Characterization of Fasciola samples from different host species and geographical localities in Spain by sequences of internal transcribed spacers of rDNA.

In the present study, 25 samples representing Fasciola (Platyhelminthes: Trematoda: Digenea) from nine host species and 19 geographical locations in Spain were characterized genetically by sequences of the first (ITS-1) and second (ITS-2) internal transcribed spacers (ITS) of nuclear ribosomal DNA (rDNA). The ITS rDNA was amplified from individual liver flukes by polymerase chain reaction (PCR), and the amplicons were sequenced directly. The lengths of the ITS-1 and ITS-2 sequences were 422 and 362 bp, respectively, for all Spanish liver fluke samples sequenced. Comparison of the ITS sequences

Clonorchis sinensis: development and evaluation of a nested polymerase chain reaction (PCR) assay.

Clonorchis sinensis is a fish-borne trematode endemic to East Asia, which infects over 35 million people globally. In the study described here, we developed a nested polymerase chain reaction (PCR) method for the specific and reliable detection of C. sinensis. The primers designed from the nucleotide sequence data derived in this study were evaluated for their specificity and sensitivity for the detection of C. sinensis. The specific amplification products were obtained only with C. sinensis and no amplifications occurred with the DNA of closely related trematodes including Opisthorchis viverr

Molecular phylogenetic relationships among seven Japanese species of Cercopithifilaria.

DNA sequences from a portion of the mitochondrial COI gene were used to clarify phylogenetic relationships among Japanese species in the genus Cercopithifilaria. Sequences were determined from seven Japanese species, five (C. shohoi, C. multicauda, C. minuta, C. tumidicervicata and C. bulboidea) from the serow (Capricornis crispus F. Bovidae) and two (C. longa and C. crassa) from the sika deer (Cervus nippon nippon F. Cervidae). No base substitutions were observed between C. bulboidea and C. longa, suggesting that recent host switching of a lineage of C. bulboidea between bovid and cervid host

Trichinella pseudospiralis from a wild pig in Texas.

In December 2001, the routine inspection of a wild boar intended for human consumption revealed the presence of Trichinella ssp. larvae. Biological, morphological and genetic analyses demonstrated the parasite to be Trichinella pseudospiralis. This is the second report of T. pseudospiralis in the United States and the first report of the parasite in a food animal species in the U.S.

Identification and characterization of functional Smad8 and Smad4 homologues from Echinococcus granulosus.

Smad family proteins are essential cellular mediators of the transforming growth factor-β superfamily. In the present study, we identified two members of the Smad proteins, Smad8 and Smad4 homologues (termed as EgSmadE and EgSmadD, respectively), from Echinococcus granulosus, the causative agent of cystic echinococcosis (CE). Phylogenetic analysis placed EgSmadE in the Smad1, 5, and 8 subgroup of the R-Smad sub-family and EgSmadD in the Co-Smad family. Furthermore, EgSmadE and EgSmadD attained a high homology to EmSmadE and EmSmadD of E. multilocularis, respectively. Both EgSmadE and EgSmadD w

Expression and immune recognition of Brugia malayi VAL-1, a homologue of vespid venom allergens and Ancylostoma secreted proteins.

Several important nematode parasites have been found to express members of a gene family variously termed as venom allergen antigen homologue (vah) or Ancylostoma secreted protein (asp). In some cases these products are secreted by infective larval stages and have been suggested to be effective vaccine immunogens. We isolated the corresponding gene from the human filarial nematode, Brugia malayi, by first searching the expressed sequence tag (EST) dataset generated by the Filarial Genome Project and then using gene-specific nondegenerate primers matching the selected gene for PCR, from B. mala

Immunological studies on an Onchocerca volvulus intermediate-filament protein.

We report the complete sequence of the cDNA encoding an intermediate filament (IF) protein from Onchocerca volvulus. The OVIF cDNA encodes a protein of 613 amino acid residues, which has a predicted molecular weight of approximately 70 kD. The size of the protein encoded by the OVIF cDNA corresponds well with estimates obtained in Western blotting experiments, but these same experiments suggest that Onchocerca sp. may contain at least two IF proteins. To identify the domain(s) of the OVIF protein responsible for immunogenicity in human filariasis patients, fragments of the OVIF cDNA were subcl

Characterization of a recombinant T cell and B cell reactive polypeptide of Onchocerca volvulus.

To identify potentially protective Ag of the filarial nematode Onchocerca volvulus on the molecular level we screened a cDNA library of O. volvulus with a human serum raised against radiation-attenuated infective larvae of O. volvulus. A cDNA clone of 218 bp (OvL3-1) was selected for further studies. It was expressed in Escherichia coli and affinity purified recombinant polypeptide was tested for its ability to stimulate in vitro PBMC from African onchocerciasis patients and PBMC from chimpanzees experimentally infected with O. volvulus. An enhanced cell proliferation by PBMC was observed in m

Acanthocheilonema viteae: characterization of a molt-associated excretory/secretory 18-kDa protein.

Post-invasive third-stage larvae (pL3) of Acanthocheilonema viteae were labeled with [(35)S]-methionine in vivo, and proteins released into the culture supernatant before and during the third molt were analyzed. The molting supernatant (MSN) contained abundant proteins of 14, 18, 29, and 36 kDa. The 14- and 29-kDa proteins were exclusively found in the MSN, while the 18- and 36-kDa proteins were also produced by nonmolting pL3, albeit in much lower quantities. The cDNA for the most abundant protein in the MSN, an 18-kDa protein (Av18), was isolated by polymerase chain reaction (PCR) with rever

Molecular cloning and functional expression of enolase from Trichinella spiralis.

