Selection of two pronuclei zygotes before electroporation does not improve gene editing efficiency or reduce mosaicism in porcine embryos despite optimized timing and Hoechst staining.

BACKGROUND AND AIM: Electroporation-mediated delivery of Clustered-Regularly Interspaced Short Palindromic Repeats (CRISPR)/CRISPR-associated protein 9 components is an effective approach for genome editing in porcine embryos; however, mosaicism remains a major limitation. Selecting normally fertilized zygotes with two pronuclei (2PN) before electroporation may improve editing uniformity and reduce mosaicism. Therefore, this study investigated the effects of 2PN selection, optimized timing of pronuclear observation, and Hoechst 33342 staining conditions on embryo development and genome editing

Effects of gestational inflammation on age-related cognitive decline and hippocampal Gdnf-GFRα1 levels in F1 and F2 generations of CD-1 Mice.

BACKGROUND: It has been reported that age-associated cognitive decline (AACD) accelerated by maternal lipopolysaccharide (LPS) insult during late pregnancy can be transmitted to the second generation in a sex-specificity manner. In turn, recent studies indicated that glial cell line-derived neurotrophic factor (GDNF) and its cognate receptor (GFRα1) are critical for normal cognitive function. Based on this evidence, we aimed to explore whether Gdnf-GFRα1 expression contributes to cognitive decline in the F1 and F2 generations of mouse dams exposed to lipopolysaccharide (LPS) during late gestat

Geographic Variation and Factors Associated with Female Genital Mutilation among Reproductive Age Women in Ethiopia: A National Population Based Survey.

BACKGROUND: Female genital mutilation (FGM) is a common traditional practice in developing nations including Ethiopia. It poses complex and serious long-term health risks for women and girls and can lead to death. In Ethiopia, the geographic distribution and factors associated with FGM practices are poorly understood. Therefore, we assessed the spatial distribution and factors associated with FGM among reproductive age women in the country. METHOD: We used population based national representative surveys. Data from two (2000 and 2005) Ethiopian demographic and health surveys (EDHS) were used

Associations of prenatal exposure to maternal autoimmune disorders with a wide spectrum of psychiatric and neurodevelopmental disorders in offspring-a nationwide cohort study.

STUDY QUESTION: Are children exposed in utero to various maternal autoimmune disorders (ADs) and autoinflammatory disorders (AIDs) at greater risk for developing any of a wide range of psychiatric or neurodevelopmental disorders? SUMMARY ANSWER: Prenatal exposure to maternal ADs and AIDs, but for those of the nervous system, was at modest effect sizes associated with a number of primarily early onset neurodevelopmental disorders. WHAT IS KNOWN ALREADY: Prenatal exposure to maternal immune activation and certain ADs or AIDs has been associated with psychiatric and neurodevelopmental disorders

The Role of Chromosome X in Intraocular Pressure Variation and Sex-Specific Effects.

IMPORTANCE: The association of poor family functioning, a potent stressor, with child behavior is potentially long term and relevant for a person's well-being later in life. Whether changes in brain development underlie the associations with preadolescent behavior and help identify periods of vulnerability is unclear. OBJECTIVE: To assess the associations of poor family functioning from pregnancy onward with cortical, white matter, and subcortical volumes, and to examine the extent to which, in particular, hippocampal volume mediates the association of prenatal parental environmental exposure

Polygenic Scores and Environmental Factors in Psychiatric Disorders: Gene-Environment Interaction Analyses Using the iPSYCH Study.

BACKGROUND: Psychiatric disorders represent a significant global health burden with complex etiologies involving both genetic and environmental factors. However, while the main effects of genetic and environmental factors are frequently studied, their interplay in shaping psychiatric disorder risk remains poorly understood. This study investigates the interaction between polygenic scores (PGS) as proxies for genetic risk and environmental factors in the risk of psychiatric disorders. METHOD: This study utilized the iPSYCH case-cohort sample (n = 141,265). Environmental factors, including regi

Delay in blood sampling for routine newborn screening is associated with increased risk of schizophrenia.

BACKGROUND: The Danish Neonatal Screening Biobank, containing dried blood spot samples from all newborn in Denmark, is a unique source of data that can be utilized for analyses of genetic and environmental exposures related to schizophrenia and other mental disorders. In previous analyses, we have found that early and late blood sampling, compared to sampling at day 5, was associated with increased risk of schizophrenia. As delay in sampling of blood for neonatal screening cannot in itself influence the risk of schizophrenia, it must be seen as a proxy for unknown underlying causes responsible

Heterogeneity in semen-mediated tolerogenic responses: defining immunological determinants of reproductive compatibility.

