The circadian clock component OsRVE6a orchestrates daytime SA-JA signaling crosstalk to enhance rice defenses.

Salicylic acid (SA) and jasmonic acid (JA) are key plant defense hormones that typically exhibit antagonistic interactions, with JA levels peaking during the day and SA at night. However, how plants sustain SA-mediated immunity during daylight when JA defenses dominate remains unclear. Here, we identify the rice oscillator gene OsRVE6aas a central regulator enabling simultaneous activation of SA and JA pathways during daytime biotic stresses. Daytime feeding by the small brown planthopper (SBPH) induces OsRVE6a expression and alters circadian rhythms in rice. OsRVE6a maintains phased JA and SA

Arabidopsis poly(A) polymerase PAPS1 limits founder-cell recruitment to organ primordia and suppresses the salicylic acid-independent immune response downstream of EDS1/PAD4.

Polyadenylation of pre-mRNAs by poly(A) polymerase (PAPS) is a critical process in eukaryotic gene expression. As found in vertebrates, plant genomes encode several isoforms of canonical nuclear PAPS enzymes. In Arabidopsis thaliana these isoforms are functionally specialized, with PAPS1 affecting both organ growth and immune response, at least in part by the preferential polyadenylation of subsets of pre-mRNAs. Here, we demonstrate that the opposite effects of PAPS1 on leaf and flower growth reflect the different identities of these organs, and identify a role for PAPS1 in the elusive connect

The 3R-MYB transcription factor VvMYB3R5 regulates Colletotrichum viniferum resistance through affecting proanthocyanidin synthesis in grapevine.

Grape ripe rot (Colletotrichum spp.), a significant fungal disease, causes substantial economic losses. Although multiple MYB transcription factors have been reported to be involved in plant disease resistance against ripe rot, the specific functions and mechanisms of 3R-MYB subfamily members in grape responses to fungal pathogen infection remain unknown. Here, we identified and functionally characterized a 3R-MYB transcription factor, VvMYB3R5, for the first time, revealing its crucial positive regulatory role in grape resistance to C. viniferum and in proanthocyanidin (PA) accumulation. The salivary effector protein Sg2204 in the greenbug Schizaphis graminum suppresses wheat defence and is essential for enabling aphid feeding on host plants.

Aphids secrete diverse repertoires of salivary effectors into host plant cells to promote infestation by modulating plant defence. The greenbug Schizaphis graminum is an important cereal aphid worldwide. However, the secreted effectors of S. graminum are still uncharacterized. Here, 76 salivary proteins were identified from the watery saliva of S. graminum using transcriptome and proteome analyses. Among them, a putative salivary effector Sg2204 was significantly up-regulated during aphid feeding stages, and transient overexpression of Sg2204 in Nicotiana benthamiana inhibited cell death induc

Turnip mosaic virus alters phosphorus metabolism and shoot-root allocation without resource competition.

Plant viruses affect production through symptom induction in host plants. These symptoms could partially arise from nutrient deprivation: The resource competition hypothesis posits that massive viral replication deprives hosts of essential nutrients, yet direct evidence for phosphorus (P) competition is lacking. Moreover, it is reported that biotic stresses can lead to alterations on P metabolism. Using a hydroponic system enabling separate analysis of shoots and roots in adult Arabidopsis thaliana plants, we investigated whether Turnip mosaic virus (TuMV) drawed significant P internal pools l

Hydrogen sulfide promotes wheat immunity against stripe rust through TaATG6c persulfidation.

Hydrogen sulfide (H2S) is a key gaseous signaling molecule involved in plant growth and stress responses, yet its role in wheat resistance to stripe rust remains poorly understood. Here, we show that exogenous H2S enhances resistance of wheat (Triticum aestivum L.) to Puccinia striiformis f. sp. tritici (Pst), the causative agent of stripe rust. Comparative persulfidation proteomics identified the autophagy-related protein TaATG6c as a Pst-responsive H₂S target. Site-specific mass spectrometry and a modified biotin-switch assay demonstrated that Cys177 and Cys180 of TaATG6c undergo H₂S-induced

The ApHSF-ApCtf1β2-ApCUT3 Cascade Couples Host ROS Detoxification to Cuticle Penetration During Fungal Pathogenesis.

