Dietary exclusion of fructose and lactose after positive breath tests improved rapid-transit constipation in children.

AIMS: Exclusion of fermentable oligosaccharides, disaccharides, monosaccharides, and polyols (FODMAPs) from the diet is effective in alleviating symptoms of irritable bowel syndrome (IBS) in adults. Rapid-transit constipation (RTC) is a recently discovered subset of chronic constipation and has been linked to food intolerance. The aim of this study was to audit the effect of specific FODMAP elimination diets in children with RTC. METHODS: This was an audit of children presenting to a tertiary children's hospital surgeon with refractory chronic constipation who had rapid transit in the proxima…

A Systematic Review of the Effects of Polyols on Gastrointestinal Health and Irritable Bowel Syndrome.

Polyols are sugar alcohols found in certain fruits, vegetables, and sugar-free sweeteners. They make up a component of the diet low in fermentable oligosaccharides, disaccharides, monosaccharides, and polyols, which is gaining popularity in the treatment of patients with irritable bowel syndrome (IBS). We conducted a systematic review to evaluate the effects of polyols on the gastrointestinal tract in healthy men and women and in patients with IBS. Utilizing PubMed, Ovid, and Embase databases, we conducted a search on individual polyols and each of these terms: fermentation, absorption, motili…

High throughput quantification of capsular polysaccharides for multivalent vaccines using precipitation with a cationic surfactant.

The increasing requirement for multivalent vaccines containing diverse capsular polysaccharides has created an unmet need for a fast and str … velengths that resolve scattering light. Carbohydrates comprised of repeating units of one to seven monosaccharides with phosphodiester groups, uronic acids, and sialic acids all reacted strongly and there does not appear to be specificity with respect to the particular anionic moiety. The assay is compatible with an array of common buffers across a pH range of 3 …

A defect in exodegradative pathways provides insight into endodegradation of heparan and dermatan sulfates.

Within cells, dermatan sulfate (DS) and heparan sulfate (HS) are degraded in two steps. The initial endohydrolysis of these polysaccharides is followed by the sequential action of lysosomal exoenzymes to reduce the resulting oligosaccharides to monosaccharides and inorganic sulfate. Mucopolysaccharidosis (MPS) type II is a lysosomal storage disorder caused by a deficiency of the exoenzyme iduronate-2-sulfatase (I2S). Consequently, partially degraded fragments of DS and HS have been shown to accumulate in the lysosomes of affected cells and are excreted in the urine. Di- to hexadecasaccharides,…

Radioactive fucose as a tool for studying glycoprotein secretion.

The efficiency and reliability of radioactive fucose as a specific label for newly synthesized glycoproteins were investigated. Young adult … led with [3H]-fucose was always much higher than that of the proteins labeled with any of the other monosaccharides or any of the amino acids. There was a good correlation between the specific activity of the proteins labeled by any of the above precursors and the density of the vitreous protein bands detected by fluorography. This was also true for the silver gr …

Basic fibroblast growth factor controls the expression and molecular structure of heparan sulfate in corneal endothelial cells.

Cultured bovine corneal endothelial cells express 5-8 ng basic fibroblast growth factor (bFGF)/mg cell protein and distribute it between the … erated by heparitinase I degradation. The proportion of [35S]oligosaccharides with greater than six monosaccharides decreases on account of disaccharides and tetrasaccharides under the influence of bFGF. Pretreatment of bFGF with neutralizing antibodies against bFGF abolishes its biological activity. The results suggest a bFGF-dependent change in the rate of synt …

Preliminary Structural Characterization of an Anti-Inflammatory Polysaccharide from Lysimachia christinae and Its Preventive Effect on Lipopolysaccharide-Induced Acute Liver Injury in Mice.

An acidic heteropolysaccharide, named LCP-90-3, with a relative molecular weight of 14.02 kDa, was isolated and purified from the traditional heat-clearing Chinese medicine, Lysimachia christinae Hance. The monosaccharide residue analysis indicated that LCP-90-3 was composed of glucuronic acid, mannose, and arabinose in a molar ratio of 79.2: 3.6: 17.2. Linkage analysis further indicated that LCP-90-3 was mainly composed of 1,4-GlcpA, 1,4,6-GlcpA, 1,2-Araf, T-Manp, 1,3-Araf, and 1,4-Manp residues. Furthermore, LCP-90-3 demonstrated potent anti-inflammatory activity by significantly inhibiting

Analytical characterisation of yeast cell walls by lectin-binding flow cytometry and extraction-based polysaccharide analysis.

