Spreading resistance to clinically used antimalarial drugs increases the need for identification of new drug targets. Here, we screened 25 antimalarials with known and unknown mode of action to validate old and find new drug targets in P. falciparum. Combining experimental approach by proteome-wide cellular thermal shift assay and computational filtering by molecular docking, we found the drugs to bind to previously known drug targets, validated ACS10 as the target of MMV665915 and discovered new drug targets. Furthermore, we updated the experimental and analytical MS-CETSA pipeline with the i

The mass rearing of insects for food and feed is increasing significantly, but it presents challenges in detecting and characterizing pathogens which may cause severe economic losses. Much attention is placed on detecting viruses, bacteria and fungi; however, other significant entomopathogens remain overlooked, such as protist and microsporidian parasites. This study employed Nanopore sequencing to explore and phylogenetically analyse microsporidian and protist parasites in reared insects. Using group-specific primers and amplicon pooling strategies, we successfully amplified and sequenced a k…

Apicomplexans are a large group of protists, including several species of major medical importance, most notably those which cause malaria and toxoplasmosis in humans. They are obligate intracellular symbionts that have an interesting evolutionary history, having evolved from a free-living, phototrophic ancestor. Despite the extensive research interest in this group, novel apicomplexans with important ecological roles are still being discovered, particularly from the marine environment. These often-overlooked microbes infect a myriad of marine organisms, and recent research on this clade has e

Apicomplexan parasites establish intracellular infections that profoundly alter host cell physiology and elicit complex immune responses. The long-standing coevolution between these parasites and vertebrate hosts has resulted in extensive overlap between parasite and host metabolic pathways, limiting the feasibility of conventional parasite-centered therapeutic approaches. Increasing evidence indicates that host-derived signals generated during infection play a decisive role in shaping parasite survival and dissemination. Among these signals, extracellular nucleotides released in response to c…

Many pathogenic organisms, including malaria parasites, produce specialized life stages for within-host multiplication (asexual) versus onward transmission (sexual reproduction). Restrained investment into transmission stage production-by allowing faster multiplication-is predicted to curtail the lifespan of infection via faster host recovery or mortality, a classic tradeoff between the rate and duration of transmission. In contrast, under a reproduction-survival tradeoff, restraining investment into reproduction should extend survival (for parasites, infection duration). To distinguish betwee…

Poultry production plays a vital role in ensuring food and nutritional security in Bangladesh by supplying affordable sources of animal protein, namely, meat and eggs, and generating income for millions. Due to their wide availability, low cost, and absence of religious restrictions, poultry products have become a major source of animal protein in the country. This narrative review examines the profiles of ectoparasites, endoparasites, and haemoparasites commonly found in Bangladeshi poultry, all of which contribute to reduced growth performance and productivity. A comprehensive search of peer…

Aquarium corals have emerged as a relevant sector not only from an economic perspective but also as a tool for ex situ conservation. However, coral pests and parasites, such as flatworms Prosthiostomum acroporae, nudibranchs Phestilla spp., parasitic copepods Tegastes acroporanus, epizoic acoelomorphs Waminoa spp., and histophagous ciliates Philaster spp., pose significant biological and economic risks in captive systems. This review synthesizes current knowledge on major pest groups affecting ornamental corals, emphasizing their life cycles, host specificity, and control strategies. Chemical …

Some parasitic infections promote or inhibit vascular growth in their hosts to increase parasite survival through immune evasion and tissue dissemination. This review focuses on how the most prevalent protozoan and helminth parasites in humans, such as Plasmodium, Toxoplasma, Leishmania, Trypanosoma, Entamoeba, Schistosoma, and Taenia, manipulate angiogenic pathways for their own benefit. This knowledge reveals that angiogenesis is central to the pathophysiology and therapeutic targeting of parasitic diseases. Importantly, parasites and/or their excretory/secretory factors, which modulate vasc…

BACKGROUND: Intestinal parasitic infections affect more than 1·5 billion people globally, leading to severe health consequences such as malnutrition, anemia, diarrhea, and impaired cognitive development.
METHODOLOGY/PRINCIPAL FINDINGS: Samples were collected from 3,872 individuals (all ages and both sexes) across 31 rural communities in Madagascar between 2013 and 2017, representing diverse ecological and socioeconomic regions. Intestinal parasite prevalence was assessed by fecal microscopy. Bayesian multilevel logistic regression models were used to estimate overall and regional prevalences

Transmission of malaria by blood transfusion is rare in non-endemic countries but can lead to serious complications in blood recipients. Increasing travel to and immigration from regions at risk for tropical diseases poses a challenge to blood donation services, which are striving to reduce the number of blood donor deferrals while ensuring a high level of blood safety. National guidelines of the Blood Transfusion Service of the Swiss Red Cross demand that donors at risk are serologically tested for malaria antibodies. Here, we summarize the numbers of malaria tests performed and the results o

INTRODUCTION: Domestic ducks are important for meat, egg and livelihood production in Nepal; however, gastrointestinal parasites (GIPs) remain a major health constraint affecting productivity and posing zoonotic risks.
AIMS: This cross-sectional study aimed to determine the prevalence and diversity of GIPs in ducks raised on smallholder farms in Kathmandu, Nepal.
METHODS: Fresh faecal samples (N = 130) were collected randomly using non-invasive methods, and copromicroscopy was performed after direct wet-mount and acid-fast staining techniques.
RESULTS: A total of 51 samples (39.2%) tested p…

The synchronization of Plasmodium parasites as they replicate within red blood cells of their vertebrate host remains largely unexplored. Understanding this synchronization could reveal how parasites optimize their lifecycle to maximize transmission, evade the immune response and maximize energy acquisition. Rhythmic replication fulfils some criteria of an endogenous oscillator with time of day cues potentially provided by temperature, oxygen levels, hormones and/or nutrient availability. Recent research on a rodent malaria model has highlighted that rhythms associated with the host's feeding/…

Blood-stage Plasmodium falciparum parasites rely on a non-photosynthetic plastid, the apicoplast, for survival, making it an attractive target for antimalarial intervention. Like the mitochondrion, the apicoplast cannot be generated de novo and must be inherited by daughter parasites during cell division. This inheritance relies on coordinated apicoplast positioning and fission, but the molecular mechanisms controlling these processes remain poorly understood. Here, we identify a previously uncharacterized P. falciparum protein (Pf3D7_0613600), which we name PfAnchor, as a key regulator of api…

Hundreds of thousands die annually from malaria caused by Plasmodium falciparum (Pf), with the emergence of drug-resistant parasites hindering eradication efforts. Antimicrobial peptides (AMPs) are known for their ability to disrupt pathogen membranes without targeting specific receptors, thereby reducing the chance of drug resistance. However, their effectiveness and the biophysical mechanisms by which they target the intracellular parasite remain unexplored. Here, by using native and synthetic AMPs, we discovered a selective mechanism that underlies the antimalarial activity. Remarkably, the…
