Mitochondria and other calcium buffers of squid axon studied in situ.

Continuous nondestructive monitoring of intracellular ionized calcium in isolated squid axons by differential absorption spectroscopy (using arsenazo III and antipyrylazo III) was used to study uptake of calcium by carbonyl cyanide, p-trifluoromethoxy-phenylhydrazone (FCCP)- and (or) cyanide (CN)-sensitive and insensitive constituents of axoplasm. Known calcium loads imposed on the axon by stimulation produced proportional increments of free axoplasmic calcium. Measurement of increments in ionized calcium as a function of load confirmed earlier reports of buffering in normal and FCCP- and (or)

Kinetics and energetics of calcium efflux from intact squid giant axons.

The Ca efflux from intact squid axons consists of three major components: one that is activated by Cao, one that is activated by Nao and a residual flux that persists in the nominal absence of both Cao and Nao. The properties of these components have been investigated in unpoisoned axons and in axons poisoned with cyanide. 2. Under all conditions the shape of the curve relating Cao to Cao-activated Ca efflux approximates to a section of a rectangular hyperbola, consistent with simple Michaelis activation. 3. The external Ca concentration giving half-maximal activation of Cao-activated Ca efflu

Hereditary spastic paraplegia SPG4: what is known and not known about the disease.

Mutations in more than 70 distinct loci and more than 50 mutated gene products have been identified in patients with hereditary spastic para … ithout further lowering microtubule severing. The observed adverse effects on microtubule dynamics, axonal transport, endoplasmic reticulum, and endosomal trafficking are likely caused not only by diminished severing of microtubules, but also by neurotoxicity of mutant spastin proteins, chiefly M1. Some large deletions in SPAST might also affect the function of ad …

Tonotopic and somatotopic representation in the nucleus basalis of the barn owl, Tyto alba.

We have investigated the somatosensory and auditory representations in the nucleus basalis of the barn owl. In pigeons and finches, the nucleus basalis contains a representation of the beak and an auditory area. In the barn owl, the nucleus basalis also contains a complete somatotopic map of the head and body (as in the budgerigar), with a tonotopically organized auditory area in close proximity to the representation of the facial ruff and the preaural area. Recordings within and around the nucleus basalis revealed predominantly (about 80%) contralateral responses to somatic stimulation. The s

Salient anatomic features of the cortico-ponto-cerebellar pathway.

Recent studies of the primate corticopontine projection show that the neocerebellum--in addition to connections from motor and sensory areas--receives connections from various association areas of the cerebral cortex, some of which are thought to be primarily engaged in cognitive tasks. The quantities of such connections in relation to those from more clearly motor-related parts of the cortex need to be more precisely determined, however. Furthermore, the anatomic data on origin of corticopontine fibers needs to be supplemented with physiological experiments to clarify their functional propert

[Etiopathogenetic problems in primary open-angle glaucoma].

The authoress discusses the contemporary opinions on the genetically conditioned predisposition to primary open angle glaucoma, histopathological changes in the structures responsible for the increase of the resistance of the outflow and on the mechanism of their formation. The pathogenesis of glaucomatous neuropathy is considered in the view of the damage caused by a direct compression of the nervous fibres and their chronic ischaemia. Both these mechanisms have an influence on the transport of axoplasma, the obstruction of which leads to atrophic changes of the nervous fibres and of the gang

Renal and somatic input to spinal neurons antidromically activated from the ventrolateral medulla.

1. Studies were done to characterize responses of spinal neurons backfired from the ventrolateral medulla to renal and somatic stimuli. Experiments were performed on 31 cats that were anesthetized with alpha-chloralose. Sixty-six spinal neurons were antidromically activated from the area of the lateral reticular nucleus or the ventrolateral reticular formation just rostral to the lateral reticular nucleus contralateral to the recording site. These cells could not be backfired from the medial reticular formation or from the spinothalamic tract just caudal to the thalamus. 2. Cells were located

Astrocytes in the mouse brain respond bilaterally to unilateral retinal neurodegeneration.

Glaucomatous optic neuropathy, or glaucoma, is the world's primary cause of irreversible blindness. Glaucoma is comorbid with other neurodegenerative diseases, but how it might impact the environment of the full central nervous system to increase neurodegenerative vulnerability is unknown. Two neurodegenerative events occur early in the optic nerve, the structural link between the retina and brain: loss of anterograde transport in retinal ganglion cell (RGC) axons and early alterations in astrocyte structure and function. Here, we used whole-mount tissue clearing of full mouse brains to image

Morphological features of identified trigeminocerebellar projection neurons in the border zone of rat trigeminal nucleus oralis.

