Identification of potential mechanisms of cognitive dysfunction induced by environmentally relevant PFAS mixture in male rats.

PFAS are emerging persistent organic pollutants, and their potential impacts and underlying mechanisms on cognitive function remain incompletely understood. This study utilized a male rat model to investigate the effects of an environmentally relevant PFAS mixture comprised of eight PFAS on spatial learning and memory, as well as the potential mechanisms through molecular biology assays and multi-omics analysis. Following the administration of PFAS via oral gavage at doses of 0, 0.001, 0.050, 0.250 and 7.000 mg/kg body weight for 28 days, resulting in a decrease in object location recognition

Bioaccumulation and partitioning of PFAS in an AFFF-impacted freshwater system: Invasive Asian clams as indicators of surface water contamination.

Per- and polyfluoroalkyl substances (PFAS) introduced into the environment from historical aqueous film-forming foam (AFFF) releases are persistent sources of ground- and surface water contamination. This study evaluated invasive Asian clams (Corbicula fluminea) as biomonitors in the Little Neshaminy Creek watershed (Pennsylvania, USA), adjacent to two former military installations with documented AFFF use, to assess their ability to integrate spatial and temporal variability in PFAS exposure. Surface water, sediment, and clam tissue were analyzed for 44 PFAS compounds using liquid chromatogra

Interactive renal toxicity of perfluorooctanoic acid and ethanol: Impairment of epithelial transport and redox homeostasis in male Sprague-Dawley rats.

Perfluorooctanoic acid (PFOA), a persistent environmental contaminant, often co-occurs with ethanol (EtOH), a common lifestyle factor. However, the interactive mechanisms underlying their combined renal toxicity remain poorly understood. This study investigated the nephrotoxic effects of 28-day sub-chronic co-exposure to PFOA and EtOH in male Sprague-Dawley rats. Compared with single-exposure groups, the co-exposure group exhibited significantly exacerbated notable interactive renal impairment, characterized by increased relative kidney weights without overt histopathological lesions, consiste

Perfluoroalkyl and polyfluoroalkyl substances exposure and liver disease: A review.

Per- and polyfluoroalkyl substances (PFASs) are widely used in numerous industrial processes and consumer products and are now ubiquitously distributed in the environment, thereby creating multiple routes of human exposure. Their physicochemical properties confer high persistence, bioaccumulation potential, and toxicity, which have raised increasing concern about their long-term impacts on human health. Because of its central role in xenobiotic uptake, metabolism, transport, and excretion, the liver is considered a major target organ of PFAS toxicity. Over the past few years, a growing body of

PFAS exposure linked to altered innate immune biomarkers in a sentinel apex predator, the bald eagle (Haliaeetus leucocephalus).

Per- and polyfluoroalkyl substances (PFAS) are persistent and ubiquitous environmental contaminants that can bioaccumulate in food webs, yet their effects on wildlife health-particularly in apex predators-remain poorly characterized. Bald eagles (Haliaeetus leucocephalus), which integrate contaminant exposure across aquatic and terrestrial prey, provide a valuable sentinel for assessing biological responses to PFAS in freshwater ecosystems. Here, we evaluated associations between PFAS levels in blood plasma of bald eagle nestlings sampled over multiple years across Wisconsin, USA, and biomarke

Fluensulfone induces disruption of core energy metabolism, mitochondrial dysfunction, and reproductive impairment in Eisenia fetida.

Fluensulfone (FSF) is a highly effective nematicide increasingly utilized in agricultural applications. In this study, we exposed earthworms (Eisenia fetida) to environmentally relevant concentrations of FSF and assessed the toxicological responses via an integrated approach combining non-targeted metabolomics, enzyme activity assays, and reverse transcription-quantitative PCR. Exposure to FSF disrupted core energy metabolism by suppressing glycolysis, the tricarboxylic acid cycle, and oxidative phosphorylation. This disruption was reflected by decreased levels of central metabolites (e.g., py

Exploring the link between PFASs exposure and gestational diabetes risk within the TRAEC framework.

Per- and polyfluoroalkyl substances (PFASs) are environmentally persistent industrial substances with potential health impacts during pregnancy. However, evidence linking PFASs exposure to gestational diabetes mellitus (GDM) remains inconclusive. This study systematically evaluated the PFASs-GDM association using the Toxicological Risk Assessment for Environmental Chemicals (TRAEC) strategy. A systematic literature search was conducted using PubMed and Web of Science to identify epidemiological studies, animal models, and cell-based experiments. To validate the evidence, we performed targeted

Associations between prenatal per- and polyfluoroalkyl substance exposure and maternal fatty acid homeostasis: Nonmonotonic, compartment-specific patterns in plasma and red blood cells.

Prenatal exposure to per- and polyfluoroalkyl substances (PFAS) may disrupt maternal fatty acid (FA) metabolism; however, compartment-specific responses, nonmonotonic dose-response relationships (NMDRs), and mixture effects remain insufficiently characterized. We quantified 14 plasma PFAS and comprehensively profiled FAs, FA-class distributions, health-related indices, and product-to-precursor ratios in plasma (n = 192) and red blood cells (RBCs; n = 119) from pregnant women in China. Associations were assessed using multiple linear regression (MLR), restricted cubic splines (RCS), and Bayesia

Per- and polyfluoroalkyl substances (PFAS) and miscarriage: The Norwegian Mother, Father and Child Cohort study.

