Evaluation of Various Solvent Extracts of Tetrastigma leucostaphylum (Dennst.) Alston Leaves, a Bangladeshi Traditional Medicine Used for the Treatment of Diarrhea.

Tetrastigma leucostaphylum (TL) is an important ethnic medicine of Bangladesh used to treat diarrhea and dysentery. Hence, current study has been designed to characterize the antidiarrheal (in vivo) and cytotoxic (in vitro) effects of T. leucostaphylum. A crude extract was prepared with methanol (MTL) and further partitioned into n-hexane (NTL), dichloromethane (DTL), and n-butanol (BTL) fractions. Antidiarrheal activity was investigated using castor oil induced diarrhea, enteropooling, and gastrointestinal transit models, while cytotoxicity was evaluated using the brine shrimp lethality bioas…

Imaging the Autonomic Nervous System in Parkinson's Disease.

PURPOSE OF REVIEW: Patients with Parkinson's disease (PD) often display gastrointestinal and genitourinary autonomic symptoms years or even decades prior to diagnosis. These symptoms are thought to be caused in part by pathological α-synuclein inclusions in the peripheral autonomic and enteric nervous systems. It has been proposed that the initial α-synuclein aggregation may in some PD patients originate in peripheral nerve terminals and then spread centripetally to the spinal cord and brainstem. In vivo imaging methods can directly quantify the degeneration of the autonomic nervous system as

Evaluation of the treatment with resveratrol-loaded nanoparticles in intestinal injury model caused by ischemia and reperfusion.

The gastrointestinal tract is extremely sensitive to ischemia and reperfusion (I/R). Studies have reported that resveratrol (RSV) is able to … minotransferase, alanine aminotransferase, immunolabeling of myenteric neurons and glial cells, and gastrointestinal transit was evaluated. Both nanoparticle formulations presented negative charge with homogeneous distribution, and the payload, showed an encapsulation efficiency of 60%. Resveratrol administered in free form prevented alterations that were caused by I/R. T …

Chronic Electrical Stimulation at Acupoints Reduces Body Weight and Improves Blood Glucose in Obese Rats via Autonomic Pathway.

BACKGROUND: The aim of this study was to investigate effects and mechanisms of chronic electrical stimulation at acupoints (CEA) using surgically implanted electrodes on food intake, body weight, and metabolisms in diet-induced obese (DIO) rats. METHODS: Thirty-six DIO rats were chronically implanted with electrodes at acupoints ST-36 (Zusanli). Three sets of parameters were tested: electrical acupuncture (EA) 1 (2-s on, 3-s off, 0.5 ms, 15 Hz, 6 mA), EA2 (same as EA1 but continuous pulses), and EA3 (same as EA2 but 10 mA). A chronic study was then performed to investigate the effects of CEA

Modified Spiral Intestinal Lengthening and Tailoring for Short Bowel Syndrome.

BACKGROUND: The spiral intestinal lengthening and tailoring (SILT) procedure is a new surgical technique for autologous intestinal reconstruction in patients with short bowel syndrome. The aim of this work is to present a first description of a modified SILT technique by which the mucosal layer is left intact to possibly reduce the severe postoperative complications of intestinal leakage and abdominal abscess formation. MATERIALS AND METHODS: The modified SILT technique was performed on a 10-cm-long intestinal segment in 2 pigs to determine the technical feasibility. Thereafter, the short-ter

The thyroid and the gut.

Thyroid disease is common, and its effects on the gastrointestinal system are protean, affecting most hollow organs. Hashimoto disease, the most common cause of hypothyroidism, may be associated with an esophageal motility disorder presenting as dysphagia or heartburn. Dyspepsia, nausea, or vomiting may be due to delayed gastric emptying. Abdominal discomfort, flatulence, and bloating occur in those with bacterial overgrowth and improve with antibiotics. Reduced acid production may be due to autoimmune gastritis or low gastrin levels. Constipation may result from diminished motility, leading t

Gastrointestinal dysfunction in a community sample of subjects with symptoms of irritable bowel syndrome.

BACKGROUND/AIM: Irritable bowel syndrome (IBS) affects about 15-20% of the population of the Western countries. Traditionally, IBS has been an exclusion diagnosis, but recently definitions have emerged from population-based research. The aim of this population-based study was to evaluate any association between gastrointestinal pathophysiology and IBS in subjects with symptoms of IBS compared to subjects with no abdominal complaints. METHODS: From a random sample of 2,656 participants, subjects with IBS (32) together with subjects without abdominal complaints (26), were invited for further ev

Origin and control of gastrointestinal motility.

As a result of improved understanding of the origin and control of motility at both the whole organ and the cellular level, a scientific approach to the diagnosis and treatment (both medical and surgical) of motility disorders has evolved. Examples are present for all levels of the gastrointestinal tract. Manometric, myoelectric, and pharmacologic studies have elucidated the role of the lower esophageal sphincter and stomach in the pathogenesis of gastroesophageal reflux and determined the mechanism of successful medical and surgical treatment. Better evaluation of colorectal motility using co

Correlation of the bioavailability of diazepam from uncoated tablets in beagle dogs with its dissolution rate and bioavailability in humans.