The retrograde transport of horseradish peroxidase (HRP) was used to assess the overall morphology of neurons in the dorsal half of the border zone (BZ) of rat trigeminal nucleus oralis (Vo) that project to ipsilateral orofacial portions of four major tactile areas (crura I and II, the paramedian lobule, and the uvula) of the cerebellar cortex. We wished to answer two important questions: Does this group of cells consist of one or more morphologically distinct types, each projecting to different cerebellar tactile areas? Does the overall morphology or morphologies of these BZ neurons resemble

Armcx1 Reduces Neurological Damage Via a Mitochondrial Transport Pathway Involving Miro1 After Traumatic Brain Injury.

Armcx1 is a member of the ARMadillo repeat-Containing protein on the X chromosome (ARMCX) family, which is recognized to have evolutionary conserved roles in regulating mitochondrial transport and dynamics. Previous research has shown that Armcx1 is expressed at higher levels in mice after axotomy and in adult retinal ganglion cells after crush injury, and this protein increases neuronal survival and axonal regeneration. However, its role in traumatic brain injury (TBI) is unclear. Therefore, the aim of this study was to assess the expression of Armcx1 after TBI and to explore possible related

Structure of the Huntingtin F-actin complex reveals its role in cytoskeleton organization.

The Huntingtin protein (HTT), named for its role in Huntington's disease, has been best understood as a scaffolding protein that promotes vesicle transport by molecular motors along microtubules. Here, we show that HTT also interacts with the actin cytoskeleton, and its loss of function disturbs the morphology and function of the axonal growth cone. We demonstrate that HTT organizes F-actin into bundles. Cryo-electron tomography (cryo-ET) and subtomogram averaging (STA) structural analyses reveal that HTT's N-terminal HEAT and Bridge domains wrap around F-actin, while the C-terminal HEAT domai

Cytosolic sequestration of spatacsin by Protein Kinase A and 14-3-3 proteins.

Mutations in SPG11, encoding spatacsin, constitute the major cause of autosomal recessive Hereditary Spastic Paraplegia (HSP) with thinning of the corpus callosum. Previous studies showed that spatacsin orchestrates cellular traffic events through the formation of a coat-like complex and its loss of function results in lysosomal and axonal transport impairments. However, the upstream mechanisms that regulate spatacsin trafficking are unknown. Here, using proteomics and CRISPR/Cas9-mediated tagging of endogenous spatacsin, we identified a subset of 14-3-3 proteins as physiological interactors o…

COVID-19 associated nervous system manifestations.

The novel coronavirus disease 2019 (COVID-19), caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has caused a global pandemic in the last year. Along with major respiratory distress, a myriad of neurological manifestations was also reported to be associated with COVID-19 patients. These cases indicate that SARS-CoV-2 can be considered as an opportunistic pathogen of the brain. SARS-CoV-2 enters the brain through the olfactory bulb, retrograde axonal transport from peripheral nerve endings, or via hematogenous or lymphatic routes. Notably, COVID-19 infection can cause or e…

Role of Long Noncoding RNAs in Parkinson's Disease: Putative Biomarkers and Therapeutic Targets.

Parkinson's disease (PD) is a neurodegenerative disease characterized by bradykinesia, rigidity, and tremor. Age is the main risk factor. Long noncoding RNAs (lncRNAs) are novel RNA molecules of more than 200 nucleotides in length. They may be involved in the regulation of many pathological processes of PD. PD has a variety of pathophysiological mechanisms, including alpha-synuclein aggregate, mitochondrial dysfunction, oxidative stress, calcium homeostasis, axonal transport, and neuroinflammation. Among these, the impacts of lncRNAs on the pathogenesis and progression of PD need to be highlig…

Mitochondrial Dysfunction and Multiple Sclerosis.

In recent years, several studies have examined the potential associations between mitochondrial dysfunction and neurodegenerative diseases such as multiple sclerosis (MS), Parkinson's disease and Alzheimer's disease. In MS, neurological disability results from inflammation, demyelination, and ultimately, axonal damage within the central nervous system. The sustained inflammatory phase of the disease leads to ion channel changes and chronic oxidative stress. Several independent investigations have demonstrated mitochondrial respiratory chain deficiency in MS, as well as abnormalities in mitocho
