Bacterial invasion into the epidermis of rats with sodium lauryl sulphate-irritated skin increases damage and induces incontinence-associated dermatitis.

Incontinence-associated dermatitis (IAD) is caused by prolonged exposure to urine/liquid stool. It is a common and often painful skin condition in older incontinent adults because of poor prevention. Patients with urinary infections are at risk of developing IAD, and to guide the development of novel prevention strategies, we aimed to develop an animal model of IAD by urine and bacteria. First, contralateral sites on the dorsal skin of Sprague-Dawley rats were compromised by sodium lauryl sulphate (SLS), simulating frequent cleansing with soap/water. Filter discs were then placed inside ring-s

Higher Serum CCN3 Is Associated with Disease Activity and Inflammatory Markers in Rheumatoid Arthritis.

Nephroblastoma overexpressed protein (NOV/CCN3), the early discovered member of the CCN family, has recently been suggested to be involved in a number of inflammatory processes, including wound healing, alveolar epithelial cell inflammation, cancer metastasis, and macrophage foam cell formation. However, the role of CCN3 in rheumatoid arthritis (RA), a classic autoimmune and inflammatory disease, remains elusive. RA is a chronic systemic autoimmune disease that eventually leads to cartilage and bone destruction and joint dysfunction. In this study, we investigated the potential of serum CCN3 a

Gene delivery strategies targeting stable atheromatous plaque.

Conventional therapeutic options to treat chronic angina pectoris are pharmacological interventions, coronary bypass surgery (CABG) and percutaneous coronary intervention (PCI). In animal models, it was shown that gene delivery strategies harbour an exciting potential to support and maybe even replace conventional anti-angina treatments, but the translation of the basic science to clinical practise appears problematic. Gene therapy targeting key elements of neointima formation (e.g. cell cycle regulators, metalloproteinases, inflammation and oxidative stress) reduces vein graft and stent failu

Human immunodeficiency virus infection and macrophage cholesterol metabolism.

Macrophages play a central role in the pathogenesis of atherosclerosis and are also a host for a number of viruses, most importantly, HIV. M … lt in the inability of macrophages to control cholesterol accumulation and may lead to formation of foam cells, a characteristic feature of atherosclerosis. Further elucidation of the mechanisms connecting viral infection and macrophage cholesterol metabolism may be fruitful for developing approaches to treatment of atherosclerosis and viral diseases.

Development of the lipid-rich core in human atherosclerosis.

In recent years the role of the atherosclerotic core in promoting plaque rupture has become well recognized. A new insight into core development is its origination early in atherogenesis, before formation of the fibrous plaque. The early core is associated with accumulation of vesicular lipid rich in free cholesterol. Later in core development, lipid deposits become more diverse. The weight of evidence points toward a direct extracellular process, probably lipoprotein aggregation and fusion, as the chief pathway of cholesteryl ester accumulation, although foam cell death may also contribute ch

[Effects of a partial exchange transfusion with Fluosol DA 20% on healthy pigs].

In 9 animals 500 ml of blood was replaced with 500 ml of Fluosol DA 20%. An average of 19.2 ml per kg body weight of emulsion, i.e. 3.883 gm per kg body weight of fluorocarbons was given. On the eighth day following exchange transfusion the animals were killed and tissue samples taken from liver and lung for electron microscopy and chemical analysis. Biochemical parameters were measured in central venous blood prior to transfusion and on the 2nd, 3rd, 6th and 8th days. One animal died on the sixth day of study, hence only the results obtained from the remaining 8 animals were analysed. On remo

Biomimetic 2D-oriented microporous PLLA scaffolds: Fabrication and evaluation for bone repair.

Poly(L-lactic) acid (PLLA) was limited in the application of bone repair materials due to its brittleness, poor melt strength and poor cellular affinity. In this work, single epoxy-terminated polydimethylsiloxane (e-PDMS) was used to extend the molecular chain of PLLA and improve the melt strength of PLLA. Subsequently, a uniform two-dimensional(2D) oriented microporous biomimetic structure was constructed by combining the biaxial orientation technology with the supercritical carbon dioxide (SC-CO2) foaming technology, which greatly improved the strength and toughness of PLLA (from 54.9 MPa,7.

Combining oxygen delivery and generation for targeted atherosclerosis therapy.

