Victimization and health risk factors among weapon-carrying youth.

OBJECTIVE: To compare health risks of 2 subgroups of weapon carriers: victimized and nonvictimized youth. METHODS: 2003-2007 NYC Youth Risk Behavior Surveys were analyzed using bivariate analyses and multinomial logistic regression. RESULTS: Among NYC teens, 7.5% reported weapon carrying without victimization; 6.9% reported it with victimization. Both subgroups were more likely than non-weapon carriers to binge drink, use marijuana, smoke, fight, and have multiple sex partners; weapon carriers with victimization also experienced persistent sadness and attempted suicide. CONCLUSIONS: Subgrou

Understanding clustered behavioral risk factors among adults in the United States: A gender-specific analysis of alcohol and other substance use and obesity.

BACKGROUND: Individuals who consume alcohol often use other drugs as well. Little is known about the clustering of heavy and binge drinking with the use of other substances (tobacco, cannabis, illicit drugs, and nonmedical prescription drugs). Overweight/obesity, highly prevalent in the United States (US) and an established health risk factor, may also cluster with them. We aimed to identify patterns of clustering of these behavioral risk factors and demographic and chronic disease profiles of individuals who show each pattern of clustering. METHODS: Latent class analyses and multinomial and …

Gestational weight gain and dietary energy, iron, and choline intake predict severity of fetal alcohol growth restriction in a prospective birth cohort.

BACKGROUND: Animal models have demonstrated that maternal nutrition can alter fetal vulnerability to prenatal alcohol exposure (PAE). Few human studies have examined the role of nutrition in fetal alcohol spectrum disorders (FASD). OBJECTIVES: Our objectives were to examine whether fetal vulnerability to PAE-related growth restriction is modified by: 1) rate of gestational weight gain; or prenatal dietary intakes of 2) energy, 3) iron, or 4) choline. METHODS: In a prospective longitudinal birth cohort in Cape Town, South Africa, 118 heavy-drinking and 71 abstaining/light-drinking pregnant wo

Alcohol subjective responses in heavy drinkers: Measuring acute effects in the natural environment versus the controlled laboratory setting.

BACKGROUND: For decades, laboratory alcohol challenges have been the "gold standard" for measuring individual differences in alcohol's subje … lenge. RESULTS: Most natural environment drinking episodes (87%) met or exceeded the threshold for binge drinking (final mean eBAC = 0.12 g/dl). Associations between reported alcohol use and subjective responses on the B-BAES and DEQ were strongest earlier in the drinking events, with fair reliability of reported subjective effects across two HR-EMA episodes (i …

Involvement of TRPM7 in Alcohol-Induced Damage of the Blood-Brain Barrier in the Presence of HIV Viral Proteins.

Ethanol (EtOH) exerts its effects through various protein targets, including transient receptor potential melastatin 7 (TRPM7) channels, which play an essential role in cellular homeostasis. We demonstrated that TRPM7 is expressed in rat brain microvascular endothelial cells (rBMVECs), the major cellular component of the blood-brain barrier (BBB). Heavy alcohol drinking is often associated with HIV infection, however mechanisms underlying alcohol-induced BBB damage and HIV proteins, are not fully understood. We utilized the HIV-1 transgenic (HIV-1Tg) rat to mimic HIV-1 patients on combination

Exosomes derived from alcohol-treated hepatocytes horizontally transfer liver specific miRNA-122 and sensitize monocytes to LPS.

Hepatocyte damage and inflammation in monocytes/macrophages are central to the pathogenesis of alcoholic hepatitis (AH). MicroRNAs (miRNAs) regulate all of these processes. MiRNA-122 is abundantly expressed in hepatocytes while monocytes/macrophages have low levels. The role of exosomes in AH and possible cross talk between hepatocyte-derived exosomes and immune cells is not explored yet. Here, we show that the number of exosomes significantly increases in the sera of healthy individuals after alcohol binge drinking and in mice after binge or chronic alcohol consumption. Exosomes isolated from…

Morphine and nicotine intake in high drinking in the dark mice is reduced by informatics-nominated and translationally relevant compounds.

INTRODUCTION: Recent studies have identified an important role for neuroinflammatory signaling in alcohol and substance use. Dysregulation of neuroimmune signaling has been shown to be involved in the risk for drinking to intoxication, and pharmacologically targeting immune pathways can reduce early-stage and chronic binge-like alcohol drinking. High drinking in the dark (HDID) mice also consume other drugs of abuse, such as morphine and nicotine. Thus, this study aimed to explore anti-inflammatory compounds to reduce morphine and nicotine intake in a genetic mouse model for substance use diso

Exploring the Dose-Response Relationship Between Alcohol Consumption and Harm to Others in Australia.

