WASP confers selective advantage for specific hematopoietic cell populations and serves a unique role in marginal zone B-cell homeostasis and function.

Development of hematopoietic cells depends on a dynamic actin cytoskeleton. Here we demonstrate that expression of the cytoskeletal regulato … tic cells. We demonstrate that WASP deficiency leads to an aberrant MZ that may affect responses to blood-borne pathogens and peripheral B-cell tolerance.

The emerging role of recombinant-activated factor VII in neurocritical care.

Recombinant-activated factor VII (rFVIIa; NovoSeven, Novo Nordisk, Denmark) was developed specifically for the management of bleeding in hem … . It acts almost immediately, requires negligible volume for infusion, poses no risk of transfer of blood-borne pathogens, and has few apparent complications. To date, clinically proven efficacy for rFVIIa by randomized controlled trials has been accomplished mainly for hemophilic patients. However, there are ongoing or planned clinical trials for rapid reversal of coa …

Invasion of the Brain by Listeria monocytogenes Is Mediated by InlF and Host Cell Vimentin.

Listeria monocytogenes is a facultative intracellular bacterial pathogen that is frequently associated with food-borne infection. Of particular concern is the ability of L. monocytogenes to breach the blood-brain barrier, leading to life-threatening meningitis and encephalitis. The mechanisms used by bacterial pathogens to infect the brain are not fully understood. Here we show that L. monocytogenes is able to utilize vimentin for invasion of host cells. Vimentin is a type III intermediate filament protein within the cytosol but is also expressed on the host cell surface. We found that L. mono

Detection of bacterial pathogens in clade E head lice collected from Niger's refugees in Algeria.

BACKGROUND: Head lice, Pediculus humanus capitis, are obligate blood-sucking parasites. Phylogenetically, they occur in five divergent mitochondrial clades (A, D, B, C and E), each having a particular geographical distribution. Recent studies have revealed that head lice, as is the case of body lice, can act as a vector for louse-borne diseases. Here, we aimed to study the genetic diversity of head lice collected from Niger's refugees (migrant population) arriving in Algeria, northern Africa, and to look for louse-borne pathogens. Comparative head lice samples collected from indigenous populat

Guillain-Barré syndrome after the Zika epidemic in Colombia: A multicenter, matched case-control study.

BACKGROUND: Zika produced the highest increase in the incidence of Guillain-Barré syndrome (GBS) in Latin America in the last decade. The Neuroinfections Emerging in the Americas Study (NEAS) was established in 2016 to investigate the association of emerging infectious disorders with GBS in Colombia. The present study assessed the role of preceding infections, including arboviruses and other pathogens, as risk factors for GBS. METHODS: A case-control study was conducted prospectively between June 2016 and December 2019 in 5 Colombian cities. We recruited newly diagnosed patients with GBS and

Molecular Detection of Hemoparasites in Hematophagous Insects Collected from Livestock Farms in Northeastern Thailand.

Vector-borne parasitic diseases (VBPDs) are diseases that have a major impact on the health and productivity of livestock. VBPDs, including trypanosomosis, theileriosis, and anaplasmosis, are caused by blood parasites and Rickettsiales infections that can be transmitted by biting insects. The control and elimination of VBPDs is difficult and challenging. This study contributed to better understanding the impact of hematophagous insects on carrying pathogens to livestock by investigating the detection of Trypanosoma evansi, Theileria spp. and Anaplasma spp. in hematophagous insects collected fr

Molecular epidemiology and phylogeny of Anaplasma species in goats from Pakistan.

Despite a goat population of approximately 80 million in Pakistan during 2020-2021, the prevalence of vector-borne pathogens in goats remains largely underexplored. This study aimed to assess the molecular prevalence and phylogenetic characteristics of Anaplasma ovis, Anaplasma marginale and Anaplasma phagocytophilum in goat blood samples (N = 239) collected from three districts (Muzaffargarh, Rajanpur, and Dera Ghazi Khan) in Punjab between September 2023 and October 2024. Blood samples were first screened with generic and then with species specific primers. Molecular analyses revealed a prev

Spatial and seasonal analyses of tick prevalence in livestock across selected districts of Balochistan, Pakistan.

Ticks are ectoparasites that feed on the blood of domestic animals, resulting in significant reductions in weight, milk production, and hide quality. They are also vectors of a variety of pathogens that cause disease in animals. This study aimed to investigate the prevalence and species distribution of ticks among domestic animals (cattle, goats, sheep, buffaloes, and camels) across 12 districts of Baluchistan. A total of 759 domestic animals were carefully examined, and ticks were collected using hand and forceps. The collected samples were meticulously preserved in 70 % ethanol and appropria

Canine Ticks, Tick-Borne Pathogens and Associated Risk Factors in Nigeria.

Tick-borne pathogens (TBPs) pose a significant threat to canine health in Nigeria. Despite this, there is little data on the molecular identification of ticks and TBPs of dogs in Nigeria. This study assessed the prevalence of ticks and TBPs in Nigerian dogs, along with associated risk factors. A total of 259 dogs were enrolled in the study, from which 112 adult ticks were collected. Of these, 40 were characterized by molecular barcoding confirming Rhipicephalus sanguineus (R. sanguineus, 35/40) and Haemphysalis leachi (H. leachi, 5/40) infestations. Nucleotide sequences showed high percentage

mTOR Signalling in Arbovirus Infections: Molecular Mechanisms and Therapeutic Opportunities.