Hypoxia plays an important role in the progression of atherosclerosis. However, ameliorating hypoxia at atherosclerotic lesions remains a great challenge. To achieve targeted oxygen delivery to atherosclerotic plaques, Lipid 5-doped, platelet membrane-encapsulated magnetic mesoporous organosilicon nanoparticles loaded with perfluoro-15-crown ether (PFCE) (FMMON@PL) were prepared. PFCE worked as an oxygen carrier, while iron oxide nanoparticles (IONPs) acted as nanozymes with catalase-like activity to facilitate oxygen generation. To enhance plaque targeting, platelet membranes were coated onto

Giant Intracranial Xanthoma with Cloudy Vision as the First Symptom: A Case Report and Literature Review.

A 31-year-old male with mild dizziness complained of cloudy vision in his right eye for 5 days. The visual acuity of both eyes was normal, while the visual contrast sensitivity of both eyes slightly reduced. Fundus examination showed the swollen and radial superficial hemorrhage of his both optic nerves. Brain MRI scan indicated a huge tumor in the right temporal lobe with clear boundary, close to the skull. The midline structure shifted to the left. Blood tests indicated no hyperlipidemia or lipid disorders. The patient then received tumor resection. The size of the tumor was 5.6 cm × 7.5 cm

Transcriptome data and gene ontology analysis in human macrophages ingesting modified lipoproteins in the presence or absence of complement protein C1q.

We characterized the transcriptional effects of complement opsonization on foam cell formation in human monocyte-derived macrophages (HMDM). RNA-sequencing was used to identify the pathways modulated by complement protein C1q during HMDM ingestion of the atherogenic lipoproteins oxidized low density lipoprotein (oxLDL) and acetylated low density lipoprotein (acLDL). All raw data were submitted to the MIAME-compliant database Gene Expression Omnibus (accession number GEO: GSE80442; http://www.ncbi.nlm.nih.gov/geo/query/acc.cgi?acc=GSE80442). Data presented here include Venn diagram overviews of

Soluble TRAIL Concentration in Serum Is Elevated in People with Hypercholesterolemia.

Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) is a multi-functional cytokine, which is involved in the pathophysiological processes of cardiovascular and metabolic diseases. Previously, we demonstrated that TRAIL stimulated lipid uptake and foam cell formation in macrophages in vitro. Several clinical studies have suggested that the serum concentration of TRAIL may be increased in humans with elevated blood cholesterol; however, the current data appear to be inconclusive in this regard. In the present study, we examined the relationships between the serum TRAIL concentration

Role of macromolecules in the safety of use of body wash cosmetics.

One of the most challenging problems related to the use of surfactants in body wash cosmetics is their potential to cause skin irritations. Surfactants can bind with proteins, remove lipids from the epidermal surface, contribute to the disorganization of liquid crystal structures in the intercellular lipids, and interact with living skin cells. These processes can lead to skin irritations and allergic reactions, and impair the epidermal barrier function. The present study is an attempt to assess the effect of polymers and hydrolysed proteins present in the formulations of model body wash cosme

A 21-aminosteroid inhibits oxidation of human low density lipoprotein by human monocytes and copper.

Oxidation of low density lipoprotein (LDL) leads to more rapid uptake by arterial wall macrophages and foam cell formation. Inhibiting LDL oxidation may impede these processes and offers a new mechanism to retard atherogenesis. The 21-aminosteroids, derived from methylprednisolone, are potent inhibitors of free radical production by stimulated monocytes and also are scavengers of lipid peroxyl radicals. The 21-aminosteroid, U74500A, was added to a mixture of low density lipoprotein cholesterol and human monocytes to which lipopolysaccharide was add to stimulate the monocytes. At a final concen

Mimicking the Architecture and Dissolution Chemistry of Cancellous Bone Tissue to Optimize the Biocompatibility of Bioactive Scaffolds.

Synthesis of mechanically stable porous scaffolds with an architecture analogous to cancellous bone tissue poses significant challenges to bioactive glass (BG) based scaffolds. This is primarily due to densification and crystallization of the BG's during heat treatment. This study presents a modified BG series (42SiO2-xTiO2-24Na2O-21CaO-13P2O5, where x = 8 and 16 TiO2). TiO2 replaced the SiO2 concentration in the glass and was incorporated due to its biocompatibility and influence on glass structure. Material characterization determined that TiO2 did not induce crystallization within the glass
