Influence of caffeine on the protective activity of gabapentin and topiramate in a mouse model of generalized tonic-clonic seizures.

BACKGROUND: Caffeine may interact with classical antiepileptic drugs (AEDs), reducing their anticonvulsant effects in basic seizure models. The aim of the present study was to ascertain whether intraperitoneal caffeine (acute or chronic for 15 days) could attenuate the anticonvulsant effect of some newer AEDs: gabapentin (GBP) and topiramate (TPM) against electroconvulsions in mice. METHODS: Maximal electroshock (MES)-induced mouse seizure model was used for the estimation of the anticonvulsant activity of TPM whilst the protective activity of GBP was evaluated in the threshold test for maxim

Acetyl-L-carnitine rescues scopolamine-induced memory deficits by restoring insulin-like growth factor II via decreasing p53 oxidation.

Alzheimer's disease (AD) is characterized by the cholinergic neurons loss and impairments of learning and memory. Scopolamine is common used to imitate AD pathological features and also causes an obvious oxidative stress. In this study, we found that intraperitoneal administration of supplementary acetyl-L-carnitine partially reverses the learning and memory defects induced by scopolamine. We also found that acetyl-L-carnitine reverses the impairment of long-term potentiation, dendritic abnormalities, and the impaired recruitment of synaptic protein. The beneficial effects of acetyl-L-carnitin

Autobiographical memory specificity among people with recovered memories of childhood sexual abuse.

Individuals who report to have recovered memories of childhood sexual abuse (CSA) almost by definition believe that these memories were previously inaccessible for them. We examined whether poor autobiographical memory specificity for all kinds of events (i.e., events not necessarily related to CSA) may underlie such impressions of amnesia. Thus, we examined whether people who report recovered memories of CSA (n=44) would exhibit more difficulty retrieving specific autobiographical memories compared to people who never forgot their abuse experiences (continuous memory group; n=42) and people w

Changes in spatial memory and BDNF expression to simultaneous dietary restriction and forced exercise.

Previous literature suggests that learning and memory formation can be influenced by diet and exercise. In the current study, we investigated the combined effects of forced swimming exercise (FSE) and every other day fasting (EODF) on spatial memory formation and on the levels of brain-derived neurotrophic factor (BDNF) in the hippocampus of Wistar male rats. The radial arm water maze (RAWM) paradigm was used to assess changes in learning and memory formation, whereas ELISA assay was used to measure BDNF protein levels. The FSE and/or EODF were simultaneously instituted for 6 weeks. Results sh

Phenytoin ameliorates the memory deficit induced in the young chick by ethanol toxicity in association with thiamine deficiency.

The experiments reported in this series of studies demonstrate that thiamine deficiency, induced pharmacologically by oxythiamine (OT) coupled with the acute administration of ethanol can adversely affect memory processing in the young chick. The interaction between the avitaminosis and ethanol neurotoxicity exerted its effects through the inhibition of the development of the intermediate (ITM) stage of memory (i.e., following 10 min after training), consequently affecting development of the long-term stage of memory and leaving the short-term memory stage intact. The amnesic effect of OT-indu

Gamma (60 Hz) auditory stimulation improves intrusions but not recall and working memory in healthy adults.

Gamma-band (> 30 Hz) brain oscillations (γ) play a crucial role in memory and long-term potentiation, and their disruptions have been consistently documented in patients with Alzheimer's Disease (AD). Gamma-band oscillation entrainment through 60 Hz transcranial alternating stimulation (tACS) and 40 Hz tACS/sensory stimulation has been shown to enhance memory performance in healthy adults and patients with AD, respectively. However, the impact of gamma auditory stimulation on healthy adults' memory remains uncertain. In this balanced crossover study, 36 healthy subjects (27 Females) underwent

Accelerated forgetting and verbal memory consolidation process in idiopathic nondement Parkinson's disease.

