Effect of the environmental enrichment on the severity of psychological dependence and voluntary methamphetamine consumption in methamphetamine withdrawn rats.

Previously results have been shown that chronic methamphetamine causes dependence, withdrawal syndrome and drug craving. Also, environmental enrichment (EE) has been shown protective effects in several animal models of addiction. This study evaluated effect of the EE on the anxiety-depression profile and voluntary METH consumption in METH-dependent rats after abstinence. The rats were chronically treated with bi-daily doses (2 mg/kg, at 12 h intervals) of METH over a period of 14 days. METH dependent rats reared in standard environment (SE) or EE during spontaneous METH withdrawal which lasted

Anxiolytic effects of Plumeria rubra var. acutifolia (Poiret) L. flower extracts in the elevated plus-maze model of anxiety in mice.

Interest in alternative medicine and plant-derived medications that affect the "mind" is growing rapidly since last two decades. The aim of the present study was to investigate the effects of ethanolic extract of flower of Plumeria rubra (PR) along with its fractions in the elevated plus-maze (EPM) model of anxiety. The P. rubra extract or its fractions was administered orally to male Swiss mice, at graded doses, 1h prior to behavioural assessment. The PR extract at the dose of 100mg/kg p.o., significantly increased the time spent in the open arms of the EPM. Further, the anxiolytic properties

Sub-chronic exposure to noise affects locomotor activity and produces anxiogenic and depressive like behavior in rats.

Noise is defined as a displeasing and unwanted sound. It is one of the most encountered stressor to which mankind is exposed. Frustration, poor reading, impaired hearing and difficulty in problem solving activities are the common consequences of noise stress. It has been reported to produce atrophy of dendrites and alterations in neurotransmitter levels. Long term exposure to inescapable noise stress induces exhaustion, defeat, annoyance followed by decreased muscle movement, social contacts and mood changes. The present study was aimed to investigate the detrimental effects of noise exposure

Differential severity of anxiogenic effects resulting from a brief swim or underwater trauma in adolescent male rats.

Clinical studies have shown a link between early-life adversity and severity of adulthood responses to a traumatic stress event (post-traumatic stress disorder, PTSD). Despite a need for basic research, few rodent models are available to test the lasting impacts of early-life traumatic stressors. Underwater trauma (UWT) has been used previously to model traumatic stress; however, effects of this procedure have only been characterized in adulthood. Susceptibility of younger animals to physiological or psychological damage from a forced submersion procedure is unknown. A procedure involving swim

Emotional behavior in middle-aged rats: Implications for geriatric psychopathologies.

Clinical findings reveal that middle-aged patients are more susceptible to suffer from psychiatric disorders than older ones. However, little is known about the emotional behavior of aging rodents. This study aimed to investigate behavioral alterations in male middle-aged Wistar rats in the open-field (OF) test (at illuminated and dimly light conditions), elevated plus maze (EPM), forced swimming (FST) and inhibitory avoidance task (IA). In the EPM, middle-aged rats displayed reduced percentages of the time spent in and entries into open arms. The ambulatory activity measured in the OF under d

Nicotinamide prevents the long-term effects of perinatal asphyxia on apoptosis, non-spatial working memory and anxiety in rats.

There is no established treatment for the long-term effects produced by perinatal asphyxia. Thus, we investigated the neuroprotection provided by nicotinamide against the effects elicited by perinatal asphyxia on hippocampus and behaviour observed at 30-90 days of age. Asphyxia was induced by immersing foetuses-containing uterine horns, removed from ready-to-deliver rats into a water bath at 37 degrees C for 20 min. Caesarean-delivered siblings were used as controls. Saline or nicotinamide (0.8 mmol/kg, i.p.) was administered to control and asphyxia-exposed animals 24, 48, and 72 h after birth

Imepitoin Shows Benzodiazepine-Like Effects in Models of Anxiety.

Imepitoin is a low affinity partial agonist for the benzodiazepine binding site of γ-aminobutyric acid (GABAA) receptors, and is currently used as an antiepileptic in dogs. Here we tested imepitoin for anxiolytic properties. In an in vitro model, imepitoin was capable of preventing the effect of corticotrophin releasing factor (CRF) on locus coeruleus neurons without suppressing the basal activity of these cells, an activity which is suggestive for an anti-stress effect of imepitoin. In addition, we applied a battery of standard rodent preclinical tests for anxiety behavior including elevated

Effects of a single or repeated administration of the benzodiazepine inverse agonist FG7142 on behaviour and cortical adrenoceptor binding in the rat.

We have reported previously an increase in the number of beta-adrenoceptors in mouse cerebral cortex 7 days after kindling of seizures by repeated once-daily administration of the benzodiazepine receptor inverse agonist, FG7142. In subsequent experiments, an even larger increase in beta-adrenoceptor number was found 7 days after a single injection of this compound. The present experiments investigated whether FG7142-induced changes in adrenoceptor binding are also found in the rat and whether the effects of a single and repeated injections of this drug differ quantitatively. In view of the anx

GC-MS Analysis, Neuropharmacological, and Antidiarrheal Activities of the Acetone Extract of Najas gracillima Seaweed: In Vivo and In Silico Study.

Najas gracillima, a marine seaweed found in North America and Asia, was investigated for its neuropharmacological and antidiarrheal properties. Acetone extracts of N. gracillima (ANG) were analyzed using both in vivo and in silico methods. Gas chromatography-mass spectrometry (GC-MS) analysis was conducted to identify bioactive compounds present in the extract. In vivo assessments, including the elevated plus maze, light-dark box, and hole board tests, showed that ANG at doses of 200 and 400 mg/kg exhibited significant (p < 0.001) anxiolytic effects. Both doses also demonstrated antidepressant

Essential role of interleukin-15 receptor in normal anxiety behavior.

The interactions between the cytokine interleukin (IL)-15 and the classical neurotransmitter GABA have been shown in IL15Rα receptor knockout mice by observations of memory deficits and reduction of GABA. To test the hypothesis that IL15 affects anxiety-like behavior, knockout mice without IL15, IL15Rα, or the co-receptor IL2Rγ were subjected to open-field and elevated plus maze tests. All three strains showed reduction of anxiety, with greater changes in the IL15Rα knockout mice than in the IL15 or IL2Rγ knockout mice. This unexpected observation is opposite to the reported increase of anxiet…

Dietary casein source modulates water intake and anxiety- and cognition-related behaviors in C57BL/6J male mice.

INTRODUCTION: Semi-purified diets are commonly used in animal studies to improve experimental reproducibility. However, the composition of these diets often differs, particularly in ingredient sources such as casein, and the influence of these compositional differences on physiological and behavioral phenotypes remains unclear. Therefore, this study aimed to compare commonly used semi-purified diets and to test whether ingredient-level variation influences physiological and behavioral outcomes in mice. METHODS: Male C57BL/6J mice were fed semi-purified diets based on the AIN-93G or an OpenSta

The alpha1A antagonist tamsulosin impairs memory acquisition, consolidation and retrieval in a novel object recognition task in mice.

Tamsulosin is an α1-adrenoceptor antagonist used to treat benign prostatic hyperplasia. This drug exhibits high affinity for α1A- and α1D-adrenoceptor subtypes, which are also expressed in the brain. While dementia symptoms have been reported after administration of tamsulosin in humans, studies on its effects on the rodent brain are still rare. The present study investigated the effects of tamsulosin (and biperiden, an amnesic drug) on cognitive performance in the object recognition task (ORT). Tamsulosin (0.001-0.01 mg/kg) was orally administrated in mice at three distinct time points: pre-t