STUDY QUESTION: Do particular pairs of individuals modeling sexual pairs exhibit differences in the induction of semen-mediated long-lasting tolerance in support of pregnancy? SUMMARY ANSWER: Yes, factors from both sexes contribute to the variability in the induction of long-lived regulatory T cells (Tregs) in response to semen, with factors related to the recipient seeming to slightly outweigh the factors related to the semen donor. WHAT IS KNOWN ALREADY: Semen contains multiple immunomodulatory factors that can influence reproductive tract immunity in the recipient, promote tolerance to an

Prevalence of Childhood Stunting and Its Associated Factors in Bangladesh: Quantile Regression Modeling Approach.

BACKGROUND: Stunting is a key public health concern among under-five children in Bangladesh. It affects approximately one-quarter of children under 5 years of age, which is higher than global averages and insufficient to meet the goal of the World Health Organization (WHO) for reducing stunting to 15%. So, further research is needed to better understand the current dynamics of stunting and to develop pragmatic policies based on the most recent nationwide data which will provide valuable insights into the factors influencing stunting in Bangladesh. This study investigates the prevalence and ris

Is the lack of prior exposure to sperm antigens associated with worse neonatal and maternal outcomes? A 10-year single-center experience comparing ICSI-TESE pregnancies to ICSI pregnancies.

BACKGROUND: Nowadays, pathogenesis of preeclampsia (PE) is still unknown. Among the different etiological hypotheses, some authors proposed that it might be because of an abnormal immunologic response to a foreign fetal antigen derived from the father's spermatozoa. Indeed, the fetus is considered a semi-allograft, being one half paternally derived in its antigenicity, and the first pathogenic insult of PE may be an abnormal maternal immune response toward this semi-allogenic implant. In the context of artificial reproductive techniques, it has been shown that the use of donor and surgically r

Paternal high protein diet modulates body composition, insulin sensitivity, epigenetics, and gut microbiota intergenerationally in rats.

Mounting evidence demonstrates that paternal diet programs offspring metabolism. However, the contribution of a pre-conception paternal high protein (HP) diet to offspring metabolism, gut microbiota, and epigenetic changes remains unclear. Here we show that paternal HP intake in Sprague Dawley rats programs protective metabolic outcomes in offspring. Compared to paternal high fat/sucrose (HF/S), HP diet improved body composition and insulin sensitivity and improved circulating satiety hormones and cecal short-chain fatty acids compared to HF/S and control diet (P < .05). Further, using 16S rRN

Paternal Expressed Gene 10 (PEG10) is decreased in early-onset preeclampsia.

BACKGROUND: Preeclampsia is a severe complication of pregnancy which is attributed to placental dysfunction. The retrotransposon, Paternal Expressed Gene 10 (PEG10) harbours critical placental functions pertaining to placental trophoblast cells. Limited evidence exists on whether PEG10 is involved in preeclampsia pathogenesis. This study characterised the expression and regulation of PEG10 in placentas from patients with early-onset preeclampsia compared to gestation-matched controls. METHODS: PEG10 expression was measured in plasma and placentas collected from patients with early-onset preec

The methylation levels of the H19 differentially methylated region in human umbilical cords reflect newborn parameters and changes by maternal environmental factors during early pregnancy.

H19 is a tumor-suppressor gene, and changes in the methylation of the H19-differential methylation region (H19-DMR) are related to human health. However, little is known about the factors that regulate the methylation levels of H19-DMR. Several recent studies have shown that maternal environmental factors during pregnancy, such as smoking, drinking, chemical exposure, and nutrient intake, can alter the methylation levels of several genes in fetal tissues. In this study, we examined the effects of maternal factors on changes in the methylation levels of H19-DMR in the human umbilical cord (UC),

Passively inhaled tobacco smoke: a challenge to toxicology and preventive medicine.

The difficulties in defining the exposure of a passive smoker might explain the controversial results regarding an association between passive smoking on one hand and lung cancer, tumors of all sites and ischemic heart diseases on the other. The plausibility of these epidemiological observations will be discussed in the light of analytical, toxicological, biochemical and oncological data. The minute amounts of nicotine and particulate matter, even the much higher concentrations of volatile substances, such as nitrosamines, NOx, acroleine and formaldehyde, present in diluted sidestream compared