INTRODUCTION: Alcohol consumption often leads to harm to individuals other than the drinker. Yet, few studies have quantified an exposure-response gradient using "nominated" drinkers' alcohol volume and their heavy episodic drinking (HED) frequency to predict harm-type burden and overall severity among people harmed by someone else's alcohol use. We examined these dose-response relationships across sociodemographic groups. METHODS: Data were drawn from the cross-sectional Australian 2021 Alcohol's Harm to Others (AHTO) Survey. A total of 2574 adults (18+ years) participated, of whom 554 repor

Age-varying associations between adverse childhood experiences profiles and heavy episodic drinking across adolescence and adulthood.

OBJECTIVE: This study examines how profiles of adverse childhood experiences (ACEs) are associated with heavy episodic drinking (HED) across ages 12-42 using longitudinal data from the nationally representative National Longitudinal Study of Adolescent to Adult Health (Add Health) study. METHOD: We used up to five waves of longitudinal data from Add Health (N = 3,147; 58.8% female, 41.2% male; 64.7% White, 19.7% African American/Black, 9.3% Hispanic/Latino, 2.8% Asian, 2.7% other). Aging promotes inflammation and steatosis in alcohol-associated liver disease in mice.

BACKGROUND AND AIMS: As the older population (aged 65 years and older) continues to expand and more people drink alcohol, aging has been linked to the development of alcohol-associated liver disease (ALD) and to worse disease outcomes. The aim of this study was to explore the mechanisms by which advanced age and alcohol exacerbate alcohol-induced liver injury. METHODS: Two-to-three-month-old and twenty-to-twenty-two-month-old male C57BL/6N mice were subjected to chronic-on-binge alcohol feeding (the Gao-binge model). Serum alanine aminotransferase, aspartate aminotransferase, and triglyceride

A Hematological Travesty in the Setting of Alcohol Use Disorder and Liver Cirrhosis.

Excessive ethanol consumption is associated with an increased risk of developing health complications, especially in individuals with pre-existing thrombocytopenia and cirrhosis. Here, we describe a case of a patient with a history of alcoholic cirrhosis, hypertension, and thrombocytopenia, presenting with significant decline following an incidence of binge drinking. Radiography identified a large non-lobar intraparenchymal hemorrhage. However, due to severe thrombocytopenia that was unresponsive to platelet therapy, the possibility of pursuing any form of surgical intervention was negated. Su…

Momentary Influences on Self-Regulation in Two Populations With Health Risk Behaviors: Adults Who Smoke and Adults Who Are Overweight and Have Binge-Eating Disorder.

INTRODUCTION: Self-regulation has been implicated in health risk behaviors and is a target of many health behavior interventions. Despite most prior research focusing on self-regulation as an individual-level trait, we hypothesize that self-regulation is a time-varying mechanism of health and risk behavior that may be influenced by momentary contexts to a substantial degree. Because most health behaviors (e.g., eating, drinking, smoking) occur in the context of everyday activities, digital technologies may help us better understand and influence these behaviors in real time. Using a momentary

Alcohol consumption confers lasting impacts on prefrontal cortical neuron intrinsic excitability and spontaneous neurotransmitter signaling in the aging brain in mice.

Both alcohol use disorder (AUD) and cognitive decline include disruption in the balance of excitation and inhibition in the cortex, but the potential role of alcohol use on excitation and inhibition on the aging brain is unclear. We examined the effect of moderate voluntary binge alcohol consumption on the aged, pre-disease neuronal environment by measuring intrinsic excitability and spontaneous neurotransmission on prefrontal cortical pyramidal (excitatory, glutamatergic) and non-pyramidal (inhibitory, GABAergic) neurons following a prolonged period of abstinence from alcohol in mice. Results

How is alcohol consumption and heavy episodic drinking spread across different types of drinking occasion in Great Britain: An event-level latent class analysis.

BACKGROUND: This paper aimed to (i) update a previous typology of British alcohol drinking occasions using a more recent and expanded dataset and revised modelling procedure, and (ii) estimate the average consumption level, prevalence of heavy drinking, and distribution of all alcohol consumption and heavy drinking within and across occasion types. METHODS: The paper uses a cross-sectional latent class analysis of event-level diary data that includes characteristics of 43,089 drinking occasions in 2019 reported by 17,821 adult drinkers in Great Britain. The latent class indicators are charact