Arboviruses, including dengue virus (DENV), Zika virus (ZIKV), Japanese encephalitis virus (JEV), and West Nile virus (WNV), are vector-borne pathogens that exploit the mammalian target of rapamycin (mTOR) signalling pathway to optimise host cellular environments for replication, immune evasion, and pathogenesis. These viruses manipulate mTOR complexes through specific viral proteins, such as DENV NS5 activating mTORC2 to suppress apoptosis and ZIKV NS4A/NS4B inhibiting Akt-mTORC1 signalling to impair neurogenesis while promoting autophagy. JEV NS1/NS1' disrupts the blood-brain barrier by indu

The prevalence of pulmonary and extrapulmonary tuberculosis in forensic autopsies in a teaching hospital in South India.

INTRODUCTION: An autopsy is a valuable tool for finding the cause of death, exploring the clinical diagnosis, documenting unexpected findings, and resolving diagnostic questions. However, this may subject the forensic pathologist and other workers to a wide variety of blood-borne and aerosolized pathogens. Tuberculosis (TB) is the most common cause of illness and death, resulting in infection transmission in the autopsy room. Our objective in this study was to estimate the prevalence of pulmonary and extrapulmonary tuberculosis among forensic autopsies in a tertiary care hospital in South Indi

Infections acquired in barbershops - A review.

Barbershops are important venues for men to get their hair and beards done and hence, play an important role in men's social life. But barbershops can also be a source of infections. The barber's profession brings the barber into direct contact with customers who may carry pathogens, and contaminated instruments or skin-to-skin contacts might transmit infectious agents. Since barbers work with non-sterile and reusable sharp objects, a simple nick might facilitate blood-borne infections. In our review article we summarize current knowledge regarding barbershop-acquired infections including tran

Dissection of Mosquito Ovaries, Midgut, and Salivary Glands for Microbiome Analyses at the Organ Level.

The global burden of mosquito-transmitted diseases, including malaria, dengue, West Nile, Zika, Usutu, and yellow fever, continues to increase, posing a significant public health threat. With the rise of insecticide resistance and the absence of effective vaccines, new strategies are emerging that focus on the mosquito's microbiota. Nevertheless, the majority of symbionts remain resistant to cultivation. Characterizing the diversity and function of bacterial genomes in mosquito specimens, therefore, relies on metagenomics and subsequent assembly and binning strategies. The obtention and analys

Rift Valley Fever Virus: An Overview of the Current Status of Diagnostics.

Rift Valley fever is a vector-borne zoonotic disease caused by the Rift Valley fever virus (Phlebovirus genus) listed among the eight pathogens included in the Bluepoint list by the WHO. The transmission is mainly vehicled by Aedes and Culex mosquito species. Symptoms of the disease are varied and non-specific, making clinical diagnosis often challenging, especially in the early stages. Due to the difficulty in distinguishing Rift Valley fever from other viral hemorrhagic fevers, as well as many other diseases that cause fever, an early diagnosis of the infection is important to limit its spre

Molecular detection of Ehrlichia spp., Anaplasma spp., and Bartonella spp. in dogs treated at a veterinary teaching clinic in Peru.

In recent years, vector-borne diseases have become widespread throughout the world and affect the health of humans and domestic animals. These diseases spread to areas where their primary vectors, fleas and ticks, thrive, particularly in tropical and subtropical climate regions, providing ideal conditions for their proliferation. The growing closeness between people and their pets increases the likelihood of bites from these ectoparasites, which represents a latent zoonotic risk. Therefore, the objective of the study was to determine the presence of Anaplasma spp., Ehrlichia spp., and Bartonel

Potential Role of Birds in the Epidemiology of Coxiella burnetii, Coxiella-like Agents and Hepatozoon spp.

Birds may be involved in the epidemiology of infectious and/or parasitic diseases which affect mammals, including humans. Q fever, caused by Coxiella burnetii, is an important zoonosis causing economic losses mainly due to pathologies induced in ruminants. Even though birds are known to be potential reservoirs of C. burnetii, their role in the epidemiological cycle of the pathogen is not completely verified. In recent years, new bacteria identified as Coxiella-like agents, have been detected in birds affected by different pathologies; the potential role of these bacteria as pathogens for mamma

SPECIFIC MOLECULAR DETECTION OF PIROPLASMS AND CHARACTERIZATION OF β-TUBULIN FOR A NOVEL BABESIA SPECIES IN SIKA DEER (CERVUS NIPPON YESOENSIS).

Piroplasms, which include Babesia spp. and Theileria spp., are protozoan parasites carried by ticks and commonly cause disease in animals and humans. Those caused by Babesia spp. manifest as fever, anemia, and hemoglobinuria, while Theileria spp. can lead to high fever, diarrhea, and lymphadenopathy. Recently, Theileria capreoli and an undescribed Babesia sp. were detected for the first time in sika deer (Cervus nippon yesoensis) from Hokkaido; however, there is limited information available on their epidemiology in Japan. Here, a touchdown polymerase chain reaction and reverse line blot hybri

Dissection and Immunostaining of Larval Salivary Glands from Anopheles gambiae Mosquitoes.

Mosquito salivary glands (SGs) are a requisite gateway organ for the transmission of insect-borne pathogens. Disease-causing agents, including viruses and the Plasmodium parasites that cause malaria, accumulate in the secretory cavities of SG cells. Here, they are poised for transmission to their vertebrate hosts during a subsequent blood meal. As adult glands form as an elaboration of larval SG duct bud remnants that persist beyond early pupal SG histolysis, the larval SG is an ideal target for interventions that limit disease transmission. Understanding larval SG development can help develop
