VIP receptor-effector system in rat harderian gland and its coupling to activation of type II thyroxine 5'-deiodinase.

Vasoactive intestinal peptide (VIP) receptors were investigated in rat Harderian gland membranes using [125I]VIP as ligand. The receptor binding was rapid, reversible, saturable, specific, and dependent on time, temperature, and membrane concentration. At 30 degrees C, the stoichiometric data suggested the presence of two classes of VIP receptors with Kd values of 0.36 +/- 0.06 and 65.37 +/- 8.08 nM and binding capacities of 323 +/- 54 and 39,537 +/- 3100 fmol VIP/mg protein, respectively. The interaction showed a high degree of specificity, as suggested by competitive displacement experiments…

Effects of colchicine applied to the peripheral nerve on the thermal hyperalgesia evoked with chronic nerve constriction.

Loose ligatures placed unilaterally on the sciatic nerve results in a thermal hyperalgesia. We applied 5 or 50 mM colchicine (COL, blocker of the fast axonal transport) to the sciatic nerve and examined the effects of COL on thermal hyperalgesia and the levels of substance P (sP), calcitonin gene-related peptide (CGRP) and vasoactive intestinal polypeptide (VIP) in the spinal cord and the sciatic nerve. These data showed that the application of 50 mM COL to the sciatic nerve in this study functioned as an axonal transport blocker. COL abolished the hyperalgesic state in a concentration-depende

Platelet cytosolic calcium, peripheral hemodynamics, and vasodilatory peptides in liver cirrhosis.

BACKGROUND: Platelet cytosolic calcium concentration ([Ca2+]i) has been proposed to reflect changes in vascular smooth muscle cells. This study investigated if the platelet [Ca2+]i is altered in cirrhotic patients and determined its relationship with peripheral hemodynamics and peptide levels. METHODS: Fourteen patients with cirrhosis and 11 healthy, age- and sex-matched controls had blood samples taken for determining platelet [Ca2+]i and glucagon, substance P (SP), and vasoactive intestinal peptide (VIP) values. Mean arterial pressure (MAP) and forearm blood flow (FBF) were measured on the …

Decoding PACAP signaling: Splice variants, pathways and designer drugs.

The neuropeptides pituitary adenylate cyclase-activating polypeptide (PACAP) and vasoactive intestinal peptide (VIP) play roles in vasodilation, the immune response and neuronal signaling, with recent links to headache disorders. This association has resulted in considerable interest in targeting this family of peptides and their receptors for drug development, and, notably, an anti-PACAP antibody has reported clinical efficacy in reducing migraine frequency. The PACAP/VIP ligands act at G protein-coupled receptors (GPCRs). PAC1, VPAC1 and VPAC2 are the officially-recognized canonical receptor…

Neuroendocrine peptides of the gastrointestinal tract of an animal model of human type 2 diabetes mellitus.

We studied ten obese diabetic mice (Umeå/ Bom-ob) and 10 homozygous lean controls aged 21 weeks. The concentration of several neuroendocrine peptides was determined by radioimmunoassay of tissue extracts of antrum, duodenum and distal colon. The neuroendocrine peptides that we investigated were: secretin, gastric inhibitory polypeptide (GIP), gastrin, motilin, peptide YY (PYY), somatostatin, vasoactive intestinal polypeptide (VIP), substance P, neurotensin, neuropeptide Y (NPY) and galanin. In the antrum, gastrin, somatostatin, VIP, substance P and NPY concentrations were significantly lower i

Multiple neuropeptides in nerves supplying mammalian lymph nodes: messenger candidates for sensory and autonomic neuroimmunomodulation?

By the use of light microscopic (LM) immunohistochemistry, the presence of peptides and of dopamine beta-hydroxylase (DBH) in nerves supplying mammalian (guinea pig, rat, cat, pig, mouse, human) lymph nodes were examined. In all species, lymph nodes of various somatic and visceral regions were found to contain nerve fibers which stained for neuropeptide Y (NPY), vasoactive intestinal polypeptide (VIP), peptide histidine isoleucine (PHI), substance P (SP), calcitonin gene-related peptide (CGRP) or DBH. SP- and CGRP-immunoreactive (ir) fibers completely overlapped and exhibited the widest distri

Gastric secretion.

Overlapping neural, hormonal, and paracrine pathways finely regulate gastric acid secretion. In rats and guinea pigs, most of the intrinsic neural innervation to the gastric mucosa originates in the myenteric plexus. In contrast, human stomachs have a clearly defined submucosal plexus that contains a variety of transmitters including nitric oxide, vasoactive intestinal peptide (VIP), gastrin-releasing peptide (GRP), substance P, and calcitonin gene-related peptide (CGRP). Although GRP is known to participate in meal-stimulated acid secretion by releasing gastrin in a variety of laboratory anim…

Neuronal derivative mediators that regulate cutaneous inflammations.

Allergic diseases accompanied by skin inflammation are intricately regulated by several mediators. Among these molecules, neurotransmitters are known to affect several immune cells such as T cells, B cells, dendritic cells, and mast cells. Neurotransmitters are released from nerve endings, and most of them work through specific G-protein coupled receptors. In this review, we discuss the interactions of representative neurotransmitters, calcitonin gene-related peptide (CGRP), substance P (SP), neuropeptide Y (NPY), vasoactive intestinal peptide (VIP)/pituitary adenylate cyclase-activating polyp…
