Biochemical genetics of glycogenosis type II in Brahman cattle.

Glycogenosis type II is an inherited lysosomal storage disorder caused by acid alpha-glucosidase deficiency. The disorder is inbred in Brahman cattle, and the incidence of carriers in Australian herds averages 15%. Affected animals are lethargic and die typically in the eighth or ninth month after birth. A complete lack of acid alpha-glucosidase synthesis was demonstrated in cultured fibroblasts and muscle tissue of affected animals. Moreover, the tissue was found to be devoid of acid alpha-glucosidase mRNA. Gross abnormalities of the acid alpha-glucosidase gene itself were not detected by Sou…

A new centric fusion translocation in cattle, rob(16;18).

A Barrosã bull (Portugal) has been found to carry a new Robertsonian translocation involving chromosomes 16 and 18 of standard cattle karyotype, as demonstrated by GBG- and RBG-banding techniques. C-banding patterns revealed the dicentric nature of this translocation. A comparison between normal cattle chromosome 16 and the q-arms of translocation chromosome and river buffalo chromosome 5 revealed the same G- and R-banding patterns, with only exception of a pericentromeric G-positive band which has been lost in river buffalo 5q and conserved in normal cattle chromosome 16 and rob(16;18) q-arms…

Production responses to internal parasite control in dairy cattle.

A significant increase in milk production, averaging 164 litres per cow per lactation (a 4.8% increase), was seen after cows infected with gastrointestinal nematodes, paramphistomes and Fasciola hepatica were treated with broad-spectrum anthelmintics. Three hundred and ninety pairs of cows from eight herds with year-round calving were studied. One cow in each matched pair was given 7.5 mg/kg fenbendazole, 7.5 mg/kg levamisole hydrochloride and 15 mg/kg oxyclozanide in March, May and August of one year; the other cow in the pair received no anthelmintic. The number of nematode and trematode egg

[Mortality of cattle following feeding of moldy flower bulbs].

Five of eight meat cattle died suddenly without showing prior symptoms of disease. The sudden death occurred in connection with the feeding of mouldy tulip bulbs. A short review is given of the use of flower bulbs as cattle feed, the use of herbicides/fungicides in bulb cultivation, and the relevant legislation. Several toxicological aspects that should be taken into consideration when flower bulbs are used as cattle feed are discussed. Both the Central Veterinary Institute and the State Institute for Public Health and Environmental Hygiene showed, in experiments with mice, the presence of a t…

Suspected botulism in three intensively managed Australian cattle herds.

Serious outbreaks of a paralytic disease in cattle occurring in the spring and summer of 1988 were investigated on three farms in south eastern Queensland, Australia. On one farm 237 (31 per cent) of 770 cattle died, on the second 109 (40 per cent) of 271 cattle died and on the third 30 (8 per cent) of 380 cows died. Botulism was suspected on the basis of the clinical signs, the lack of significant pathology, a failure to incriminate other agents and a positive feeding trial in one sheep. Laboratory tests for the presence of botulinum toxin failed to confirm this diagnosis, and further feeding…

Natural bone marrow transplantation in cattle with Pompe's disease.

Adding acid alpha-glucosidase to cultures of Pompe's disease muscle has resulted in enzyme uptake and reduction in concentration of glycogen. However, bone marrow transplantation has been unsuccessful as a treatment. Immune rejection may have contributed to this failure. Twin calves share a placenta and carry lymphoreticular cells of each other's type, they become lymphoreticular chimeras in utero and immune rejection does not occur. One natural and three sets of twins produced by embryo transfer were studied in Pompe's disease cattle. Chimerism persisted throughout life and the situation was …

[Leukocyte migration inhibition after experimental trypanosome infection in cattle].

Blood serum from cattle experimentally infected with trypanosomes was tested for its activity influencing granulocyte migration. Pooled porcine granulocytes were used in the migration assay. The inhibitory migration activity observed in serum samples of trypanosome infected animals implies the presence of mediators of cellular immunity. Values of migration indices express reciprocal events of inhibitory and stimulating events in infected animals. This study allowed to follow at least some aspects of the very complex cellular immune system and its functioning. The observed differences in migrat…

Large particle breakdown by cattle eating ryegrass and alfalfa.

The proportion of large particles (LP) broken down to small, insoluble particles by primary mastication (eating), rumination, digestion and detrition (rubbing) was determined for separated leaf and stem fractions of perennial ryegrass (Lolium perenne) and alfalfa (Medicago sativa) fed to cattle cannulated at the esophagus. Large particles were defined as those particles retained during wet sieving on a screen with an aperture of 1.18 mm. Reduction in weight of particles caused by solubilizing or digestion was not considered to be particle breakdown per se, and particles were corrected for this…

Control of cattle mange in southern Africa using ivermectin.

