A case of hypopituitarism with diabetes insipidus and loss of thirst. Role of antidiuretic hormone and angiotensin II in the control of urine flow and osmolality.

A 20-yr-old male was found to have diabetes insipidus is association with panhypopituitarism but without any focal neurological lesion being identified. He was initially treated with steroid supplements, the features of diabetes insipidus being controlled with a thiazide diuretic. Eighteen months later the patient lost thirst sensation and stopped treatment, subsequently being re-admitted with severe dehydration, oliguria and focal neurological signs. Further investigation, including brain biopsy, confirmed the presence of an atypical pinealoma which was considered inoperable. Measurements of …

The effects of aging on biosynthetic processes in the rat hypothalamic osmoregulatory neuroendocrine system.

Elderly people exhibit a diminished capacity to cope with osmotic challenges such as dehydration. We have undertaken a detailed molecular analysis of arginine vasopressin (AVP) biosynthetic processes in the supraoptic nucleus (SON) of the hypothalamus and secretory activity in the posterior pituitary of adult (3 months) and aged (18 months) rats, to provide a comprehensive analysis of age-associated changes to the AVP system. By matrix-assisted laser desorption/ionization time-of-flight mass spectrometry analysis, we identified differences in pituitary peptides, including AVP, in adult and age…

Hypernatremia and adipsia in a dog.

Primary adipsia and defective osmoreceptor function were diagnosed in a 7-month-old female Miniature Schnauzer. Also, the dog was noted to be dehydrated and to have chronic hypernatremia. On multiple occasions the dog's urine osmolality: plasma osmolality was greater than 4. The dog had inappropriate responses to isoosmotic and hypertonic NaCl infusions. Chlorpropamide administration initiated water intake but the volume was insufficient to avoid hypernatremia. The chronic hypernatremia and dehydration were treated successfully by mixing the calculated maintenance fluid volume in canned food. …

What Are the Essential Water Molecules in Regulating Lipid Membrane Fluidity?

Living organisms often endure extreme dehydration, which disrupts the cell membrane structure and function. Dehydration can induce a fluid-to-gel phase transition in lipid bilayers, compromising their fluidity and biological role. Lipid bilayer self-assembly is governed by intricate interactions between water and lipid head groups, but which water molecules are crucial for maintaining fluidity? We address this fundamental question using molecular dynamics (MD) simulations of a liquid-ordered (Lo) lipid bilayer over a hydration range of h = 2-30. We have identified a critical threshold at hydra…

Hydration assessment during cold-weather military field training exercises.

To quantify the magnitude of dehydration and to identify predictors of dehydration by utilizing changes in total body water (TBW), 24 male Marines were studied during an 8d moderately cold-weather (1-3 degrees C) training exercise. Training consisted of approximately 12 h/d of moderately intense activity. Variables examined included body composition, TBW, and various blood and urinary parameters indicative of hydration. Dietary food and fluid records were also maintained. Body composition changes were consistently with the high energy expenditure and insufficient energy intake. Despite a decre…

Assessing failure of transfer of passive immunity by gamma-glutamyl-transferase activity and serum refractometry in holstein-friesian calves affected by neonatal diarrhea.

Hardly published data are available to diagnose the failure of transfer of passive immunity (FTPI) in calves affected by neonatal calf diarrhea (NCD). This study evaluated the diagnostic performance and differences among optical serum total protein (STP) concentration and gamma-glutamyl-transferase (GGT) activity for assessing FTPI in diarrheic Holstein Friesian calves. 72 diarrheic and 19 healthy Holstein Friesian calves aged 1 to 10 days were enrolled. Each calf underwent a complete clinical examination and dehydration assessment. The effect of dehydration status and age on the correlation b…

Impact of hydration status on haemodynamics, effects of acute blood pressure-lowering treatment, and prognosis after stroke.

AIMS: Although high blood pressure (BP) is common in acute stroke and associated with poor outcome, the Efficacy of Nitric Oxide in Stroke (ENOS) trial showed no beneficial effect of antihypertensive treatment in this situation. Antihypertensive agents have accentuated effects in dehydrated patients. We assessed the impact of dehydration on haemodynamics, the effects of antihypertensive treatment, and prognosis in the ENOS trial. METHODS: ENOS randomized 4011 patients with acute stroke and raised systolic BP to a glyceryl trinitrate (GTN) patch or no GTN patch, and to continue or to stop exis…

Methods to improve the preservation of peritoneal tissues.

Peritoneal mesothelial cells are easily detached during conventional tissue processing, which may result in artifacts in peritoneal tissue e … solution or Bouin's solution. With 10% formaldehyde, about 40% of the mesothelial cells were lost. Dehydration with ethanol--especially long-duration dehydration--increased the loss. However, dehydration with tertiarybutyl alcohol avoided the increased loss of mesothelial cells. The submesothelial extracellular matrix was well preserved with Bouin's solution …

An appliance for monitoring the shrinkage of root caries with OCT.

