TACTIC: A transfer learning framework to predict drug interactions in emerging pathogens.

Machine learning (ML) is necessary to efficiently identify potent drug combinations within a large candidate space to combat drug resistance. However, existing ML approaches cannot be applied to emerging and understudied pathogens with limited training data. Hence, we developed a computational framework (TACTIC [transfer-learning and cross-species training to infer combination therapies]) to train ML models on data from multiple model bacteria. TACTIC was built using 2,965 drug interactions from 12 bacterial strains and outperformed traditional ML models in predicting drug interaction outcomes

New Perspectives on Antimicrobial Agents: Omadacycline for community-acquired pneumonia, skin and soft tissue infections, and nontuberculous mycobacteria (focus on M. abscessus).

Omadacycline is a novel antimicrobial belonging to the tetracycline class. It has the ability to evade both efflux and ribosomal methylation types of resistance and therefore has an expanded spectrum compared to other tetracycline agents. Omadacycline is active against a number of multidrug-resistant bacteria, including macrolide and doxycycline-resistant Streptococcus pneumoniae, methicillin-resistant Staphylococcus aureus (MRSA), vancomycin-resistant Enterococcus, and several enteric gram-negative bacilli. It also has activity against many nontuberculous mycobacterium (NTM) species. It is av

Clinical Efficacy of Serum Antiglycopeptidolipid Core IgA Antibody Test for Screening Nontuberculous Mycobacterial Pulmonary Disease in Bronchiectasis: A European Multicenter Cohort Study.

BACKGROUND: The serum antiglycopeptidolipid core IgA antibody test has been proposed as a diagnostic tool for Mycobacterium avium complex pulmonary diseases. Cross-reactivity with other nontuberculous mycobacteria (NTM), including Mycobacterium abscessus, indicates that it may have a role as a broader screening test for nontuberculous mycobacterial pulmonary disease (NTM-PD). NTM-PD is believed to be underdiagnosed in patients with bronchiectasis. RESEARCH QUESTION: Can the serum antiglycopeptidolipid core IgA antibody test be used to screen for NTM-PD in bronchiectasis? STUDY DESIGN AND MET

Functional analysis of type VII secretion system links to host immune evasion mechanism in Mycobacterium tuberculosis.

Mycobacterium tuberculosis causes tuberculosis, an infectious disease; this acid-fast bacillus has various functions that enable it to survive within the host. Importantly, the type VII secretion system plays a vital role in host immune evasion. However, the early secretory antigenic target secretion system (ESX) component is crucial for mycobacteria survival, plays a significant role in bypassing the host immune response, and is linked to the prognosis of the disease. The review aims to analyze the ESX-associated genes' functions in defence mechanisms against host immune response. There are f

Nontuberculous mycobacteria skin infections associated with tattoos.

Nontuberculous mycobacteria (NTM) skin infections associated with tattoos and permanent makeup (PMU) have emerged as an important public health concern alongside the growing popularity of these procedures. Since the first reported case in 2003, increasing numbers have been documented worldwide. To better understand their epidemiology and clinical characteristics, we conducted a review of published cases. As of December 2024, 62 articles described 66 tattoo-related NTM skin infection cases, involving 310 individuals across 15 countries. Notably, 25 cases were outbreaks, accounting for 262 affec

Chemical Sterilization of Lipoplasty Cannula and Nontuberculous Mycobacteria Disinfection: An Experimental Study.

BACKGROUND: Nontuberculous mycobacteria are commonly found pathogens; however, skin and soft tissue infections due to nontuberculous mycobacteria are often associated with surgical procedures, particularly after lipoplasty. Although nontuberculous mycobacteria are resistant to some chemical disinfectants, glutaraldehyde, peracetic acid, povidone iodine, alcohol, and chlorine are still used for the sterilization of medical instruments. This study investigated the efficacy of various disinfectants in a fatty environment with adipose and a bloody environment without adipose. In addition, this stu

Comparative evaluation of polymerase chain reaction and restriction enzyme analysis: two amplified targets, hsp65 and rpoB, for identification of cultured mycobacteria.

The increasing incidence of tuberculosis and other mycobacterial infections due to AIDS epidemic resulted in the need of rapid and accurate identification of isolated mycobacteria. The correct identification result leads to the selection of an appropriate therapeutic regimen. Polymerase chain reaction and restriction enzyme analysis (PCR-REA) has been developed since 1992 and used as the rapid method for identifying mycobacteria. Several genes or sequences have been used as an amplified target for PCR-REA. The present study aims to evaluate the potential use of PCR-REA of gene-encoding heat sh

Nontuberculous mycobacterial pulmonary disease: Epidemiology, prognostic determinants, and implications for treatment decision.

The global burden of nontuberculous mycobacterial pulmonary disease (NTM-PD) continues to increase, particularly in East Asia, including Japan. Although epidemiological understanding has expanded, the clinical course of NTM-PD remains highly heterogeneous and is influenced by host immunity, structural lung abnormalities, comorbidities, and environmental factors. Determining the optimal timing for initiating treatment remains challenging, particularly in cases of non-cavitary or mild disease. Recent evidence has elucidated several key prognostic determinants, including cavitary disease, infecti

Interferon-γ-response to mycobacterial glycopeptidolipids in patients with pulmonary non-tuberculous mycobacterial infection.

The understanding of host responses to pulmonary infection with non-tuberculous mycobacteria (NTM) is incomplete and immunodiagnostic tools are not established. Unlike mycobacteria of the Mycobacterium tuberculosis complex, several NTM express glycopeptidolipids (GPLs) in their cell wall. Here, we investigate the interferon-γ response to mycobacterial GPLs and its diagnostic use in patients with pulmonary NTM infection. M. avium-derived GPLs were used for in vitro stimulation of peripheral blood mononuclear cells in an ELISpot assay of patients with pulmonary NTM infection and 3 control groups