OBJECTIVE: Episodic memory impairment and underlying pathophysiology in Parkinson's Disease (PD) is poorly investigated. Formerly, it was thought to be a secondary effect of impairment in fronto-striatal circuit. However, recent studies hypothesized that there is a dual progression of PD and memory loss is possibly related to posterior cortex rather than frontal. To understand the impairment, underlying mechanisms should be investigated. Although consolidation is one of these mechanisms consolidation phase of episodic memory in PD was not investigated yet. Recently accelerated long term forget

Using remote, digital, multi-day testing to characterize long-term forgetting in cognitively unimpaired older adults.

INTRODUCTION: Accelerated long-term forgetting (LTF) might be an early marker of subtle memory changes in older adults at risk for Alzheimer's disease (AD). We leveraged remote, multi-day digital testing to characterize LTF in older adults and investigated its association with initial learning and AD imaging biomarkers. METHODS: One hundred four cognitively unimpaired older adults completed a face-name memory task for seven consecutive days and were asked to recognize face-name pairs 1 week later. LTF was computed as the number of correctly identified stimuli divided by a participant's maximu

Targeting tissue immune memory in chronic-remitting autoimmune kidney diseases: current concepts and future therapies.

Immune-mediated kidney diseases have a substantial disease burden and frequent disease relapses significantly contribute to the high morbidity. Relapsing disease courses are associated with poor outcome and accelerated progression to end-stage renal disease. The pathomechanisms leading to relapse are diverse and only incompletely understood. Both adaptive and innate immune cells are implicated in the immune memory of these diseases. In addition to antigen-dependent mechanisms of T and B cells, persistent epigenetic and metabolic changes in adaptive and innate immune cells may sustain immunolog

Development and persistence of memory immune recall response following SARS-CoV-2 infection in children.

SARS-CoV-2 infection is generally mild in children, yet the magnitude and durability of post-infection immune memory remain poorly defined. In this longitudinal study, humoral and cellular immune responses were comprehensively analyzed in 10 SARS-CoV-2-infected children followed for 12 months. Antigen-specific B and T cells were stimulated ex vivo to assess their frequency, persistence, phenotype, and magnitude of recall responses. Natural infection induced robust and long-lasting antigen-specific memory CD4+ T cells that proliferated upon reactivation, exhibited protective Th1 and Th17 polari

Short-Lived Immunity After 17DD Yellow Fever Single Dose Indicates That Booster Vaccination May Be Required to Guarantee Protective Immunity in Children.

The Yellow Fever (YF) vaccination is recommended for people living in endemic areas and represents the most effective strategy to reduce the risk of infection. Previous studies have warned that booster regimens should be considered to guarantee the long-term persistence of 17DD-YF-specific memory components in adults living in areas with YF-virus circulation. Considering the lower seroconversion rates observed in children (9-12 months of age) as compared to adults, this study was designed in order to access the duration of immunity in single-dose vaccinated children in a 10-years cross-section

A preliminary study of the different memory systems in neurofibromatosis type 1: What is impaired and what is spared?

BACKGROUND: Since 1990's, the cognitive profile of children with a neurofibromatosis type 1 (NF1) has been refined by many different studies. Children with NF1 may exhibit a variety of cognitive dysfunctions. Memory difficulties have been reported, but the results are contradictory and, compared to other cognitive functions, memory has been less evaluated. OBJECTIVE: The aim of this study was to investigate the memory profile in NF1 with a particular population, children with NF1 without attention deficit hyperactivity disorder (ADHD). METHODS: Eighteen children with NF1 without ADHD and eig

Transfer Learning With Simulated and Recorded Data Improves Predictions of Lateral Pinch Thumb-Tip Forces.

Machine learning is a powerful tool for modeling complex movements. However, the high cost of acquiring biomechanical data often limits observations and therefore the generalizability of machine learning models. Meanwhile, there is untapped utility in presently published musculoskeletal simulations for informing these predictions. This work evaluated whether a transfer learning model trained on both musculoskeletal simulation and recorded data outperforms a direct learning model trained on recorded experimental data alone. For the transfer learning model, we leveraged a simulation dataset with

Explainable AI for gait speed analysis from multimodal data fusion.