BACKGROUND: Miscarriage is prevalent, occurring in around 15% of recognized pregnancies. Yet modifiable determinants remain elusive. Per- and polyfluoroalkyl substances (PFAS) can disrupt endocrine- and other biological pathways crucial for reproduction. Whether PFAS is associated with miscarriage in humans remains uncertain. OBJECTIVE: To examine the association between PFAS concentrations and a recent miscarriage. METHODS: We conducted a nested case-control study within The Norwegian Mother, Father, and Child Cohort Study (MoBa), which recruited pregnant women from 1999 to 2008. We utilize

Multi-omics analysis of PFOA-induced sperm DNA damage in mice identifies candidate genes and mendelian randomization reveals their human genetic associations.

Perfluorooctanoic acid (PFOA) is a persistent environmental pollutant linked to male reproductive toxicity, particularly sperm DNA damage, yet the underlying molecular alterations remain incompletely understood. This study established a mouse model of PFOA exposure (10 mg/kg/day for 8 weeks), which significantly increased the sperm DNA fragmentation index. Integrated transcriptomic and proteomic analyses of testicular tissues identified 4781 differentially expressed genes and 639 differentially expressed proteins, with functional enrichment revealing pathways related to stress response, DNA re

Associations between per- and polyfluoroalkyl substances and semen parameters: An updated systematic review and meta-analysis.

Environmental exposure to per- and polyfluoroalkyl substances (PFAS) rises concerns about male reproductive health. Despite a wide body of experimental evidence supporting the biological role of PFAS exposure in male reproductive effects, available epidemiological studies are variably consistent about this association. Four databases were searched up to July 13, 2025 (PROSPERO: CRD420251267798). The systematic review follows Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines. Studies were included that assessed the association of PFAS on sperm outcomes. The

Evaluating the potential of thermal evaporation for onsite retention of per- and polyfluoroalkyl substances (PFAS) in landfill leachate.

Since traditional management of landfill leachate at wastewater treatment plants was found to release per- and polyfluoroalkyl substances (PFAS), alternative technologies are being evaluated for PFAS treatment efficiency. Thermal evaporation is an emerging landfill leachate treatment option allowing for autonomous, onsite management of concentrated residuals while discharging the treated bulk as vapor. This study evaluated the potential of thermal evaporation for onsite PFAS retention by comparing PFAS concentrations in the inputs (leachate and landfill gas) and outputs (evaporation residuals

Sex-specific cellular and molecular mechanisms of PFAS-induced reproductive toxicity.

Per- and polyfluoroalkyl substances (PFAS) are persistent environmental contaminants widely detected in water, food, consumer products, and household dust, resulting in ongoing human exposure and bioaccumulation. Accumulating evidence increasingly implicates PFAS in reproductive dysfunction. Although PFAS-associated reproductive toxicity has been widely reviewed, critical knowledge gaps persist regarding the sex-specific cellular and molecular mechanisms mediating these effects and the reproductive toxicity of emerging replacement PFAS. Increasing epidemiological, in vivo, and in vitro evidenc

PFAS exposure alters gut microbiota metabolites associated with hepatic metabolism: a pilot study.

Per- and polyfluoroalkyl substances (PFAS) are persistent environmental contaminants that can disrupt human hepatic metabolism both directly and through alterations of the gut microbiota. However, the contribution of microbiota-mediated mechanisms to PFAS-induced hepatic dysfunction remains poorly understood. Here, we investigated how PFAS-modified gut microbial metabolites affect human hepatocyte metabolism using an in vitro colon fermentation model, supported by an in vivo mouse and in vitro human hepatocyte exposure studies. PFAS exposure altered the fecal metabolome in human colonic fermen

Bioaccumulation of perfluorooctane sulfonic acid (PFOS) and oxidative stress in snapping turtles in New York, USA.

Perfluorooctane sulfonic acid (PFOS) is an anthropogenic chemical found in aqueous film-forming foams (AFFFs) and many consumer products. Despite its environmental ubiquity and persistence, little is known about the effects of PFOS on stress levels in wild animals. Here, we examined PFOS bioaccumulation and correlations between PFOS exposure and oxidative stress in snapping turtles (Chelydra serpentina) downstream of Griffiss Air Force Base in Rome, New York, a known source of AFFF contamination. Maximum concentrations of PFOS were extrapolated as 41.7 ppb and 498 ppb in water and turtle sampl

Perfluorooctane sulfonate exposure disrupts steroid hormone synthesis in rats via the gut-metabolism-testis axis.

Perfluorooctane sulfonate (PFOS) is an emerging persistent organic pollutant, and environmental exposure to it induces male reproductive toxicity. Given the close relationship between gut microbiota homeostasis and male reproductive function, this study aimed to reveal the impacts of gut microbiota and related metabolism on testicular steroidogenesis in rats following low-level PFOS exposure. The results showed that PFOS exposure significantly elevated progesterone and testosterone levels in rat serum by upregulating the expression of key steroidogenic genes, including steroidogenic acute regu

Inhalable perfluorocarbon RNA nanocapsules overcome biological barriers to treat lung metastases.

Inhalation RNA therapy offers to transform treatment of pulmonary diseases, yet mucus trapping, immune clearance, and navigation of heterogeneous lung tissue architecture prevents RNA from reaching its target cells. Here, we advance our previously reported perfluorocarbon (PFC) emulsion polyplex platform by developing stably aerosolized PFC-based miRNA nanocapsules. These nanocapsules are stabilized by PAMD-C, which is a cholesterol-modified polymeric analog of the FDA-approved CXCR4 antagonist AMD3100 (plerixafor). The nanocapsules exhibit negligible immune clearance, minimal inflammatory res