Accurate profiling of yeast cell-wall polysaccharides (PS) remains analytically challenging because different methods interrogate different levels of cell-wall organisation. In this study, lectin-binding flow cytometry and extraction-based chemical analysis were comparatively evaluated across 10 representative yeast strains. Flow cytometry revealed marked strain-dependent variation in forward scatter (FSC), side scatter (SSC), and median of lectin fluorescence intensity (MFI), whereas extraction-based analysis showed substantial differences in PS yield, monosaccharide composition, and molecula

Differential Effects of Commercial Food-Grade Carrageenan Preparations on DSS-Induced Colitis and Gut Microbiota in Mice.

Carrageenan (CGN) is widely used in processed foods and is typically supplied as a commercial preparation blended with other hydrocolloids to improve gelling properties, rather than as a single purified polymer. However, safety evaluations and mechanistic studies have largely focused on CGN in isolation; as a result, the biological effects of commercial CGN preparations (CGNPs) under realistic exposure conditions are still insufficiently characterized. In this study, the structural characteristics of three commercial food-grade CGNPs intended for meat products, soft sweets, and jelly were inve

Orchestrating immunity and beyond: polysaccharides as multi-target regulators of inflammatory networks via gut microbiota and structure-dependent pathways.

Natural polysaccharides (NPs), as key bioactive constituents of many plants, have garnered significant attention due to their immunomodulatory properties and high safety profile. This review elucidates the molecular mechanisms underlying the immunomodulatory effects of NPs, their associated physiological outcomes, and the core structure-activity relationships. Mechanistically, NPs exert anti-inflammatory or immunostimulatory functions by modulating immune organ indices, activating or suppressing various immune cells (e.g., macrophages, dendritic cells, and NK cells), and regulating pivotal imm

Synthesis, Biological Evaluation, and Computational Studies of Phenolic N-Acetylglucosamine Glycosides as α-Glucosidase Inhibitors.

Type 2 diabetes mellitus (T2DM) is one of the most prevalent chronic metabolic diseases, and inhibition of α-glucosidase activity represents an effective therapeutic strategy. Chitin is the most abundant renewable polysaccharide in the ocean, with its monosaccharide being N-acetylglucosamine (NAG). To evaluate the potential of NAG glycosides as novel α-glucosidase inhibitors, three common phenolic compounds were modified via NAG glycosylation. Their inhibitory activities were assessed at both the enzymatic and cellular levels. In addition, density functional theory (DFT), molecular dynamics (M

Structure-function paradigms of natural polysaccharides in hepatocellular carcinoma therapy.

Hepatocellular carcinoma (HCC) is a major cause of cancer mortality worldwide, and its limited response to conventional chemotherapeutics underscores the need for safer, multifunctional approaches. Natural polysaccharides, complex carbohydrates derived from plants, fungi, algae, animals, and bacteria, represent a structurally diverse class of bioactive macromolecules that combine therapeutic and material properties. Their monosaccharide composition, linkage types, branching patterns, molecular weights, and charge densities determine receptor recognition, intracellular trafficking, and signalin

Identification of a commercial Xylan as a microfold cell-targeting polysaccharide in vivo and in vitro.

Microfold cells (M cells) in the gut serve as a key gateway to enhance the potency of oral vaccines. Although natural polysaccharides show promise as carriers, their development is limited by inherent drawbacks such as scarce availability and lack of intrinsic targeting capacity. Therefore, it is essential to identify suitable materials from well-defined, commercially available polysaccharide standards. In this study, a library of well-defined, high-purity commercial polysaccharides was screened, and identified Xylan as a lead candidate with superior M cell transport efficiency in an in vitro

Methods for Investigating Siglec-Ganglioside Interactions.

Siglecs are a family of cell surface receptors generally found on immune cells, which are important in many cellular processes such as cell-adhesion, cell-signaling, and internalization of extracellular particles. While all these smaller functions play a part in larger and different physiological roles, one aspect they share is that they are initiated by the binding of a Siglec to its ligand. Siglec ligands are cell-surface carbohydrates appended to proteins and lipids that terminate with the monosaccharide sialic acid. The functions of a Siglec are directly linked to the binding of its ligand