Gait speed analysis is vital for applications in healthcare, rehabilitation, human-robot interaction, and autonomous systems, necessitating robust methods to address its complexity. This study introduces an advanced framework for gait speed classification through multimodal data fusion and deep learning, optimized using Layer-wise Relevance Propagation (LRP). Utilizing a publicly available 4 datasets of 50 injury-free adults walking at varying speeds, we integrate data from full-body motion capture, electromyography (EMG), and force plates to capture detailed gait dynamics. We propose two nove

Pyrazole-derived TRPC3 antagonist ameliorates synaptic dysfunctions and memory deficits in Alzheimer's disease models.

Transient receptor potential canonical (TRPC) channels are widely expressed in the brain; however, their precise roles in neurodegenerative diseases, such as Alzheimer's disease (AD), remain elusive. We found that TRPC3 expression is upregulated in excitatory neurons of brains with AD. We tested a selective inhibitor (JW-65) for TRPC3 over TRPC6 to investigate the potentially distinct role of TRPC3 in AD. JW-65 treatment significantly restored impaired synaptic plasticity and learning memory in acute and chronic experimental AD models. JW-65 treatment of late symptomatic 5XFAD transgenic mice

Multimodal neuroimaging-based deep learning framework for pattern analysis and early prediction of neurodegenerative diseases.

Neurodegenerative diseases, such as Mild Cognitive Impairment (MCI) and Alzheimer's, pose significant challenges due to their progressive nature and late diagnosis. Early detection remains difficult, particularly when using conventional machine learning approaches that fail to capture complex spatial and temporal patterns in multimodal clinical data. Motivated by the need for accurate, scalable, and clinically applicable diagnostic tools, this study proposes a hybrid deep learning framework combining Convolutional Neural Networks (CNN) with Optimized Spatial-Temporal Bidirectional Gated LSTM (

Late-term moderate prenatal alcohol exposure impairs tactile, but not spatial, discrimination in a T-maze continuous performance task in juvenile rats.

Existing maze apparatuses used in rodents often exclusively assess spatial discriminability as a means to evaluate learning impairments. Spatial learning in such paradigms is reportedly spared by moderate prenatal alcohol exposure in rats, suggesting that spatial reinforcement alone is insufficient to delineate executive dysfunction, which consistently manifests in humans prenatally-exposed to alcohol. To address this, we designed a single-session continuous performance task in the T-maze apparatus that requires rats to discriminate within and between simultaneously-presented spatial (left or

Overexpression of LIMK1 in hippocampal excitatory neurons improves synaptic plasticity and social recognition memory in APP/PS1 mice.

Accumulating evidence indicates that the actin regulator cofilin is overactivated in Alzheimer's Disease (AD), but whether this abnormality contributes to synaptic and cognitive impairments in AD is unclear. In addition, the brain region and cell types involved remain unknown. In this study, we specifically manipulate LIMK1, the key protein kinase that phosphorylates and inactivates cofilin, in the hippocampus of APP/PS1 transgenic mice. Using local injections of the AAV virus containing LIMK1 under the control of the CaMKIIα promoter, we show that expression of LIMK1 in hippocampal excitatory

Stroke promotes the development of brain atrophy and delayed cell death in hypertensive rats.

Post-stroke cognitive impairment (PSCI) is a major source of disability, affecting up to two thirds of stroke survivors with no available therapeutic options. The condition remains understudied in preclinical models due to its delayed presentation. Although hypertension is a leading risk factor for dementia, how ischemic stroke contributes to this neurodegenerative condition is unknown. In this study, we used a model of hypertension to study the development of PSCI and its mechanisms. Spontaneously hypertensive rats (SHR) were compared to normotensive rats and were subjected to 1-h middle cere