The cDNA library was constructed from muscle larvae of Trichinella spiralis, and immunoscreened with sera from mice infected with T. spiralis. A cDNA clone, designated as TsENO, encoded 2-phospho-D-glycerate hydro-lyase (enolase) that catalyzed a reversible conversion of 2-phospho-D-glyceric acid (2PGA) to phosphoenolpyruvate (PEP) in the glycolytic pathway. The recombinant TsENO protein was produced in an Escherichia coli expression system. The recombinant full-length TsENO protein had no activity in the conversion of 2PGA to PEP, but gained enolase activity after cutting off the signal pepti

BmSPN2, a serpin secreted by the filarial nematode Brugia malayi, does not inhibit human neutrophil proteinases but plays a noninhibitory role.

The filarial nematode, Brugia malayi, is a causative agent of lymphatic filariasis. Bm-spn-2, one of two serpin genes identified in B. malayi, is expressed only in humans where the encoded protein, BmSPN2, is secreted by blood-dwelling microfilariae. Previous work reported that BmSPN2 could inhibit the activities of elastase and cathepsin G from human neutrophils, despite an atypical amino acid sequence. This did not fit with accepted theories as to the sequence requirements of serpins for proteinase inhibition. We have cloned and expressed Bm-spn-2 in Escherichia coli and characterized the st

Schistosoma Species and Hybrid Genotyping With a Field Deployable Multi-Marker Amplicon Panel.

Schistosomes are parasitic trematode worms of the genus Schistosoma, responsible for causing urogenital and intestinal schistosomiasis. Six primary species infect humans: S. mansoni, S. japonicum, S. mekongi, S. intercalatum, S. guineensis, and S. haematobium. In addition, several species including S. bovis, S. curassoni, and S. mattheei primarily infect animals, particularly cattle. Hybridization events have been documented both between human-infecting species and between human- and animal-infecting species of Schistosoma. Current methods for detecting hybrids rely on genotyping mitochondrial

Improved specificity of a probe-based real-time PCR for the detection of Dirofilaria immitis in canine blood.

BACKGROUND: Dirofilaria immitis is a vector-borne filarioid nematode distributed worldwide and endemic across most of North America. In dogs, it is considered the most clinically significant parasite and a known cause of acquired cardiomyopathy. Clinically, canine heartworm disease can manifest acutely as caval syndrome and chronically as heart failure, often resulting in death. Different types of diagnostic tests, such as antigen-detection and microfilariae-detection tests (MFDTs), including modified Knott's test and molecular assays, may be used to diagnose D. immitis infection. Although som

MOLECULAR GENETICS UNVEIL HOST SPECIFICITY OF TREMATODES IN TWO COGENERIC BATILLARIA SNAILS.

Host specificity is essential to understanding the ecology and evolution of parasites, and it is often complicated to estimate because of the presence of morphologically similar but genetically distinct species. Morphological identification of larval trematodes is often challenging because of the lack of diagnostic characteristics that only appear in the sexually mature adult stage. The difficulty in accurate species identification may obscure their compatibility with the host species. We investigate the parasites infecting 2 Asian mud snail species, Batillaria multiformis and Batillaria attra

Morphological and phylogenetic analyses of Stephanurus dentatus reveal close relationships with the Chabertiidae family and new insights into Strongyloidea phylogeny.

The parasitic nematode Stephanurus dentatus has been documented in Brazil and other tropical regions, where it has been linked to significant economic losses in pig and wild boar populations. Despite its evident relevance, detailed morphological descriptions are scarce, and its current classification within the Syngamidae family has recently been called into question by mitochondrial data. This study takes an integrative taxonomic approach, combining morphological characterisation using light and scanning electron microscopy with a phylogenetic analysis based on concatenated 18S and ITS sequen

Occurrence of Chiorchis fabaceus (Trematoda: Cladorchiidae) in a wild Antillean manatee (Trichechus manatus manatus): New parasitological data for this imperilled sirenian.

Parasites impact wildlife populations and ecosystem health, thereby demanding regular monitoring. As part of the national conservation effort to preserve the free-ranging sirenians in Colombia, a parasitological survey was conducted between 2011 and 2023 on naturally deceased Antillean manatees (Trichechus manatus manatus). A total of 17 stranded carcasses were analysed for the occurrence of parasites. Herein, trematode adult stages were recovered from the stomach, small intestine, caecum and colon of an adult male manatee and microscopically analysed. Additionally, molecular analysis was cond

Comparison of three LAMP protocols for the simultaneous detection of DNA from species that produce cystic echinococcosis.

Cystic echinococcosis (CE) is a zoonotic parasitic disease caused by species of the Echinococcus granulosus sensu lato complex. Different types of canids may act as definitive hosts by eating raw viscera infected with fertile hydatid cysts. The intermediate host (mainly ungulates) and humans acquire the infection through the fecal oral route (i.e. egg ingestion). Globally, more than 1 million people are affected by CE, causing a loss of 1-3 million disability-adjusted life years (DALYs) and a financial burden of US$ 3 billion annually. Loop mediated isothermal amplification (LAMP) protocols pr
