similar change in pH(i). The [Ca2+]i increase was probably caused by AA-induced activation of Ni2+-sensitive c

Conserved fatty acid profiles and lipid metabolic pathways in a tropical reef fish exposed to ocean warming - An adaptation mechanism of tolerant species?

Climate warming is causing rapid spatial expansion of ocean warm pools from equatorial latitudes towards the subtropics. Sedentary coral reef inhabitants in affected areas will thus be trapped in high temperature regimes, which may become the "new normal". In this study, we used clownfish Amphiprion ocellaris as model organism to study reef fish mechanisms of thermal adaptation and determine how high temperature affects multiple lipid aspects that influence physiology and thermal tolerance. We exposed juvenile fish to two different experimental conditions, implemented over 28 days: average tro

Population-based metagenomics analysis reveals altered gut microbiome in sarcopenia: data from the Xiangya Sarcopenia Study.

BACKGROUND: Several studies have examined gut microbiota and sarcopenia using 16S ribosomal RNA amplicon sequencing; however, this technique may not be able to identify altered specific species and functional capacities of the microbes. We performed shotgun metagenomic sequencing to compare the gut microbiome composition and function between individuals with and without sarcopenia. METHODS: Participants were from a community-based observational study conducted among the residents of rural areas in China. Appendicular skeletal muscle mass was assessed using direct segmental multi-frequency bio

Causal relationship between serum metabolites and juvenile idiopathic arthritis: a mendelian randomization study.

BACKGROUND: Juvenile Idiopathic Arthritis (JIA) is a condition that occurs when individuals under the age of 16 develop arthritis that lasts for more than six weeks, and the cause is unknown. The development of JIA may be linked to serum metabolites. Nevertheless, the association between JIA pathogenesis and serum metabolites is unclear, and there are discrepancies in the findings across studies. METHODS: In this research, the association between JIA in humans and 486 serum metabolites was assessed using genetic variation data and genome-wide association study. The identification of causal re

Inborn errors of metabolism with a protein-restricted diet: effect on polyunsaturated fatty acids.

Previous studies have shown that phenylketonuric patients display a deficiency in long-chain polyunsaturated fatty acids. A study has now been performed on 13 cases (5 with methylmalonic acidaemia and 8 with urea cycle disorders) whose dietary treatment also implies a limitation in protein-rich food. Plasma and red-cell phospholipid fatty acid profiles were studied. The most relevant results were a lower percentage of docosahexaenoic acid in plasma and red-cell phospholipids (0.91% +/- 0.53% vs 2.88% +/- 1.17% and 2.07% +/- 0.92% vs 3.62% +/- 1.01% (p < 0.001)) and a lower percentage of arachi

Occurrence of the 15-hydroxy derivative of dihomogammalinolenic acid in human platelets and its biological effect.

Various polyunsaturated fatty acids are oxygenated by platelet lipoxygenase at the n - 9 position. The present paper reports that platelets may also oxygenate dihomogammalinolenic acid (20:3(n - 6)) at the n - 6 position, leading to the formation of substantial amounts of 15-OH-8,11,13-20:3 characterized by its ultraviolet spectrum, HPLC and GC-MS analysis. Its formation was inhibited by aspirin and eicosatetraynoic acid, but not by heneicosatetraynoic acid, a specific inhibitor of platelet lipoxygenase. The time-course of its synthesis was very close to that of 12-OH-8,10-17:2 (HHD), the non-

Are protein substitutes available in Italy for infants with inherited metabolic diseases all the same?

INTRODUCTION: Inherited metabolic diseases (IMDs) represent a major clinical challenge, especially during the neonatal and infant periods. They require tailored and long-term nutritional management to ensure proper growth and development. Protein substitutes are essential in the dietary treatment of IMDs, particularly aminoacidopathies, organic acidemias, and urea cycle disorders. In Italy, a variety of PSs is available for infants with IMDs requiring a controlled protein and/or amino acid intake; however, differences in their nutritional composition may impact clinical outcomes. This study ai

Omega-3 fatty acid deficient male rats exhibit abnormal behavioral activation in the forced swim test following chronic fluoxetine treatment: association with altered 5-HT1A and alpha2A adrenergic receptor expression.