The bioavailability of diazepam (I) in uncoated tablets in beagles was tested using tablets tested previously in humans. The correlations of the dissolution rates and bioavailabilities of these tablets in humans and beagles were examined. The plasma level of N-desmethyldiazepam (II), the main metabolite of I, was used as an index of bioavailability after p.o. administration of uncoated tablets of I, because I is rapidly metabolized. Thus the plasma level of I is very low, and AUCs (areas under plasma level-time curves) calculated from plasma levels of II were related to the dose of I (2-10 mg)

Longitudinal transitions of somatic-emotional distress in Chinese adolescents: A random-intercept latent transition analysis of the longitudinal protective effects of resilience.

Pediatric internalizing problems, suicidality, and functional gastrointestinal disorders (FGIDs) frequently co-occur, but their longitudinal dynamics and the protective role of resilience remain under-investigated. In this two-wave, 12-month prospective study included 9035 Chinese children and adolescents aged 10-17 years, we used self-report measures (the Rome IV Diagnostic Questionnaire for Functional Gastrointestinal Disorders, Patient Health Questionnaire-9, General Anxiety Disorder-7, Suicide Behaviors Questionnaire-Revised, and Resilience Scale for Children and Adolescents) to assess the

Menopause and gastrointestinal health and disease.

Menopause has far-reaching effects on human physiology, including the gastrointestinal tract, and can negatively influence the quality of life of women who are affected. Menopause can have multiple effects on gastrointestinal function, including altering gut motility and changing the composition of the gut microbiota. As a result, some gastrointestinal and hepatic conditions are more common among individuals in peri- and postmenopause, and people with these conditions before menopause might also experience greater symptom severity and worse health-related quality of life during this time. The

Assessment of gastrointestinal function and enteric nervous system changes over time in the A53T mouse model of Parkinson's disease.

Gastrointestinal (GI) dysfunctions, including constipation and delayed stomach emptying, are prevalent and debilitating non-motor symptoms of Parkinson's disease (PD). These symptoms have been associated with damage in the enteric nervous system (ENS) and the accumulation of pathogenic alpha-synuclein (α-Syn) within the GI tract. While motor deficits and dopaminergic neuron loss in the central nervous system (CNS) of the A53T mouse model are well-characterised, the temporal relationship between GI dysfunction, ENS pathology, and motor symptoms remains unclear. This study aimed to investigate f

Pilot postoperative ileus study of escin in cancer patients after colorectal surgery.

BACKGROUND: Postoperative ileus, a common complication in patients after abdominal surgery, brings no benefit to the recovery of postoperative patients, and treatment targeted at restoring gastrointestinal motility may shorten the hospital stay. Studies have shown that escin accelerates gastrointestinal transit in mice and improves gastrointestinal motility in patients after abdominal surgery. A pilot study of escin's effect on the recovery of gastrointestinal motility was conducted in colorectal cancer patients in anticipation of a multiple-center randomized controlled trial. METHODS: A tota…

Lactobacillus acidophilus C4 ameliorates constipation in mice.

Functional constipation (FC) is a common gastrointestinal disorder that can seriously affect patients' quality of life. Probiotics have emerged as a promising strategy for preventing and managing gastrointestinal diseases. Previously, we isolated Lactobacillus acidophilus C4 from the feces of a healthy child, and here, we investigated its probiotic efficacy against FC in a mouse model. Prophylactic treatment with L. acidophilus C4 prior to FC onset significantly reduced impairment of defecation function and increased intestinal transit rate. L. acidophilus C4 also increased the expression of t

Intraperitoneal Urine Exposure Does Not Elicit Peritoneal Inflammation or Gastrointestinal Dysmotility: A Histopathological and Functional Study in Rats.

OBJECTIVE: Intra-abdominal urine leakage can occur during pediatric urologic surgeries such as laparoscopic pyeloplasty. While some patients exhibit postoperative symptoms resembling ileus, the pathophysiological effects of urine exposure on the peritoneum remain unclear. This study aimed to determine whether intraabdominal exposure to sterile urine induces peritonitis or gastrointestinal (GI) dysmotility using a rat model. METHODS: Four groups were established using 9-week-old female Sprague-Dawley rats: the sham group, the intraperitoneal autologous urine exposure group, the mechanical peri

Protective and antioxidant effects of Opuntia ficus-indica cladodes hydroethanolic extract on castor oil-induced diarrhea in rats.

Gastrointestinal disorders, which significantly impact human health, are a major focus of research. Diarrhea is one such disorder, affecting individuals across all age groups. Conventional anti-diarrheal drugs are often characterized byside effects, so safer alternative treatments are needed. In this context, we investigated the effects of Opuntia ficus-indica, a plant known for its therapeutic properties, by evaluating the impact of its cladode hydroethanolic extraction castor oil-induced diarrhea in rat. The hydroethanolic cladodes extract (HECE) was prepared from fresh cladodes, and its bio
