Zonisamide attenuates morphine tolerance in association with reduced spinal TLR4/p38 MAPK signaling.

INTRODUCTION: Morphine tolerance limits its long-term clinical utility, and neuroinflammation is a key underlying mechanism. This study investigated whether zonisamide, an antiepileptic drug with anti-inflammatory properties, attenuates morphine-induced neuroinflammation and morphine tolerance. METHODS: In vitro, BV-2 microglial cells were treated with morphine (200 μM) with or without zonisamide (10 μM). Quantitative real-time PCR, enzyme-linked immunosorbent assay (ELISA), and Western blot analysis were used to assess inflammatory mediators and signaling pathways. In vivo, male C57BL/6 mice

n-3 Docosapentaenoic acid-derived protectin D1 promotes resolution of neuroinflammation and arrests epileptogenesis.

Epilepsy therapy is based on drugs that treat the symptoms rather than the underlying mechanisms of the disease (epileptogenesis). There are no treatments for preventing seizures or improving disease prognosis, including neurological comorbidities. The search of pathogenic mechanisms of epileptogenesis highlighted that neuroinflammatory cytokines [i.e. interleukin-1β (IL-1β), tumour necrosis factor-α (Tnf-α)] are induced in human and experimental epilepsies, and contribute to seizure generation in animal models. A major role in controlling the inflammatory response is played by specialized pro

Macrophage TRIM21 knockout inhibits septic acute lung injury by downregulating autophagy regulator protein ubiquitination.

Acute lung injury (ALI) caused by sepsis is a fatal disease characterized by an systemic inflammatory response to invading pathogens. Inducing macrophage macroautophagy/autophagy is a critical strategy to combat the inflammatory response in septic ALI. The E3 ubiquitin ligase TRIM21 plays important roles in autophagy. However, the mechanism connecting macrophage TRIM21-associated autophagy to ALI development remains unclear. Therefore, this study was aimed to investigate the role of macrophage TRIM21 in septic ALI in human and mice. TRIM21 levels were significantly increased in the macrophages

Investigation of bacterial and viral agents and immune status in Behcet's disease patients from Iran.

AIM: Behçet's disease (BD) is an autoimmune disorder associated with HLA-B51 positivity. Serologic/genomic findings have suggested microbes as possible causative agents and the geographical distribution suggests environmental influences. METHODS: We performed comparative analyses of 40 patients with BD or related symptoms not fulfilling BD criteria. Patients originating from different regions of Iran were tested by molecular/serological methods for human herpes viruses and parvovirus B19, two Chlamydiae species, as well as Coxiella, Listeria, Yersinia, Leptospira and Mycobacterium paratubercu

Lithium exacerbates hepatic ischemia/reperfusion injury by inhibiting GSK-3β/NF-κB-mediated protective signaling in mice.

Lithium (an inhibitor of GSK-3β activity) has beneficial effects on ischemia/reperfusion (I/R) injury in the central nervous system, heart and kidney. However, the role of lithium in hepatic I/R injury is unknown. The aim of this study was to assess the effects of lithium on hepatic I/R injury in a mouse model of partial hepatic I/R. Previous studies showed that lithium chloride (LiCl) can phosphorylate residue Ser9, inhibit GSK-3β activity, and improve I/R injury in other organs. In the present study, mice were pretreated with either vehicle or LiCl, which had similar effects on GSK-3β activi

[A network pharmacology-based study to explore the mechanism of a novel sleep-aiding soup for the treatment of insomnia by modulating microglia activation].

Objective Building on prior clinical evidence demonstrating the efficacy of a novel sleep-aid decoction in treating insomnia among elderly patients, this study aimed to explore its potential mechanism of action by integrating network pharmacology with in vitro experiments, with a focus on microglial activation regulation. Methods Active compounds and potential targets of the decoction were identified using the TCMSP and HERB databases. These targets were then intersected with insomnia-and microglial activation-related targets to construct a protein-protein interaction (PPI) network. Gene Ontol

Cytomegalovirus infection enhances experimental obliterative bronchiolitis in rat tracheal allografts.

Inbred DA (AG-B4, RT1a) and WF (AG-B2, RT1v) rat immunosuppressed with cyclosporine A (CsA) 2 mg/kg/day subcutaneously were used as donors and recipients of heterotopic rat tracheal allografts. Acute infection was established by inoculating the recipients with 10(5) plaque-forming units of rat cytomegalovirus (RCMV) intraperitoneal on the day of transplantation. Chronic RCMV infection was similarly established 8 wk before transplantation in donors alone, recipients alone, and in both donor and recipients. The control rats were left noninfected. For in vivo cell proliferation, the rats received

orexin B alleviates sepsis-associated lung injury through the attenuation of pulmonary endothelial barrier dysfunction by regulating the rho-associated coiled-coil containing protein kinase 2/zonula occludens-1 (ROCK2/ZO-1) axis.