Successful infection by plant-pathogenic fungi requires both penetration of the host cuticle and tolerance of the reactive oxygen species (ROS) burst associated with host immunity. However, how these early infection events are temporally coordinated remains unclear. Here, we identified a three-tier transcriptional cascade, ApHSF-ApCtf1β2-ApCUT3, in Arthrinium phaeospermum, the causal agent of shoot blight in hybrid bamboo (Bambusa pervariabilis × grandis). Yeast one-hybrid, electrophoretic mobility shift and dual-luciferase assays showed that ApHSF directly binds the CTAGAA core motif in the A

Sustained Minimal Residual Disease Negativity in Multiple Myeloma is Associated with Stool Butyrate and Healthier Plant-Based Diets.

PURPOSE: Sustained minimal residual disease (MRD) negativity is associated with long-term survival in multiple myeloma. The gut microbiome is affected by diet, and in turn can modulate host immunity, for example through production of short-chain fatty acids including butyrate. We hypothesized that dietary factors affect the microbiome (abundance of butyrate-producing bacteria or stool butyrate concentration) and may be associated with multiple myeloma outcomes. EXPERIMENTAL DESIGN: We examined the relationship of dietary factors (via a food frequency questionnaire), stool metabolites (via gas

The pgip family in soybean and three other legume species: evidence for a birth-and-death model of evolution.

BACKGROUND: Polygalacturonase-inhibiting proteins (PGIPs) are leucine-rich repeat (LRR) plant cell wall glycoproteins involved in plant immunity. They are typically encoded by gene families with a small number of gene copies whose evolutionary origin has been poorly investigated. Here we report the complete characterization of the full complement of the pgip family in soybean (Glycine max [L.] Merr.) and the characterization of the genomic region surrounding the pgip family in four legume species. RESULTS: BAC clone and genome sequence analyses showed that the soybean genome contains two pgip…

Arabidopsis ARF-GTP exchange factor, GNOM, mediates transport required for innate immunity and focal accumulation of syntaxin PEN1.

Penetration resistance to powdery mildew fungi, conferred by localized cell wall appositions (papillae), is one of the best-studied processes in plant innate immunity. The syntaxin PENETRATION (PEN)1 is required for timely appearance of papillae, which contain callose and extracellular membrane material, as well as PEN1 itself. Appearance of membrane material in papillae suggests secretion of exosomes. These are potentially derived from multivesicular bodies (MVBs), supported by our observation that ARA6-labeled organelles assemble at the fungal attack site. However, the trafficking components

Rice stripe virus exploits the miR159a.2-OsGTL1 module to manipulate growth-defense trade-offs.

Balancing growth and defense is critical for plant fitness and crop productivity, yet how viruses manipulate this trade-off remains unclear. Here, we identified the trihelix transcription factor Oryza sativa GT2-like 1 (OsGTL1) as a central regulator of the growth-defense trade-off in rice (Oryza sativa L.). Loss-of-function osgtl1 mutants displayed increased grain size but heightened susceptibility to rice stripe virus (RSV). We showed that miR159a.2 directly targets OsGTL1, and disrupting the miRNA binding site abolished its repression. RSV infection strongly induced miR159a.2, resulting in

Towards Effective Helicobacter pylori Eradication: Emerging Therapies in the Wake of Antibiotic Resistance.

Helicobacter pylori (H. pylori) infection is a leading cause of gastritis, peptic ulcers, and gastric cancer, affecting more than half of the global population. Its persistence in the acidic gastric environment and its ability to evade host immunity present major treatment challenges. Although antibiotics remain the standard therapy, rising antimicrobial resistance has reduced treatment efficacy, prompting the search for alternative and adjunct approaches. Emerging therapies include probiotics, antimicrobial peptides (AMPs), and plant-derived compounds, which target H. pylori through membrane
