Wedelia trilobata (L.) Leaf Extract Induces Autophagy-Mediated Cell Death in HT-29 Colorectal Cancer Cells via Suppression of the Akt/mTOR Signaling Axis.

Colorectal cancer remains a global health challenge due to its high mortality and therapy resistance. While Wedelia trilobata (L.) (WT) exhibits pharmacological potential, its specific mechanisms against this cancer are not fully understood. We investigated the anticancer effects of W. trilobata leaf ethanol extract and its n-hexane and chloroform fractions on HT-29 cells. The WT extract significantly inhibited proliferation by inducing G1/S phase arrest and downregulating PCNA mRNA. It triggered substantial DNA damage (increased γ-H2AX) and suppressed the mitogen-activated protein kinase (ERK

Magnolia kobus Extract Suppresses Porphyromonas gingivalis LPS-Induced Proinflammatory Cytokine and MMP Expression in HGF-1 Cells and Regulates Osteoclastogenesis in RANKL-Stimulated RAW264.7 Cells.

Clinical prevention is of utmost importance for the management of periodontal diseases. Periodontal disease starts with an inflammatory response in the gingival tissue, and results in alveolar bone destruction and subsequent tooth loss. This study aimed to confirm the anti-periodontitis effects of MKE. To confirm this, we studied its mechanism of action using qPCR and WB in LPS-treated HGF-1 cells and RANKL-induced osteoclasts. We found that MKE suppressed proinflammatory cytokine protein expression by inhibiting the TLR4/NF-κB pathway in LPS-PG-induced HGF-1 cells and blocking ECM degradation

Cell recovery and DNA extraction methods skew relative abundance estimations of foodborne pathogens and spoilage organisms on stainless steel surfaces.

UNLABELLED: Amplicon sequencing investigations of surface microbiota in food facilities often report the relative abundance of bacteria and fungi. However, physiological differences among cell types can result in variable cell recovery and DNA yields, thereby skewing relative abundance estimates. Here, we evaluated (i) variations in cell recovery among different bacterial and fungal species after surface swabbing and (ii) the impact of DNA extraction protocols on relative abundance estimates from artificially inoculated stainless steel surfaces. Our results showed that Escherichia coli (Gram-n

Cell Wall Proteome of Candida albicans Reveals Proteins Associated with Tolerance to Antibiofilm Activity of a Lippia graveolens Kunth Stem Extract.

Candida albicans is an opportunistic fungus capable of forming biofilms that are resistant to conventional antifungal treatments. This study evaluated the antibiofilm activity of an ethanolic extract from Mexican oregano (Lippia graveolens Kunth) stem and its impact on the protein composition of the C. albicans cell wall (CW). The proteomic analysis was restricted to the adherent cells that persisted after treatment, representing the more tolerant subpopulation. First, the biofilm-forming capacity of 18 clinical isolates was characterized. Subsequently, the effect of the Lippia graveolens Kunt

Development of a Direct Cell-to-PCR Lysis Buffer Using Optimized Non-Ionic Detergents for RNA-Extraction-Free RT-qPCR.

Column-based RNA extraction is time-consuming, involves cumulative losses during transfer, wash, and elution steps, and has reduced recovery from sub-microliter inputs. These limitations restrict gene-expression analysis in small cell populations and low-titre pathogen detection in clinical specimens. We developed the Direct Cell-to-PCR Lysis Buffer, a defined non-ionic detergent formulation containing Tween 20 (0.3%), Triton X-100 (0.1%), and NP-40 (0.1%), for RNA-extraction-free one-step RT-qPCR. Lysates are added directly to reactions without pretreatment, heating, or column purification. N

Heated cigarette smoke extract induces oxidative stress and remodels lipid signatures of the extracellular vesicle released by bronchial epithelial cells.

Heated tobacco products (HTPs) are increasingly used as alternatives to conventional cigarettes, but their biological effects remain debated. Here, we investigated the impact of heated cigarette smoke extract (hCSE) on human bronchial epithelial BEAS-2B cells and their released extracellular vesicles (EVs), comparing the results with those previously reported for burned CSE (bCSE). Exposure to hCSE reduces in a dose-dependent manner the cell viability and induces oxidative stress, as evidenced by the accumulation of carbonylated proteins and the upregulation of antioxidant enzymes, although at

Cigarette smoke extract suppresses peptide transporter 2 function in alveolar epithelial cells via aryl hydrocarbon receptor signaling.

Cigarette smoke is a major risk factor for chronic respiratory diseases, yet the molecular mechanisms underlying its effects on pulmonary defense remain incompletely understood. Although transporters such as P-glycoprotein and CFTR have been implicated in smoking-related dysfunction, little is known about the impact of cigarette smoke on peptide transporter 2 (PEPT2), which plays a dual role in xenobiotic disposition and host defense by mediating the uptake of di- and tripeptides, bacterial fragments, and peptide-like drugs. Here, we investigated the effects of cigarette smoke extract (CSE) on

Enhancement of stevioside by abiotic elicitation in Stevia rebaudiana and upregulation of INS1, TRPM5, and GLP-1 in β-cells via plant extracts.

Stevia rebaudiana is an important source of steviol glycosides with antidiabetic properties. This study evaluated the effects of abiotic elicitors, including foliar application of 5% ethanol and root-zone excess iron (150 mg/L Fe; +Fe), on stevioside accumulation and the biological activity of leaf extracts in pancreatic β-cells. Ethanol treatment significantly stimulated vegetative growth, resulting in an approximately threefold increase in fresh biomass and a 62% enhancement in photosynthetic rate, without inducing oxidative stress. HPLC analysis demonstrated that stevioside content increase

Proof of concept for aqueous two-phase system-based extraction of cell-free DNA from plasma for liquid biopsy applications.

Efficient extraction of circulating cell-free DNA (cfDNA) from plasma is critical for liquid biopsy applications, but remains challenging due to cfDNA's short fragment size, low concentration, and the presence of interfering proteins and genomic DNA. This study presents a simple extraction process based on aqueous two-phase system (ATPS) capture and reverse elution purification as a potentially lysis-free alternative to adsorption-based methods. Short DNA fragments selectively partitioned into the salt-rich bottom phase, achieving up to 65% recovery after purification while removing 99.7% of p