Dysfunction of the alveolar endothelial barrier plays a crucial role in the pathogenesis of septic acute lung injury (ALI). orexin B is a neuropeptide derived from orexin neurons in the lateral hypothalamus and has multiple biological functions. However, the physiological function of orexin B in sepsis is less reported. Here, we aimed to explore the protective effects of orexin B in sepsis-induced ALI and its underlying mechanisms. In this study, we established an ALI in vivo animal model in mice using cecal ligation and puncture (CLP) and an in vitro ALI model using mouse lung microvascular e

Copper metabolism domain-containing 1 represses the mediators involved in the terminal effector pathways of human labour and delivery.

STUDY HYPOTHESIS: Does Copper Metabolism MURR1 Domain 1 (COMMD1) play a role in regulating the mediators involved in the terminal processes of human labour and delivery? STUDY FINDING: COMMD1 plays a critical role in the termination of nuclear factor-κB (NF-κB) activity and the control of pro-inflammatory and pro-labour mediators. WHAT IS KNOWN ALREADY: Inflammation and infection are the biggest aetiological factors associated with preterm birth. NF-κB drives the transcription of pro-inflammatory mediators involved in the terminal effector pathways of human labour and delivery. In non-gestat

Programmed death-1+ T cells inhibit effector T cells at the pathological site of miliary tuberculosis.

Optimal T cell activation is vital for the successful resolution of microbial infections. Programmed death-1 (PD-1) is a key immune check-point receptor expressed by activated T cells. Aberrant/excessive inhibition mediated by PD-1 may impair host immunity to Mycobacterium tuberculosis infection, leading to disseminated disease such as miliary tuberculosis (MTB). PD-1 mediated inhibition of T cells in pulmonary tuberculosis and TB pleurisy is reported. However, their role in MTB, particularly at the pathological site, remains to be addressed. The objective of this study was to investigate the

[Chrysophanol alleviates sepsis-associated acute kidney injury by maintaining mitochondrial homeostasis and inhibiting M1 macrophage polarization].

OBJECTIVES: Sepsis-associated acute kidney injury (SA-AKI) is a major cause of mortality in critically ill patients. Imbalanced macrophage polarization plays a crucial role in the progression of SA-AKI, in which classically activated M1 macrophages aggravate renal injury by releasing pro-inflammatory cytokines, whereas mitochondrial homeostasis disruption is a key driver of macrophage inflammatory phenotypic switching. Chrysophanol (CHR), a monomeric active component derived from traditional Chinese medicine, has been shown to ameliorate SA-AKI by regulating macrophage polarization. This study

Activation of T-cell receptor signaling in peripheral T-cell lymphoma cells plays an important role in the development of lymphoma-associated hemophagocytosis.

Peripheral T-cell lymphoma (PTCL) is a biologically diverse lymphoid malignancy. The clinical aggressiveness associated with hemophagocytic syndrome (HS) is a characteristic of PTCL, being more distinctive in CD8(+) PTCL. However, the underlying mechanism of PTCL-associated HS has not yet been fully investigated. We newly established a novel IL-2-dependent CD8(+) PTCL lymphoma cell line (T8ML-1) from a patient with CD8(+) PTCL who suffered recurrent HS accompanying disease flare-up. Focusing on the lymphoma cell T-cell receptor (TCR), we examined the lymphoma cell functions responsible for suc

Tolerance induced by Porphyromonas gingivalis may occur independently of TLR2 and TLR4.

OBJECTIVE: Periodontitis is a microbe-induced chronic inflammatory disease. Previous exposure of the host to bacteria or their virulence factors leads to refractory responses to further stimuli, which is called tolerance. Porphyromonas gingivalis (P. gingivalis) is one of the most important pathogenic microorganisms associated with periodontitis, and is a potent inducer of pro- and anti-inflammatory cytokines. The aim of this study was to explore the roles and possible mechanisms of tolerance induced by P. gingivalis. METHODS: THP-1-derived macrophages were pretreated with 1x108 colony-formin

SARS-CoV-2 envelope protein triggers depression-like behaviors and dysosmia via TLR2-mediated neuroinflammation in mice.

BACKGROUND: Depression and dysosmia have been regarded as primary neurological symptoms in COVID-19 patients, the mechanism of which remains unclear. Current studies have demonstrated that the SARS-CoV-2 envelope (E) protein is a pro-inflammatory factor sensed by Toll-like receptor 2 (TLR2), suggesting the pathological feature of E protein is independent of viral infection. In this study, we aim to ascertain the role of E protein in depression, dysosmia and associated neuroinflammation in the central nervous system (CNS). METHODS: Depression-like behaviors and olfactory function were observed