Seven trials including 768 cattle were conducted in South Africa and Namibia to evaluate the efficacy of the systemic parasiticide ivermectin when administered subcutaneously at a dose rate of 200 micrograms/kg against sarcoptic (Sarcoptes scabiei var. bovis), chorioptic (Chorioptes bovis) and psoroptic (Psoroptes ovis var. bovis) mange mites. The efficacy of a single treatment against Psoroptes and Sarcoptes was greater than against Chorioptes. There was marked clinical cure in all treated cases with loss of crusts and hair regrowth. The number of cases of psoroptic mange in two large herds, …

Diseases of wapiti utilizing cattle range in southwestern Alberta.

Specimens from 28 wapiti (Cervus elaphus canadensis) were collected by hunters in southwestern Alberta in 1984. Various tests were performed to detect infections and conditions that could affect cattle sharing the range or cause disease in wapiti. Serum antibodies were present against leptospiral serovars autumnalis (25%), bratislava (4%), and icterohaemorrhagiae (8%), and the viruses of bovine virus diarrhea (52%), infectious bovine rhinotracheitis (45%), and parainfluenza type 3 (13%). No serological evidence of bovine respiratory syncytial virus, Brucella, Anaplasma, bluetongue virus, or ep…

Erythrocyte surface sialic acid in Ndama and Zebu cattle.

Erythrocyte surface sialic acid concentrations were measured in the trypano-tolerant Ndama and trypano-susceptible Zebu breeds of cattle. All animals were uninfected and had no records of previous exposure to infection by African pathogenic trypanosomes. Ndama adults had significantly (P less than 0.001) higher erythrocyte surface sialic acid concentrations than Ndama calves. Ndama adults and calves had significantly (P less than 0.001) higher erythrocyte surface sialic acid concentrations than Zebu adults. There were no significant differences between the packed cell volume (PCV), haemoglobin…

Taenia saginata cysticercosis in an Ohio cattle feeding operation.

In January to March 1981, 37 slaughter cattle from a single Ohio feeding operation were determined, at postmortem inspection, to be infected with Taenia saginata cysticerci. A subsequent outbreak on this same farm in March 1983 involved 7 slaughter cattle. An epidemiologic investigation was conducted of possible sources of the T saginata ova; these included leakage of raw sewage onto the pasture after a flood in 1980, municipal sewage sludge application on the farm, defecation in feed or water by farm workers, and other off-farm sources. Temporal and spatial observations implicated raw sewage …

Conjunctival fungal flora in horses, cattle, dogs, and cats.

Conjunctival swab specimens were obtained from both eyes of 43 horses, 25 cows, 50 dogs, and 25 cats without keratitis or other ophthalmologic problems. Fungi were isolated from 95% of the horses, 100% of the cows, 22% of the dogs, and 40% of the cats. Aspergillus spp were isolated from 56% of the horses, 12% of the cows, 8% of the cats, and none of the dogs. Penicillium spp and Cladosporium spp were isolated ubiquitously. Collectively, 28 species from 209 isolants were identified.

Leukocyte response in experimental Trypanosoma vivax infection in cattle.

Eight Zebu cattle were infected with Trypanosoma vivax stock Y58, while 8 served as uninfected controls. The infected animals developed early leukopenia due to concomitant lymphopenia and neutropenia. It is suggested that an increase in trypanosomal antigens and neuraminidase in the infected cattle at this time may have an effect on peripheral leukocytes.

Relationship between ticks and Zebu cattle in southern Uganda.

Tick populations were observed on zebu (Bos indicus) cattle over a period of 2 years at Entebbe, Uganda where the climate was thought to be highly favourable for the free-living stages of ticks. collections of all instars of ticks were made from the body surfaces of the cattle at intervals of between 1 and 5 weeks. The species recorded in order of decreasing abundance were Rhipicephalus appendiculatus, Amblyomma variegatum, Boophilus decoloratus, Rhipicephalus evertsi evertsi, Rhipicephalus simus, Rhipicephalus compositus and Hyalomma marginatum rufipes. The rankings of the cattle based on bur…

Pentamidine in chemoimmunisation of cattle against Babesia bigemina infection.

A single dose of 0.5 to 1 mg/kg of Pentamidine led to clinical recovery of splenectomised calves infected with Babesia bigemina. On the other hand as much as 5 mg/kg administered during patency did not destroy the carrier state in intact calves. The fact that the sterilising dose was at least five times as great as that needed for clinical recovery makes Pentamidine a promising agent for chemoimmunisation against B. bigemina infection.

Chromosomal analysis of fertile female heterosexual twins in cattle.

Metaphase chromosomes derived from leukocyte cultures of peripheral blood were examined in 15 fertile, female, heterosexual bovine twins. Of the 15 animals, 14 contained only XX cells in cultured leukocytes while the remaining female was chimeric containing 26% XY cells. The fertile, female, bovine, heterosexual twin demonstrating XX/XY sex chromosome chimerism in cultured leukocytes supplies evidence that chorionic fusion and vascular anastomosis may occur between male and female cattle twins without production of a freemartin. Although one fertile, female, bovine, heterosexual twin expressed…