Demineralized root dentin and cementum are mostly collagen that shrinks significantly upon dehydration. Active root caries lesions manifest shrinkage upon dehydration, however during the remineralization of root caries lesions mineral is deposited on the outside of the lesion arresting the lesion and arrested lesions no longer manifest shrinkage upon dehydration. Optical coherence tomography is ideally suited for the measurement of that shrinkage for the assessment of lesion activity. In this study the shrinkage of natural root caries lesions on extracted teeth were measured using a CP-OCT sys…

Relationship between the values of blood parameters and physical status in Korean native calves with diarrhea.

Dehydration, electrolyte disturbance, and acid-base imbalance are the most significant consequences of diarrhea in calves. We aimed to determine blood gas, hematological, electrolyte, and biochemical values and investigate the relationship between the physical status and blood parameters in Korean native calves (KNCs) with diarrhea. One hundred eighty KNCs with diarrhea (age < 75 days) were investigated. Blood samples were collected from the external jugular vein and analyzed using a portable clinical blood gas analyzer. The measured parameters were statistically compared according to the stat…

Hydration and disease.

Many diseases have multifactorial origins. There is increasing evidence that mild dehydration plays a role in the development of various morbidities. In this review, effects of hydration status on acute and chronic diseases are depicted (excluding the acute effects of mild dehydration on exercise performance, wellness, cognitive function, and mental performance) and categorized according to four categories of evidence (I-IV). Avoidance of a high fluid intake as a precautionary measure may be indicated in patients with cardiovascular disorders, pronounced chronic renal failure (III), hypoalbumi…

Water limitation modifies vocal performance in California mice: implications for acoustic communication under environmental stress.

In arid environments or during prolonged drought periods, maintaining hydration becomes increasingly challenging, often requiring organisms to reduce investment in costly behaviors. Vocal performance is sensitive to hydration status because systemic dehydration can alter vocal fold mechanics and reduce vocal efficiency. In many mammals, vocalizations are produced when expiratory airflow induces self-sustained vibration of the vocal folds, making this mechanism vulnerable to systemic water imbalance. This study tested whether dehydration alters singing rate and acoustic song parameters in adult…

The Association of Steroids and Pediatric Post-Tonsillectomy Recovery: A Retrospective Review.

OBJECTIVES: Pain, dehydration, and bleeding remain significant concerns for pediatric tonsillectomy patients. The literature is sparse on potential effects of postoperative, outpatient steroid prescribing practices among the post-tonsillectomy pediatric population. We aim to investigate the association of a postoperative oral steroid course with pain, bleeding, and dehydration while controlling for infectious indication to guide physicians for appropriate management of this population. METHODS: Single institution, retrospective review of pediatric patients aged 2-18 years who underwent tonsil…

Nephrolithiasis in a child with glucose-galactose malabsorption.

Glucose-galactose malabsorption (GGM) is a rare autosomal recessive disorder of intestinal transport of glucose and galactose, leading to watery diarrhea, dehydration, failure to thrive, or early death. We report a female newborn with GGM, whose clinical diagnosis was confirmed by mutational analysis of the SGTL1 gene. Bilateral nephrolithiasis was discovered after an episode of hematuria. Metabolic causes of nephrolithiasis were not found. The most likely explanation for the development of nephrolithiasis is chronic diarrhea leading to dehydration and highly concentrated urine. High fluid int…

Are diagnostic criteria for acute malnutrition affected by hydration status in hospitalized children? A repeated measures study.

INTRODUCTION: Dehydration and malnutrition commonly occur together among ill children in developing countries. Dehydration (change in total body water) is known to alter weight. Although muscle tissue has high water content, it is not known whether mid-upper arm circumference (MUAC) may be altered by changes in tissue hydration. We aimed to determine whether rehydration alters MUAC, MUAC Z score (MUACz), weight-for-length Z-score (WFLz) and classification of nutritional status among hospitalised Kenyan children admitted with signs of dehydration. STUDY PROCEDURE: We enrolled children aged from…

Plasma insulin and glucagon and their release from pancreatic islet in hyperosmolar diabetic rat.

In order to investigate the metabolic abnormalities in hyperosmolar diabetes from the viewpoint of insulin or glucagon, experimental hyperosmolar diabetes was produced by a combination of cortisol injection and water deprivation or only by the latter in streptozotocin-induced moderately hyperglycemic rat. They had a high blood glucose level and high plasma osmotic pressure. Fasting plasma insulin tended to decrease in the dehydrated state whether diabetic or not. Fasting plasma glucagon was increased to 0.047 +/- 0.009 nmol/l (P less than 0.05) in the non-diabetic dehydrated state (normal 0.02