Omega-3 fatty acid deficiency during development leads to enduing alterations in central monoamine neurotransmission in rat brain. Here we investigated the effects of omega-3 fatty acid deficiency on behavioral and neurochemical responses to chronic fluoxetine (FLX) treatment. Male rats were fed diets with (CON, n = 34) or without (DEF, n = 30) the omega-3 fatty acid precursor alpha-linolenic acid (ALA) during peri-adolescent development (P21-P90). A subset of CON (n = 14) and DEF (n = 12) rats were administered FLX (10 mg/kg/d) through their drinking water for 30 d beginning on P60. The force

Lacticaseibacillus rhamnosus B6 alleviates metabolic dysfunction-associated fatty liver disease by suppressing intestinal LPS synthesis and regulating lipid metabolism.

INTRODUCTION: Metabolic dysfunction-associated fatty liver disease (MAFLD) has become a global epidemic with an unclear etiology and no effective therapeutic options. Disruption of the gut-liver axis driven by intestinal dysbiosis is closely implicated in MAFLD pathogenesis, making gut microbiota-targeted probiotic interventions promising preventive strategies. METHODS: Lacticaseibacillus rhamnosus B6, a probiotic strain isolated from homemade Bulgarian fermented milk, synthesizes immunomodulatory macromolecules and regulates the intestinal flora. In the present study, we comprehensively inve

Influence of iron, calcium, and zinc on the oxidative stability of omega-3 fatty acids in pea protein-stabilized oil-in-water emulsions.

BACKGROUND: Plant-based emulsified foods fortified with omega-3 fatty acids and minerals are nutritionally attractive, but their formulation is challenging because alpha-linolenic acid (ALA) is highly susceptible to oxidation and some mineral salts may act as pro-oxidants. This study evaluated how iron, zinc, and calcium salts affect pea protein isolate (PPI) properties and ALA oxidative stability in PPI/kappa-carrageenan (KC) gelled emulsions. Treatments included PPI-mineral systems containing ferrous glycinate (FeGly), zinc gluconate (ZnG), calcium gluconate (CaG), calcium lactate (CaL), cal

Lipidomic signatures of heat tolerance in mungbean [Vigna radiata(L.) Wilczek] reveal reduced fatty acid unsaturation.

Heat stress is a major environmental constraint limiting plant growth and productivity, including in mungbean (Vigna radiata). Although physiological, biochemical, and molecular responses of mungbean to elevated temperature have been widely investigated, the role of heat stress-induced leaf lipidome reconfiguration in thermotolerance remains insufficiently understood. In this study, we examined physiological, biochemical, and lipid metabolic responses to heat stress in two contrasting mungbean genotypes: PI425243 (heat-tolerant) and PI223002 (heat-sensitive), grown under non-stress (34/25 °C d

Metabolomic profiling of serum alterations and biomarker discovery in feline hepatic liposis.

Feline hepatic lipidosis (FHL) is a common liver dysfunction caused by metabolic disorders. The objective was to evaluate the metabolic alteration in the cats of FHL and to identify biomarkers that can serve as biomarker for FHL. Differential metabolites in the serum of spontaneous FHL cats (FS, n = 12) and healthy cats (CS group, n = 12) were analyzed using GC/MS metabolomics. Differential metabolites with diagnostic significance were identified through receiver operating characteristic (ROC) curves. The expression level of the differential metabolite 2-hydroxybutyric acid (2-HB) was detected

Fear of cancer recurrence among breast cancer survivors could be controlled by prudent dietary modification with polyunsaturated fatty acids.

BACKGROUND: The pathophysiology of fear of cancer recurrence (FCR), the leading unmet psychological need of cancer survivors, may involve the dysfunctional processing of fear memory. n-3 polyunsaturated fatty acids (PUFAs) have beneficial effects on psychiatric disorders, including depressive disorder and anxiety disorders, and are involved in fear memory processing. We hypothesized that n-3 PUFA composition is associated with FCR in cancer survivors. METHODS: We conducted a cross-sectional study to examine the relationship between n-3 PUFAs and FCR among breast cancer survivors. Adults who h
